O objetivo do presente trabalho foi estudar as diferentes respostas morfogenéticas de embriões zigóticos de açaizeiro (Euterpe oleracea Mart.) submetidos a várias condições de cultura in vitro. Os experimentos foram conduzidos em laboratório com material vegetal coletado de plantas de açaí na Embrapa Amazônia Oriental, Belém, PA, Brasil. Verificou-se a expressão de um modelo de embriogênese somática, direto, repetitivo a assincronizado em embriões zigóticos maduros cultivados em meio primário MS suplementado com 339.36 ? A propagação in vitro de teca tem sido realizada por meio da cultura de tecidos vegetais de diferentes genótipos. Nesse contexto, o uso da micropropagação a partir de plantações de teca no Brasil pode ser uma importante ferramenta, pois permite a produção de grandes quantidades de novas plantas em um curto espaço de tempo, a partir de um único propágulo vegetativo retirado de uma planta matriz. Assim, o objetivo deste trabalho é descrever protocolos de micropropagação da espécie Tectona grandis a partir de genótipos estabelecidos na Amazônia Sul-Ocidental, no Estado do Acre.

Somatic embryogenesis and in vitro plant regeneration from pejibaye adult plant leaf primordia PAB

The aim of this work was to evaluate a protocol for plant regeneration by means of somatic embryos obtained from isolated adult pejibaye leaf primordia, and to describe histological origin of embryos and morphogenetic response. Explants were cultivated in modified MS medium. Mesophyll parenchymatous cells originated meristemoids (preembryonic complex formation) induced with 7.1 µM BAP in the first two subculture periods. After polarized structures with 12.9 µM NAA and 3.55 µM BAP were formed in the third subculture, somatic embryos developed and regenerated normal plants. The mesophyll parenchymatous cells display high capacity of direct response to the auxin and cytokinin.

Acclimatization of Tapeinochilos ananassae plantlets in association with arbuscular mycorrhizal fungi PAB

The objective of this work was to assess the potential of three isolates of arbuscular mycorrhizal fungi to promote growth of micropropagated plantlets of Tapeinochilos ananassae during acclimatization. The experiment was carried out in greenhouse, in a completely randomized block design, with four inoculation treatments: non‑inoculated control and plants inoculated with Glomus etunicatum, Acaulospora longula or Gigaspora albida, with ten replicates. After 90 days, the following parameters were evaluated: survival rate, height, leaf and tiller number, leaf area, fresh and dry biomass, contents of macro‑ and micronutrients in the root and shoot, glomerospore number, and mycorrhizal colonization. Somatic embryogenesis and plant regeneration in elite clones of Theobroma cacao PAB

The objective of this work was to evaluated a procedure for somatic embryogenesis and regeneration of cacao (Theobroma cacao L.) elite clones. Petal explants from cacao clones TSH 565 and TSH 1188 were cultured on PCG and SCG-2 media, for calli growth. Somatic embryos were formed on the surface of embryogenic calli after transfer to embryo development (ED) medium. Clone TSH 565 showed a higher embryogenic potential than TSH 1188. The best combination of carbon source for embryo induction in ED medium was genotype-specific. Embryogenic callus formations increased in micropore tape-sealed Petri dishes, irrespective of cacao genotype. Mature somatic embryos were successfully converted into plantlets.

ANATOMICAL STUDIES OF IN VITRO ORGANOGENESIS INDUCED IN LEAF-DERIVED EXPLANTS OF PASSIONFRUIT PAB

With the aim of studying the organogenesis in vitro in Passiflora edulis Sims f. flavicarpa Deg., the passionfruit, leaf-derived explants were cultured on media containing NAA or BAP and incubated either in continuous darkness or in light. The histological events leading to de novo organ formation were evaluated. Darkness induces rhizogenesis in the presence of NAA, whereas direct shoot regeneration is stimulated by light and BAP. This latter condition is recommended for passionfruit micropropagation as several adventitious shoot buds were formed from meristemoids of parenchymal origin.

