Amin,Adnan; Khan,Muhammad Ayaz; Ehsanullah,Malik; Haroon,Uzma; Azam,Sheikh Muhammad Farooq; Hameed,Abdul.
A total of 112 soil samples were taken from differents areas of district D.I.Khan and Kohat (KPK) Pakistan and screened for production of antibiotics against the Micrococcus luteus and Staphylococcus aureus. Widest zone of inhibition (18mm) was produced by microorganism isolated from saline soil. The strain was later identified as Bacillus GU057 by standard biochemical assays. Maximum activity (18mm inhibition zone) was observed against Staphylococcus aureus after 48 hours of incubation at pH 8 and 4% concentration of glucose. The antibiotic was identified by autobiography as bacitracin. The Bacillus strain GU057 was confirmed as good peptide antibiotic producer and can effectively be indulged as biocontrol agent.

A rapid screening method for cutinase producing microorganisms BJM
Macedo,Gabriela Alves; Pio,Tatiana Fontes.
Both cutinase and lipase belong to the esterase group of enzymes (EC 3.1.1.X), which are capable of catalyzing the hydrolysis of ester bonds. Cutinase catalyzes the hydrolysis of cutin, an insoluble biopolyester which is the structural component of plant cuticles. As cutinase is an efficient catalyst in watery or organic media, it is potentially appropriate for the detergent, food and cosmetic industries. The objective of this work was to isolate microorganisms from plants and perform a pre-selection of molds showing esterase producing ability. The selected strains were then submitted to fermentation in media containing cutin. The lipolytic and cutinolytic activities of the supernatant were determined in order to differentiate lipase producers from...

Physiological and biochemical traits as tools to screen sensitive and resistant varieties of tomatoes exposed to salt stress Braz. J. Plant Physiol.
Fariduddin,Qazi; Mir,Bilal Ahmad; Ahmad,Aqil.
The present study was conducted to evaluate salt tolerance in seven different pure-line cultivars of tomato (Solanum lycopersicum L.) viz. K-21, Pusa Ruby, Pusa Gorav, Hera research, Selection N5, PKM-1 and S-22 based on several physiological and biochemical traits. Seedlings were transplanted to the pots, being exposed to different salinity levels in the form of NaCl (0, 50, 100, or 150 mM) at a 35-day stage of growth for six days. The plants exposed to salt stress presented a significant decline in growth, photosynthetic parameters, maximum quantum yield of PSII and leaf water relations, which were drastically reduced in variety S-22, while variety K-21 was the least affected. Electrolyte leakage was superior in proportion to an increase in salinity...

Evaluation of Resistance of Cucumber Cultivars to the Vegetable Leafminer (Liriomyza sativae Blanchard) (Diptera: Agromyzidae) in Greenhouse Chilean J. Agric. Res.
Basij,Moslem; Askarianzaeh,Alireza; Asgari,Shahriyar; Moharramipou,Saeid; Rafezi,Ramin.
The leafminer, Liriomyza sativae Blanchard, is distributed around the world and is an important pest of vegetables and ornamentals. Given the resistance potential of the leafminer to current insecticides, the use of resistant plant cultivars and parasitoids could be effective integrated pest management (IPM) strategies against it. Cucumber (Cucumis sativus L.) is a preferred host for this insect. Seventeen cultivars of cucumber have been evaluated to study resistance mechanisms to L. sativae. All cucumber cultivars were evaluated in screening tests in greenhouse with indices such as the number of leafminer stings, the number of larval mines, the proportion of larval mines to leafminer stings, and the rate of injury. Significant differences (p < 0.01)...

An alternative method for screening lactic acid bacteria for the production of exopolysaccharides with rapid confirmation Ciênc. Tecnol. Aliment.
Paulo,Elinalva Maciel; Vasconcelos,Murilo Pinho; Oliveira,Ivelise Santiago; Affe,Helen Michelle de Jesus; Nascimento,Rosely; Melo,Itamar Soares de; Roque,Milton Ricardo de Abreu; Assis,Sandra Aparecida de.
The accumulation of exopolysaccharides (EPS) produced by microorganisms occurs in the presence of excess substrate and limiting conditions of elements that are essential to growth, such as nitrogen, phosphorus, sulfur, and magnesium. The presence of EPS produced by bacterial cells contributes to slime colonies formation in solid medium and increased viscosity in liquid medium. This paper proposes an alternative method for screening EPS-producing lactic acid bacteria using solid medium-containing discs of filter paper that are saturated with active cultures. The screening was carried out under different culture conditions varying the type of sugar, pH, and temperature. EPS production was visualized by the presence of mucoid colonies on the discs, which was...

The lift pool method for isolation of cDNA clones from lambda phage libraries Electron. J. Biotechnol.
LeBlanc-Straceski,Janine; Sobrado,Pablo; Betz,Sharon; Zerfas,Julie; Morgan,Karen.
A PCR based strategy was developed to screen a Xenopus oocyte λgt10 cDNA library. The PCR-based lift pool (LP) method follows the same two tiered strategy as conventional screening of phage libraries by filter hybridization. Two rounds of plating, one at high density to detect the clone, and one at low density to purify the clone to homogeneity, are performed. In the first round, lysates from high density plates, termed plate pools (PP), serve as template for PCR. In the second round, phage particles adhering to plaque lifts of low density plates are washed off nitrocellulose filters to create LPs, which are used as template for PCR. The integrity of the plaques on the low-density plates is preserved. Once a positive LP has been identified,...

