The aim of this study was to test the ability of magnetic resonance imaging (MRI) technique to characterize gonad development and to determine the sex of live Pacific oysters through their shells. A preliminary nuclear magnetic resonance (NMR) retaxometry study was conducted to characterize T-1 and T-2 NMR relaxation parameters for the main oyster organs. This showed that T-1-weighted MRI sequences were most appropriate to optimize contrasts between tissues in images. The results showed that gray levels of gonads in images acquired with gradient-echo sequence were variably affected by T-2* weighting effect. Herbicides are one of the major classes of pollutants contaminating coastal waters over the world. Among them, diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is a phenylurea herbicide frequently detected in oyster-producing area, known to be toxic for this important exploited non-target species. Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. Estuaries and coastal areas are often subjected to much pollution, including pesticides and polycyclic aromatic hydrocarbons (PAHs) produced by anthropic activities. Bay of Marennes-Oléron is an estuarine coastal zone in which there is a high level of aquacultural production and this has adverse effects in aquatic organisms. The chemiluminescence technique has been used to estimate the phagocytosis capacity of Crassostrea gigas hemocytes according to different culture media and times. We describe two methods of producing viable and fertile autotetraploid Pacific oyster (Crassostrea gigas Thunberg) based on the use of normal-sized oocytes produced by normal diploid females. Our methods showed that the oocyte size is not a limiting factor for the success of the induction to autotetraploidy. These methods offer means of direct introgression of genetic progress from elite diploid lines to tetraploids used as broodstock, avoiding a triploid step with the risk of transferring undesirable traits from highly fecund triploids. This study reports mortality under laboratory conditions in unselected controls and two lines of juvenile Pacific oysters Crassostrea gigas, previously selected for their high or low survival in the field during the summer period.
