Bellini,Marilanda Ferreira; Cabrioti,Leonardo Neves; Terezan,Ana Paula; Jordão,Berenice Quinzani; Ribeiro,Lúcia Regina; Mantovani,Mário Sérgio.
Functional food investigations have demonstrated the presence of substances that could be beneficial to human health when consumed. However, the toxic effects of some substances contained in foods have been determined. Reported medicinal and nutritive properties have led to the extensive commercialization of the basidiomycete fungi Agaricus blazei Murrill (sensu Heinemann), also known as Agaricus brasiliensis Wasser et al., Agaricus subrufescens Peck or the Brazilian medical mushroom (BMM). Different methanolic extract fractions (ME) of this mushroom were submitted to the cytokinesis-block micronucleus (CBMN) clastogenic assay and the hypoxanthine-guanine phosphoribosyl transferase locus (HGPRT) assay for gene mutation, both using Chinese hamster ovary...

Oliveira,Rodrigo Juliano; Pesarini,João Renato; Salles,Maria José Sparça; Kanno,Tatiane Yumi Nakamura; Lourenço,Ana Carolina dos Santos; Leite,Véssia da Silva; Silva,Ariane Fernanda da; Matiazi,Hevenilton José; Ribeiro,Lúcia Regina; Mantovani,Mário Sérgio.
β-glucan is a well-known polysaccharide for its chemopreventive effect. This study aimed to evaluate the chemopreventive ability of β-glucan in somatic and germ cells through the dominant lethal and micronucleus assays, and its influence on the reproductive performance of male mice exposed to cyclophosphamide. The results indicate that β-glucan is capable of preventing changes in DNA in both germ cells and somatic ones. Changes in germ cells were evaluated by the dominant lethal assay and showed damage reduction percentages of 46.46% and 43.79% for the doses of 100 and 150 mg/kg. For the somatic changes, evaluated by micronucleus assay in peripheral blood cells in the first week of treatment, damage reduction percentages from 80.63-116.32% were found. In...

Abstract Metal ions such as iron can induce DNA damage by inducing reactive oxygen species (ROS) and oxidative stress. Vitamin C is one of the most widely consumed antioxidants worldwide, present in many fruits and vegetables, especially inMalpighia glabra L., popularly known as acerola, native to Brazil. Acerola is considered a functional fruit due to its high antioxidant properties and phenolic contents, and therefore is consumed to prevent diseases or as adjuvant in treatment strategies. Here, the influence of ripe and unripe acerola juices on iron genotoxicity was analyzed in vivo using the comet assay and micronucleus test. The comet assay results showed that acerola juice exerted no genotoxic or antigenotoxic activity. Neither ripe nor unripe acerola...

Comparison of nuclear abnormalities in Astyanax bifasciatus Cuvier, 1819 (Teleostei: Characidae) of two sections of rivers from the middle Iguaçu Biological Sciences
Melo-Silva, Merieli; Oliveira, Felipe Rafael; Rosa, Jonathan; Gemelli, Emanuelli; Santos, Lilian; Bueno-Krawczyk, Ana Carolina de Deus.
Most changes in water bodies are result of human activities that have the potential to undermine the environmental integrity of aquatic ecosystems. Changes in genetic material can be evidenced by the frequency of nuclear abnormalities in fish blood cells, in response to genotoxic agents even at low concentrations. Thus, we aimed at comparing the frequencies of nuclear abnormalities of fish kept in acclimation, with fish collected in Timbó river (Santa Cruz do Timbó, Santa Catarina State), under low anthropogenic interference, and fish collected in Iguaçu river (União da Vitória, Paraná State), a polluted river. The highest frequency of changes in nuclear morphology of fish erythrocytes was found in the urbanized area around the Iguaçu river, while, the...

