Oxidation of the albumin thiol to sulfenic acid and its implications in the intravascular compartment BJMBR

Human serum albumin (HSA) is the most abundant protein in the intravascular compartment. It possesses a single thiol, Cys34, which constitutes ~80% of the total thiols in plasma. This thiol is able to scavenge plasma oxidants. A central intermediate in this potential antioxidant activity of human serum albumin is sulfenic acid (HSA-SOH). Work from our laboratories has demonstrated the formation of a relatively stable sulfenic acid in albumin through complementary spectrophotometric and mass spectrometric approaches. Stability indicating RP-HPLC method for simultaneous determination of gatifloxacin and dexamethasone in binary combination BJPS

Abstract In this study, conditions were optimized for development of a simple RP-HPLC method for simultaneous analysis of gatifloxacin and dexamethasone in different matrices like pharmaceuticals, human serum and urine. Good separation of gatifloxacin and dexamethasone from the induced degradation products was accomplished using C8 as stationary phase; 0.02 M phosphate buffer (pH 3.0) and methanol (42:58 v/v) as mobile phase. The concentration was measured with DAD at 270 nm. Linearity was observed in the range of 0.000040-0.000280 mol/L for gatifloxacin (r2≥0.999) and 0.000013-0.000091 mol/L for dexamethasone (r2≥0.999). Development of a new HPLC method for in vitro and in vivo studies of haloperidol in solid lipid nanoparticles BJPS

ABSTRACT A simple and sensitive HPLC method was developed and validated for the quantification of haloperidol in solid lipid nanoparticles (SLNs). The developed method was used for detection of shelf life of haloperidol in SLNs. Calibration curve of haloperidol was also constructed in rat plasma using loratidine as internal standard. In vivo studies were performed on rats and concentration of haloperidol in brain and blood was measured for the determination of various pharmacokinetic and hence brain targeting parameters. Chromatogram separation was achieved using C18 column as stationary phase. The mobile phase consisted of 100 mM/L potassium dihydrogen phosphate-acetonitrile-TEA (10:90:0.1, v/v/v) and the pH was adjusted with o-phosphoric acid to 3.5....

Determination of Gemifloxacin in human plasma by high performance liquid chromatography using Ultra Violet detector and its application to a bioequivalence study BJPS

A liquid chromatography method was developed and validated for the determination of gemifloxacin in human plasma using chloramphenicol as internal standard to achieve lower quantification limit. Acetonitrile was used to precipitated and extracted analyte and internal standard from plasma by Protein Precipitation. Analysis was performed isocratically on C18 column using 25% acetonitrile and 75% 0.02 M phosphate buffer as mobile phase. The method was demonstrated to be linear from 0.003 µg/mL to 5 µg/mL with the lower limit of quantitation of 0.003 µg/mL. The method was successfully applied for the bioequivalence study of gemifloxacin after a single oral administration of 320 mg gemifloxacin mesylate tablets to 12 healthy volunteers.

Eletroforese das proteínas plasmáticas em emas (Rhea americana) de diferentes faixas etárias Ciência Rural

A rheacultura vem crescendo em todo o Brasil. Porém, ainda há uma escassez de informações sobre esta espécie e de médicos veterinários especializados na área. Este trabalho teve como objetivo verificar as diferenças existentes na eletroforese plasmática (gel de agarose) em idades distintas de emas (Rhea americana). Utilizaram-se 45 emas, separadas em quatro grupos: grupo 1 (n=10), animais com 15 dias de idade; grupo 2 (n=10), animais com 30 dias; grupo 3 (n=10), animais com 45 dias e grupo 4 (n=15), animais com 1 ano. Verificou-se homogeneidade nos valores eletroforéticos analisados nas aves dentro de cada faixa etária. Currently, the use of alternative energy ingredients for horses has increased because these foods contain "superfibers", making safer diet for these animals. To study the influence of diets containing increasing levels of citrus pulp on albumin, triglycerides, cholesterol, glucose, insulin and short chain fatty acid (SCFA) concentrations in the blood, five animals were used, with an average age of 3.5 years and a live weight of 460.66±76.86 kg, they were fed twice a day, at 7:00 am and 4:00 pm. It was intended with the present work to verify the effect of phytase on the performance, bone mineralization and content of plasma urea of swine. 80 swine (30 ± 3.1 kg/LW), crossbred (LD x LW) males and females, allocated to a randomized block design (RBD) with four treatments and five replicates. The treatments consisted of a growing swine diet on the basis of corn, soybean meal and defatted rice bran (DRB) supplemented with four levels of phytase (0, 400, 800 and 1200 FTU/kg) The diet was formulated to meet the requirements of growing swine except for available calcium and phosphorus, the levels of which were 0.64 and 0.25%, respectively.
