Influence of genotype and explant source on indirect organogenesis by in vitro culture of leaves of Melia azedarach L. Biocell

In vitro regeneration of shoots from leaf explants of the Paradise tree (Melia azedarach L.) was studied. Three different portions (proximal portion, distal portion and rachis of the leaflets) of three developmental stages (folded, young still expanding and completely expanded) of leaves of 10 - 15 years old plants of seven genotypes were cultured on Murashige and Skoog (1962) medium (MS) supplemented with 1mg.l-1 benzylaminopurine (BAP) + 0.1mg.l-1 kinetin (KIN) + 3 mg.l-1 adenine sulphate (ADS). The rachis of the leaflets of the completely expanded leaves was found to be the most responsive tissue, in most of the genotypes employed. Rapid in vitro plant regeneration from leaf explants of Launaea sarmentosa (Willd.) Sch. Bip. ex Kuntze Biol. Res.

An efficient protocol for organogenesis through leaves has been established for Launaea sarmentosa (Willd.) Sch. Bip. ex Kuntze, a highly valuable medicinal plant. The leaf explants produced microshoots on MS basal medium when fortified with cytokinins and auxins. A combination of 6-benzylaminopurine (BAP) at 0.5mg/l and naphthaleneacetic acid (NAA) at 0.2mg/l resulted in the induction of high frequency microshoots in 30 days. The microshoots were successfully subcultured for shoot elongation and eventually for rooting on MS medium supplemented with indole-3-butyric acid (IBA) at 0.5mg/l. The regenerated plantlets were hardened under greenhouse conditions and transferred to garden, resulting in a 90% survival rate.

Plant regeneration of natural tetraploid Trifolium  Hum pratense L Biol. Res.

The regeneration of natural tetraploid T. pratense, originated from Erzurum-Turkey, is reported in this study. This plant has low seed setting and hard seed problems due to polyploidy. Hypocotyl, cotyledon, apical meristems, epicotyl and young primary leaves were inoculated on MS and PC-L2 media containing different concentrations of BAP and NAA as growth regulators. The best shoot formation has been observed on explants initiated from apical meristem placed on PC-L2 medium that includes 2 mg dm-3 BAP and 1 mg dm-3 NAA. 94.4% of the shoots originated from calli were rooted on PC-L2 medium with 1 mg dm-3 NAA. Morphogenetic response of cotyledon and leaf explants of melon (Cucumis melo L.) cv. Amarillo Oro BABT

Callus cultures from cotyledon and leaf explants of a Spanish cultivar of melon (Amarillo Oro) were tested for growth and morphogenic capacity on several culture media with different concentrations of IAA (indole-3-acetic acid) in combination with 1.0 mg.L-1 BA (6-benzylaminopurine) or 6.0 mg.L-1 KIN (kinetin). The best results were achieved with cotyledon explants. The leaf explants presented low bud formation capacity. Bauhinia forficata link shoot regeneration: histological analysis of organogenesis pathway BABT

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. Histological studies indicated that in vitro plant regeneration in B. forficata occurred through indirect organogenesis. Organogenesis from in vitro-derived leaf and internode explants of Hoya wightii ssp. palniensis -a vulnerable species of Western Ghats BABT

An efficient system was developed for indirect plant regeneration from in vitro-derived leaf and internode explants of Hoya wightii ssp. palniensis. Maximum percentage of the organogenic callus was obtained on MS medium supplemented with NAA (1.0 mg/l) and 2,4-D (2.0 mg/l). The best shoot bud induction was observed on MS medium with BA (1.0 mg/l) +IBA (0.5 mg/l). The coconut water (15%) was better, resulting in a differentiation of the shoot initials in to well-developed shoots. Growth curve and development of the internal calli structure of Eucalyptus camaldulensis Dehn BABT

The objective of this work was to elucidate the growth curve of Eucalyptus camaldulensis Dehn. calli analyzing their anatomical modifications. A sigmoid aspect of the growth curve of the calli fresh matter was observed, with five different phases (lag, exponential, linear, deceleration and decline). In the lag phase, the highest growth percentage 87%, was observed, which reduced during the evaluation period to 17% in the linear phase. As for the anatomical analyses, cellular multiplications was observed during the lag and exponential phases and increase in cell size during the linear phase, promoting the calli volume growth and the establishment of the globular conformation.

Plant regeneration from cotyledonary explants of Eucalyptus camaldulensis dehn and histological study of organogenesis in vitro BABT

The present work aimed at regenerating plants of Eucalyptus camaldulensis from the cotyledonary explants and describing the anatomy of the tissues during callogenesis and organogenesis processes, in order to determine the origin of the buds. The cotyledonary leaves of E. camaldulensis were cultured in Murashige and Skoog (MS), WPM and JADS media supplemented with 2.7 µM NAA and 4.44 µM BAP. The best results for bud regeneration were obtained on MS and WPM media (57.5 and 55% of calluses formed buds, respectively). Different concentrations of 6-benzyladenine (6-BA) were used in vitro to induce buds in three types of explants: leaf discs, root segments and the seedling itself obtained from in vitro seed germination of Passiflora cincinnata Mast. Callus induction and plant regeneration of tomato (Lycopersicon esculentum cv. IPA 5) via anther culture Ciência Rural

Different growth regulators combinations were tested on the production of anther callus in tomato cultivar IPA 5. Calli were induced on media supplemented with 1.0mgL-1 gibberellic acid (GA3), 0.05mgL-1 alpha-naphthaleneacetic acid (NAA) plus 0.1mgL-1 6-benzylaminopurine (BAP), or with 1.0mgL-1 BAP plus 1.0mgL-1 NAA. A shoot regeneration system for Plectranthus neochilus was studied from leaf explants. Leaves developed under in vitro conditions were cultured on Wood Plant Medium supplemented with 0.2mg dm-3 α-naphthaleneacetic acid (NAA) and different 6-benzilaminopurine (BAP) or thidiazuron (TDZ) concentrations (0, 1.5, 3.0, 4.5 and 6.0mg dm-3).
