Effect of the kappa-casein gene polymorphism, breed and seasonality on physicochemical characteristics, composition and stability of bovine milk R. Bras. Zootec.

The objective of this study was to evaluate the effect of genetic polymorphism of kappa-casein, breed and seasonality on the physicochemical characteristics, composition and stability of milk in commercial dairy herds. A total of 879 milk and blood samples were collected from 603 Holstein and 276 Girolando cows, obtained during rainy and dry seasons. Milk samples were analyzed to determine the physicochemical characteristics, composition and ethanol stability, while blood samples were subjected to polymerase chain reaction to identify the kappa-casein genotype. PORCINE PARVOVIRUS INFECTIONS IN CUBA Rev Salud Anim.

To obtain information about the porcine parvovirus (PPV) infection status of pigs in Cuba, tissue samples collected from ill pigs were evaluated using polymerase chain reaction (PCR). The PCR analysis showed the presence of PPV in 12 of the 34 (35.3%) field clinical samples assessed. Pigs from three of the seven swine herds of six different geographic regions were detected to be positive for PPV. A field PPV strain was isolated for the first time in the country. A swine herd showed 57 of 60 sera sampled (95%) positive for PPV specific antibodies by a commercial ELISA. This study is the first report of PPV infecting pigs in Cuban swine herds.

The objective of this research was to develop a primer for a polymerase chain reaction specific for Xylella fastidiosa strains that cause Pierce's Disease (PD) in grapes (Vitis vinifera). The DNA amplification of 23 different strains of X. fastidiosa, using a set of primers REP1-R (5'-IIIICGICGIATCCIGGC-3') and REP 2 (5'-ICGICTTATCIGGCCTAC-3') using the following program: 94 ºC/2 min; 35 X (94 ºC/1 min, 45 ºC/1 min and 72 ºC/1 min and 30 s) 72 ºC/5 min, produced a fragment of 630 bp that differentiated the strains that cause disease in grapes from the other strains. ABSTRACT Invasion of trophoblast into endometrium is vital for successful pregnancy development. MMP9 and uPA are key proteases in this process, but it is still not clear the regulation of its expression by Transforming Growth Factor Beta (TGF-β), a known negative regulator of trophoblast invasion. We evaluated the effect of TGF-β on the transcriptional expression of uPA and MMP9 over time, in HTR-8/SVneo trophoblast cells cultured with or without 0.5 % fetal bovine serum, via RT qPCR. This study evaluated an indirect immunofluorescence assay (IFA) to detect feline immunodeficiency virus infection (FIV) antibody in a comprehensive epidemiological survey of FIV in Argentina. IFA modified in our laboratory, was compared with two other immunoassays, western blot (WB) and a sandwich immunochromatographic commercial kit (SI), and also with a direct polymerase chain reaction (PCR) method that detects proviral DNA. IFA showed to be a test with high sensitivity and specificity, and could be useful as a diagnostic tool in epidemiological studies. The presence of a low percentage of results with non-specific reactivity in the IFA could be resolved with further testing or use of an alternative method.

A técnica da reação em cadeia pela DNA polimerase (PCR) foi usada para a amplificação rápida de um fragmento de 456bp da região única curta (Us) do genoma do BHV-1. Iniciadores de 18pb do gene da ORF1 foram usados para a amplificação das amostras-padrão e brasileiras. Uma amplificação clássica não foi bem sucedida. ABSTRACT Mastitis caused by Mycoplasma spp., regardless of species, are considered highly contagious pathogens and, usually was not responsive to antimicrobial therapy. Five dairy herds, comprising 489 animals and 1,956 mammary glands, were used in this study. Milk samples were obtained from bulk tanks and subjected to polymerase chain reaction (PCR) for the identification of Mollicutes, Mycoplasma spp., and Mycoplasma bovis. Moreover, individual samples from cases of clinical and subclinical mastitis in quarters of the dairy herds’ animals that yielded a positive PCR upon bulk tank analysis were subjected to molecular analysis. ABSTRACT Meningoencephalitis caused by Bovine herpesvirus 5 (BoHV-5) is an important neurological disease that affects Brazilian cattle herds. The present study investigated the presence of BoHV-5 DNA in cattle diagnosed with meningoencephalitis at Faculdade de Medicina Veterinária e Zootecnia, UNESP - Univ Estadual Paulista from 1980 to 2009. Investigation of natural infection by Leishmania in sandflies of Paraná State, Southern Brazil BABT

