The regeneration of natural tetraploid T. pratense, originated from Erzurum-Turkey, is reported in this study. This plant has low seed setting and hard seed problems due to polyploidy. Hypocotyl, cotyledon, apical meristems, epicotyl and young primary leaves were inoculated on MS and PC-L2 media containing different concentrations of BAP and NAA as growth regulators. The best shoot formation has been observed on explants initiated from apical meristem placed on PC-L2 medium that includes 2 mg dm-3 BAP and 1 mg dm-3 NAA. 94.4% of the shoots originated from calli were rooted on PC-L2 medium with 1 mg dm-3 NAA. In vitro organogénesis has been accomplished in the natural tetraploid T. pratense regenerated plants successively transferred to the field |

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. Histological studies indicated that in vitro plant regeneration in B. forficata occurred through indirect organogenesis. Meristemoids consisting of small cells with dense cytoplasm and prominent nuclei were randomly distributed throughout the callus surface indicating early stages of shoot bud differentiation. Shoots developed de novo from... |

The objective of this work was to elucidate the growth curve of Eucalyptus camaldulensis Dehn. calli analyzing their anatomical modifications. A sigmoid aspect of the growth curve of the calli fresh matter was observed, with five different phases (lag, exponential, linear, deceleration and decline). In the lag phase, the highest growth percentage 87%, was observed, which reduced during the evaluation period to 17% in the linear phase. As for the anatomical analyses, cellular multiplications was observed during the lag and exponential phases and increase in cell size during the linear phase, promoting the calli volume growth and the establishment of the globular conformation. |

Different concentrations of 6-benzyladenine (6-BA) were used in vitro to induce buds in three types of explants: leaf discs, root segments and the seedling itself obtained from in vitro seed germination of Passiflora cincinnata Mast. The 0.5 mg.L -1 6-BA concentration was most suitable for all the three explants, however, bud formation time and means (direct/indirect) were different for each type of explant. In seedlings used as explants, it had shoot regeneration from the primary root. Histological events leading to bud formation via meristemoids were described. The origin was indirect in leaf discs, with callus formation in subepidermal cells of the chlorophyll parenchyma layers. In root segments and in seedling roots, the buds were formed directly in... |

H. Mancilla, K. Quiroz, A. Arencibia, B. Carrasco, and R. García-Gonzales. 2013. In vitro culture of Luma chequen from vegetative buds. Cien. Inv. Agr. 40(3): 609-615. Luma chequen, a small tree or large shrub belonging to the Myrtaceae family, is endemic to South America and has medicinal, nutritional and ornamental potential. However, its native habitat is deteriorating gradually, and it is suffering from the effects of fragmentation that is being caused by the conversion of forest land to agricultural land and the natural expansion of monocultural plantations of exotic species, such as Pinus radiata. The purpose of this work is to develop an effective procedure for establishing in vitro cultures of the native Chilean species L. chequen. Aseptic nodal... |

A shoot regeneration system for Plectranthus neochilus was studied from leaf explants. Leaves developed under in vitro conditions were cultured on Wood Plant Medium supplemented with 0.2mg dm-3 α-naphthaleneacetic acid (NAA) and different 6-benzilaminopurine (BAP) or thidiazuron (TDZ) concentrations (0, 1.5, 3.0, 4.5 and 6.0mg dm-3). An increase in percentage of responsive explants (85.3%) and in the number of shoots developed per explant (3.2) was observed when the explants were treated with 5.3 and 4.7mg dm-3 BAP, respectively. The leaf explants cultured on media supplemented with TDZ became vitreous and did not form buds. The regeneration system used is efficient for boldo bud induction and shoot development, showing high potential for advanced cellular... |

Syngonanthus mucugensis Giul. subsp. mucugensis is an herbaceous plant with significant economic value in the ornamental dry flower business. The restricted occurrence of the municipality Mucugê-BA, Brazil, exclusively associated with extractive exploitation, has considered this species as endangered. The objective of this work was to evaluate the organogenic potential of three different types of S. mucugensis subsp. mucugensis explants to promote the development of an alternative method to the propagation of the genetic resources of this important plant. The morphogenetic capacities of the leaf, stem and root this species was tested using Murashige and Skoog culture medium at half salt concentration and different concentrations of growth of regulators... |
