RESUMO: A eliminação de microrganismos é uma condição essencial para o estabelecimento de um processo de regeneração in vitro, pois eles além de competirem pelos nutrientes também produzem toxinas que causam a morte do explante. O trabalho visou desenvolver metodologias de controle de crescimento de microrganismos provenientes de explantes de mangueira inoculados em meio de cultura, como fase inicial do estabelecimento de um protocolo de micropropagação de gemas laterais de mangueira para apoiar o programa de melhoramento desenvolvido na Embrapa. O objetivo do presente trabalho foi estudar as diferentes respostas morfogenéticas de embriões zigóticos de açaizeiro (Euterpe oleracea Mart.) submetidos a várias condições de cultura in vitro. Os experimentos foram conduzidos em laboratório com material vegetal coletado de plantas de açaí na Embrapa Amazônia Oriental, Belém, PA, Brasil. Verificou-se a expressão de um modelo de embriogênese somática, direto, repetitivo a assincronizado em embriões zigóticos maduros cultivados em meio primário MS suplementado com 339.36 ? The objective of this work was to assess the potential of three isolates of arbuscular mycorrhizal fungi to promote growth of micropropagated plantlets of Tapeinochilos ananassae during acclimatization. The experiment was carried out in greenhouse, in a completely randomized block design, with four inoculation treatments: non‑inoculated control and plants inoculated with Glomus etunicatum, Acaulospora longula or Gigaspora albida, with ten replicates. The objective of this work was to evaluate in vitro storage of Piper aduncum and P. hispidinervum under slow-growth conditions. Shoots were stored at low temperatures (10, 20 and 25°C), and the culture medium was supplemented with osmotic agents (sucrose and mannitol - at 1, 2 and 3%) and abiscisic acid - ABA (0, 0.5, 1.0, 2.0 and 3.0 mg L-1). After six-months of storage, shoots were evaluated for survival and regrowth. Low temperature at 20ºC was effective for the in vitro conservation of P. aduncum and P. hispidinervum shoots. In vitro cultures maintained at 20ºC on MS medium showed 100% survival with slow-growth shoots. Abstract: The objective of this work was to evaluate the effect of silicon and agar concentrations on cockscomb (Celosia cristata) (Amaranthaceae) development in vitro. Phytotechnical, anatomical, and ultrastructural analyses were carried out in a completely randomized design, in a 4x3 factorial arrangment, with four silicic acid concentrations (0.0, 0.5, 1.0, and 1.5 g L-1) and three agar concentrations (5.5, 8.0, and 10.5 g L-1), totaling 12 treatments with seven replicates. The objective of this work was to evaluate the influence of substrate and preconditioning treatments on the acclimatization of in vitro plants of Arachis retusa. Plants were transferred to Plantmax or sand, and fertilized with Hoagland's nutrient solution. Plants maintained in sand, with or without fertilizer, showed the highest survival rates. In order to evaluate the influence of in vitro preconditioning treatments, stem segments were cultured on MS medium supplemented with different sucrose concentrations. The highest survival and developmental rates were observed in plants from two accessions cultured on MS supplemented with 1.5% and 3% sucrose. A robust protocol for culturing meristems of Sorghum is required to assist with rapid genetic improvement of the genus. Through meristem culture, an efficient method for rapid micropropagation was developed for Sorghum bicolor (L.) Moench. hybrids, namely NC+262, NC+6C21 and NC+6B50. Complete plants were regenerated directly from shoot meristems without an intervening callus phase. Regeneration frequencies varied between the studied genotypes and according to the growth regulator combinations present in the medium. The combination of BAP and TDZ showed a synergistic effect on shoot multiplication. The induction and control of in vitro somatic embryogenesis is dependent of a number of factors. The genotype of the mother plant donor of explants and the kind of auxin play essential roles in conferring embryogenic competence.