Erolu,Erhan H; Hamzaolu,Ergin; Aksoy,Ahmet; Budak,Ümit; Özkul,Yusuf.
Helichrysum sanguineum, Helichrysum pamphylicum, Helichrysum orientale, Helichrysum noeanum (Asteraceae) are medicinal plants. For centuries, they have been used as tea in Turkey because of their medicinal properties. So far no scientifc evidence has been found in a literature survey regarding the genotoxic effects of these plants. This work evaluated the genotoxic effects on human lymphocyte cultures induced by methanol extracts of these plants, assayed in different concentrations (0.01, 0.05, 0.1, 0.5 and 1 mg/mL). According to the results, Helichrysum noeanum, Helichrysum pamphylicum and Helichrysum sanguineum induced the formation of micronuclei and decreased the mitotic and replication indexes. Helichrysum orientale did not affect these parameters,...

Microscopic Studies on Erythrocytes of Channa punctata Exposed to Commercial Grade Lindane BABT
Bhattacharjee,Debasish; Das,Suchismita.
ABSTRACT Light and scanning electron microscopic studies of erythrocytes of Channa punctata exposed to two sublethal doses of lindane over a period of 21 days revealed myriads of anomalies. High frequency of micronucleus appeared progressively in the erythrocyte, ranging from 3.8-7.5%, when monitored weekly for 21 d at 0.25 g/L and 1.0-3.0% at 0.025 g/L lindane. Prominent structural anomalies of erythrocytes included abnormal shapes, vacuolation, membrane invagination and disintegration. At higher dose and at later duration, a unique tendency of abnormal cells to stick together was observed. It could be concluded that commercial formulation of lindane could induce genotoxicity and structural anomalies in the erythrocytes of fish.

The cytogenetic effects of black tea and green tea on cultured human lymphocytes BABT
Eroğlu,Halil Erhan.
In this study, the cytogenetic effects of black tea and green tea were determined in cultured peripheral blood lymphocytes. Results showed that black tea and green tea induced the mitotic and replication indexes and decreased micronuclei. But these data were not statistically significant for green tea. The effects of black tea on the micronucleus formation and mitotic index were statistically significant. The decrease in micronucleus counts indicated that black tea and green tea had considerable anticlastogenic and antigenotoxic effects as observed in vitro in human lymphocytes. Thus, it could be concluded that tea polyphenols protected the normal cells from genotoxic or carcinogenic agents, which indicated the therapeutic and antioxidative role of...

Zamani,Ehsan; Shokrzadeh,Mohammad; Modanloo,Mona; Shaki,Fatemeh.
ABSTRACT Acrylamide (AA), is an important industrial compound, formed during food processing under high temperature. Due to its potential carcinogenic effect, it has been recognized as a human health concern. In this study, we investigated the role of oxidative stress in acrylamide’s genotoxicity and protective role of L- carnitine (LCA) in human lymphocytes. The micronucleus test (MNT) was used to monitor AA genotoxicity after 20h exposure to concentrations of 0.25,50 and 100 mM. Furthermore we evaluated oxidative stress markers, such as reactive oxygen species (ROS), lipid peroxidation (LPO) and glutathione (GSH) content. In MNT, It has been found that AA at high concentrations (50 and 100 µM) significantly increased the micronuclei (MN) frequencies. On...

Cell cycle synchronization and BrdU incorporation as a tool to study the possible selective elimination of ErbB1 gene in the micronuclei in A549 cells BJMBR
Lauand,C.; Niero,E.L.; Dias,V.M.; Machado-Santelli,G.M..
Lung cancer often exhibits molecular changes, such as the overexpression of the ErbB1 gene that encodes epidermal growth factor receptor (EGFR). ErbB1 amplification and mutation are associated with tumor aggressiveness and low response to therapy. The aim of the present study was to design a schedule to synchronize the cell cycle of A549 cell line (a non-small cell lung cancer) and to analyze the possible association between the micronuclei (MNs) and the extrusion of ErbB1 gene extra-copies. After double blocking, by the process of fetal bovine serum deprivation and vincristine treatment, MNs formation was monitored with 5-bromo-2-deoxyuridine (BrdU) incorporation, which is an S-phase marker. Statistical analyses allowed us to infer that MNs may arise both...