The purpose of this work was to verify the occurrence of Leishmania in naturally infected sandflies. The insects were collected with Falcão, Shannon and HP light-traps, in Doutor Camargo and Maringá municipalities between November 2004 and October 2005. Of the 11,033 sandflies collected in Doutor Camargo, 2,133 surviving females were dissected, particularly those of the Nyssomyia neivai species (86.87%). Analysis of seven STR human loci for paternity testing by microchip electrophoresis BABT

The aim of this work was to evaluate two paternity cases by microchip electrophoresis and the validation of the methodology by comparison of the results with those obtained in a commercial genetic analyzer. It was observed that when working with tetranucleotide regions, in which the minimal difference between the alleles was only four base pairs, the commercial microchip system did not present the resolution and repeatability needed. Nevertheless, the relative standard deviation was between 0 and 1.2% and the fragments detected were within the expected size ranges as described in the literature.

Development of a polymerase chain reaction and its comparison with agar gel immunodiffusion test in the detection of bovine leukemia virus infection Braz. J. Vet. Res. Anim. Sci.

Polymerase chain reaction (PCR) was used for bovine leukemia virus (BLV) detection in the peripheral leukocytes of the infected bovines. The primers used were designed to amplify a part of env gene of BLV. PCR products were analyzed by agarose gel electrophoresis stained by ethidium bromide. The analytical specificity of PCR was confirmed by enzymatic restriction analysis of the PCR product with Bam HI and also by nucleotide sequence analysis of three PCR samples. Sixty five animals were tested for anti-BLV antibody, by agar gel-immunodiffusion test (AGID) and for direct BLV detection by PCR. There was a 73.80% concordance rate between the two tests. Enteropathogenic Escherichia coli (EPEC) comprise one of the six categories of diarrhoeagenic E. coli (DEC). EPEC is subgrouped into typical (tEPEC) and atypical (aEPEC). The identification of DEC cannot be based only on cultural and biochemical criteria, since they are indistinguishable from the non-pathogenic E. coli commonly found in human feces. Characterization of rickettsia rickettsii in a case of Fatal Brazilian spotted fever in the city of Rio de Janeiro, Brazil BJID

A lethal case of Brazilian spotted fever (BSF) is presented. Clinical features were initially of gastrointestinal involvement and evolved with progression to septic shock, meningoencephalitis and death on the 6th day of illness. Indirect immunofluorescence assay (IFA) for spotted fever group rickettsia (SFGR) was non-reactive. Diagnosis was confirmed by the polymerase chain reaction (PCR) and the nucleotide sequencing of a fragment of the ompA gene showed 100% homology to Rickettsia rickettsii. Comparison of different primes for PCR-based diagnosis of cutaneous leishmaniasis BJID

OBJECTIVE: The objective of this study was to analyze different primers that are commonly used in epidemiological studies for the detection of Leishmania DNA by PCR, and to compare them to the conventional direct parasite search for American cutaneous leishmaniasis (ACL) diagnosis. MATERIAL AND METHODS: Five pairs of primers, four of them derived from Leishmania kDNA sequences (MP3H-MP1L; B1-B2; LBF1-LBR1; 13A-13B), and one derived from the SL RNA (mini-exon) gene repeat (LU5A-LB3C), reported previously, were used. RESULTS: The MP3H-MP1L primers were the best at amplifying the DNA, detecting 2 fg of Leishmania spp. DNA. The 13A-13B primers presented the worst performance, detecting 512 x 10³ fg of DNA.
