Evaluation of the mutagenic effect of the iodinated contrast medium Urografina® 292 using the micronucleus test in mouse bone marrow cells Anais da ABC (AABC)
BELLE,MONICA B.B.; LEFFA,DANIELA D.; MAZZORANA,DALIANE; ANDRADE,VANESSA M. DE.
Contrast media (CM) are frequently used in diagnostic radiology and in radiotherapy as a diagnostic tool and in treatment planning. Previous studies have demonstrated that these compounds induce chromosomal aberrations. This study evaluates the mutagenic effects induced by the contrast medium Urografina® 292 (meglumine amidotrizoate and sodium-ionic dimmer) in bone marrow cells (BMC) of mice in vivo. Micronuclei assay was performed in BMC of CF-1 mice injected with CM 1.5 and 3.0 mL/kg intravenous doses and 1.0, 2.0, 3.0 mL/kg intraperitoneal doses. The animals were beheaded 24 h after treatment by cervical dislocation, and femur BMC from each animal were used in the micronucleus test. The group treated with the highest intravenous injection of Urografina®...

Physalis angulata L (Solanaceae) is a medicinal plant from North of Brazil, whose different extracts and infusions are commonly used in the popular medicine for the treatment of malaria, asthma, hepatitis, dermatitis and rheumatism. However, the genotoxic effects of P. angulata on human cells is not well known. The main purpose of the present study was to evaluate the in vitro genotoxic effects of aqueous extract of P. angulata using the comet assay and the micronucleus assay in human lymphocytes provided from 6 healthy donors. Treatments with P. angulata extracts were performed in vitro in order to access the extent of DNA damage. The comet assay has shown that treatments with P. angulata at 0.5, 1.0, 2.0, 3.0 and 6.0 μg/mL in culture medium were...

Haveric,Sanin; Hadzic,Maida; Haveric,Anja; Mijanovic,Mirjana; Hadziselimovic,Rifat; Galic,Borivoj.
ABSTRACT Genotoxic effects of inorganic molecule dipotassium-trioxohydroxytetrafluorotriborate, K2(B3O3F4OH), a promising new therapeutic for the epidermal changes treatment, have been evaluated. In vitro analysis included evaluation of genotoxic and cytotoxic potential of K2(B3O3F4OH) in concentrations of 0.01, 0.02, 0.05 and 0.06 mg/mL applying cytokinesis-block micronucleus cytome assay in human lymphocyte culture. With the increase of concentration the frequency of micronuclei elevated but the differences were not significant. Also, there were no significant differences among the frequencies of nuclear buds and nucleoplasmic bridges between controls and treated cultures. Nuclear division index and nuclear division cytotoxycity index values did not...

Colocalization of coilin and nucleolar proteins in Cajal body-like structures of micronucleated PtK2 cells BJMBR
Silva,N.P.; Christofolini,D.M.; Mortara,R.A.; Andrade,L.E.C..
Cajal bodies (CB) are ubiquitous nuclear structures involved in the biogenesis of small nuclear ribonucleoproteins and show narrow association with the nucleolus. To identify possible relationships between CB and the nucleolus, the localization of coilin, a marker of CB, and of a set of nucleolar proteins was investigated in cultured PtK2 cells undergoing micronucleation. Nocodazol-induced micronucleated cells were examined by double indirect immunofluorescence with antibodies against coilin, fibrillarin, NOR-90/hUBF, RNA polymerase I, PM/Scl, and To/Th. Cells were imaged on a BioRad 1024-UV confocal system attached to a Zeiss Axiovert 100 microscope. Since PtK2 cells possess only one nucleolus organizer region, micronucleated cells presented only one or...

Micronucleated lymphocytes in parents of Down syndrome children BJMBR
Silva-Grecco,R.L.; Navarro,G.C.; Cruz,R.M.; Balarin,M.A.S..
Down syndrome (DS) is the most common disease due to an autosomal aneuploidy in live born children and also the major known genetic cause of mental retardation. The risk of a DS pregnancy increases substantially with increasing maternal age. However, several women aged less than 35 years at conception have a child with DS. The micronucleus (MN) assay can identify chromosome breakage or chromosome malsegregation and is an ideal biomarker to investigate genomic instability. The aim of the present study was to determine the frequency of peripheral lymphocytes with MN in the parents of DS individuals. The subjects were 17 couples, 1 father and 9 mothers, and 24 couples who had at least one healthy child formed the control group. For each individual we...

Tomato and garlic by gavage modulate 7,12-dimethylbenz[a]anthracene-induced genotoxicity and oxidative stress in mice BJMBR
Bhuvaneswari,V.; Velmurugan,B.; Abraham,S.K.; Nagini,S..
Chemoprotection by dietary agents is a promising strategy for cancer prevention. The aim of the present study was to evaluate the combined effect of tomato and garlic against 7,12-dimethylbenz- [a]anthracene (DMBA)-induced genetic damage and oxidative stress in 12-14-week-old male Swiss albino mice. The animals were randomized into experimental and control groups and divided into eight groups of five animals each. Group 1 animals were injected intraperitoneally with 35 mg/kg body weight DMBA suspended in peanut oil as a single dose. Groups 2-4 animals received tomato (500 mg/kg body weight), garlic (125 mg/kg body weight) and a combination of tomato and garlic for 5 days by gavage, respectively, followed by DMBA 1.5 h after the final feeding. The doses of...

Genetic stability in synthetic wheat accessions: cytogenetic evaluation as a support in breeding programs Ciência Rural
Frizon,Patrícia; Brammer,Sandra Patussi; Lima,Maria Imaculada Pontes Moreira; Castro,Ricardo Lima de; Deuner,Carolina Cardoso.
ABSTRACT: Synthetic wheat is developed by crossing tetraploid species ( Triticum turgidum , AABB) with a diploid species ( Aegilops tauschii , DD), followed by chromosome duplication through the use of colchicine to restore the resultant sterile hybrid to a fertile hexaploid plant. The main importance of producing synthetically improved wheat is to increase their genetic variability and to incorporate genes that code for resistance to biotic and abiotic stressors. This study aimed to evaluate the presence of micronuclei (MN) and the meiotic index (MI) in the tetrad phase in synthetic wheat accessions and cultivars ( Triticum aestivum ) stored at the Germplasm Bank of Embrapa Trigo (Brazil), in order to identify and select genetically stable accessions for...