In vitro conservation of Piper aduncum and Piper hispidinervum under slow-growth conditions PAB

The objective of this work was to evaluate in vitro storage of Piper aduncum and P. hispidinervum under slow-growth conditions. Shoots were stored at low temperatures (10, 20 and 25°C), and the culture medium was supplemented with osmotic agents (sucrose and mannitol - at 1, 2 and 3%) and abiscisic acid - ABA (0, 0.5, 1.0, 2.0 and 3.0 mg L-1). After six-months of storage, shoots were evaluated for survival and regrowth. Low temperature at 20ºC was effective for the in vitro conservation of P. aduncum and P. hispidinervum shoots. In vitro cultures maintained at 20ºC on MS medium showed 100% survival with slow-growth shoots. Rescue of a non-viable accession and rapd analysis of recovered plants of Arachis retusa PAB

In vitro regeneration of Arachis retusa was examined for the purpose of germplasm renewal and conservation. Random amplified polymorphic DNA (RAPD) fingerprinting was used to evaluate the genetic stability of plants derived from embryo axes and apical segments. Ten arbitrary decamer primers were screened and five of them were selected. Ninety genomic regions were evaluated, with an average of 18 loci per clone. All amplified segments were monomorphic. The results indicate that recovered plants are genetically stable at the assessed genomic regions and that both regeneration processes are suitable for in vitro germplasm preservation of Arachis species.

Abstract: The objective of this work was to evaluate the effect of silicon and agar concentrations on cockscomb (Celosia cristata) (Amaranthaceae) development in vitro. Phytotechnical, anatomical, and ultrastructural analyses were carried out in a completely randomized design, in a 4x3 factorial arrangment, with four silicic acid concentrations (0.0, 0.5, 1.0, and 1.5 g L-1) and three agar concentrations (5.5, 8.0, and 10.5 g L-1), totaling 12 treatments with seven replicates. Short-term storage in vitro and large-scale propagation of grapevine genotypes PAB

The objective of this work was to evaluate the large-scale propagation of grapevine genotypes after short-term storage in vitro. Microshoots from ten grapevine genotypes were used. The following storage temperatures were evaluated: 10, 20, and 25°C. After short-term storage, the shoots were propagated in up to five successive subcultures, to assess the large-scale propagation of the germplasm maintained under conditions of minimal growth. The propagated shoots were rooted in different concentrations of indolbutiric acid (IBA) and acclimatized in greenhouse. Influence of substrates and in vitro preconditioning treatments on ex vitro acclimatization of Arachis retusa PAB

The objective of this work was to evaluate the influence of substrate and preconditioning treatments on the acclimatization of in vitro plants of Arachis retusa. Plants were transferred to Plantmax or sand, and fertilized with Hoagland's nutrient solution. Plants maintained in sand, with or without fertilizer, showed the highest survival rates. In order to evaluate the influence of in vitro preconditioning treatments, stem segments were cultured on MS medium supplemented with different sucrose concentrations. The highest survival and developmental rates were observed in plants from two accessions cultured on MS supplemented with 1.5% and 3% sucrose. Flowering and fruit production were observed after five months.

Solidifying agents and activated charcoal for in vitro culture of Solanum sessiliflorum PAB

Abstract: The objective of this work was to evaluate the effects of the solidifying agents agar and phytagel and of activated charcoal on the in vitro cultivation of two maná cubiu (Solanum sessiliflorum) varieties: Thaís and Santa Luzia. The phytotechnical characteristics analyzed included number of leaves, number of roots, shoot and root length, and fresh matter of shoot and root. Regardless of the variety, phytagel was superior to agar as a culture medium. A greater number of leaves and longer shoots were observed in the Santa Luzia variety, in the absence of charcoal. The Thaís variety showed longer shoots and roots in the presence of charcoal.

An efficient protocol for culturing meristems of sorghum hybrids Phyton

A robust protocol for culturing meristems of Sorghum is required to assist with rapid genetic improvement of the genus. Through meristem culture, an efficient method for rapid micropropagation was developed for Sorghum bicolor (L.) Moench. hybrids, namely NC+262, NC+6C21 and NC+6B50. Complete plants were regenerated directly from shoot meristems without an intervening callus phase. Regeneration frequencies varied between the studied genotypes and according to the growth regulator combinations present in the medium. The combination of BAP and TDZ showed a synergistic effect on shoot multiplication. The induction and control of in vitro somatic embryogenesis is dependent of a number of factors. The genotype of the mother plant donor of explants and the kind of auxin play essential roles in conferring embryogenic competence.