A reliable begomovirus inoculation method for screening Lycopersicon esculentum lines Horticultura Brasileira
Inoue-Nagata,Alice K; Nagata,Tatsuya; Ávila,Antonio C de; Giordano,Leonardo de B.
Due to failures of infection after mechanical inoculation of begomovirus on tomato (Lycopersicon esculentum Mill.) plants, an efficient and reliable begomovirus inoculation method was developed using the whitefly vector (Bemisia tabaci). Virus acquisition was carried out using a 'Tube-cage', made of a polypropylene tube, containing one to two day-old whiteflies and a tomato plant apex infected with a begomovirus isolate. After 48 h of acquisition access period, inoculation was done using a 'Ring-cage', containing viruliferous whiteflies, attached to the abaxial side of the leaf of young tomato plants. 48 hours after acquisition, the whiteflies were eliminated and the inoculated plants were incubated for 28 days under greenhouse conditions. Three...

Search for sources of resistance to Meloidogyne enterolobii in commercial and wild tomatoes Horticultura Brasileira
Silva,Alysson J da; Oliveira,Gustavo HF de; Pastoriza,Rhuan JG; Maranhão,Eduardo HA; Pedrosa,Elvira MR; Maranhão,Sandra RVL; Boiteux,Leonardo S; Pinheiro,Jadir B; Carvalho Filho,José Luiz S de.
ABSTRACT Meloidogyne enterolobii (=M. mayaguensis) is an emerging plant pathogen capable of inducing root galls and yield reduction in a wide range of host species. This pathogen has also been reported as a global threat for tomato (Solanum lycopersicum) crop production mainly due to its ability to overcome the resistance meditated by the Mi-1 gene. Despite the potential importance of this nematode, sources of resistance to M. enterolobii are not yet available for breeding purposes. The main objective of the present work was to evaluate a large Solanum (section Lycopersicum) germplasm (comprising nine species and one botanic variety) aiming to identify useful sources of resistance to M. enterolobii. In the first screening assay, 101 accessions and the...

Lactic acid bacteria selection for biopreservation as a part of hurdle technology approach applied on seafood ArchiMer
Wiernasz, Norman; Cornet, Josiane; Cardinal, Mireille; Pilet, Marie-france; Passerini, Delphine; Leroi, Francoise.
As fragile food commodities, microbial and organoleptic qualities of fishery and seafood can quickly deteriorate. In this context, microbial quality and security improvement during the whole food processing chain (from catch to plate), using hurdle technology, a combination of mild preserving technologies such as biopreservation, modified atmosphere packaging and superchilling, are of great interest. Thirty-five lactic acid bacteria (LAB) belonging to the species Carnobacterium maltaromaticum, Carnobacterium inhibens, Lactococcus piscium, Leuconostoc gelidum, Vagococcus fluvialis, Vagococcus penaei and Aerococcus viridans, known to possess interesting antimicrobial activity, were chosen for their potential application as bioprotective agents as a part of...

Is generalization of exhaled CO assessment in primary care helpful for early diagnosis of COPD? ArchiMer
Molinari, Nicolas; Abou-badra, Mathieu; Marin, Gregory; Ky, Chin-long; Amador, Noemi; Gamez, Anne Sophie; Vachier, Isabelle; Bourdin, Arnaud.
Background: COPD is largely under-diagnosed and once diagnosed usually at a late stage. Early diagnosis is thoroughly recommended but most attempts failed as the disease is marginally known and screening marginally accepted. It is a rare cause of concern in primary care and spirometry is not very common. Exhaled carbon monoxide (eCO) is a 5-seconds easy-to-use device dedicated to monitor cigarette smoke consumption. We aimed to assess whether systematic eCO measurement in primary care is a useful tool to improve acceptance for early COPD diagnosis. Methods: This was a two-center randomized controlled trial enrolling 410 patients between March and May, 2013. Whatever was the reason of attendance to the clinic, all adults were proposed to measure eCO during...

Lutte contre les salissures marines : approche par procédés enzymatiques. ArchiMer
Leroy, Celine.
Fouling on marine underwater surfaces causes critical and economic problems such as important material biodamages and industrial performances reduction. We chose to test antifouling potential of enzymatic commercial preparations like hydrolases (proteases, glycosidases and lipases) in order to inhibit the first fouling adhesion step: bacterial biofilm formation. An evaluation test of antifouling properties onto marine bacterial adhesion was designed using a mono-incubation of Pseudoalteromonas sp. D41 in microtiter plate and in sterile natural sea water. This test was adapted to screen agents for bacterial adhesion removal or inhibition activities and allowed to test enzymatic preparations toxicity on non adhered bacteria. Inhibition rates according to...

A marine bacterial adhesion microplate test using the DAPI fluorescent dye: a new method to screen antifouling agents ArchiMer
Leroy, Celine; Delbarre Ladrat, Christine; Ghillebaert, F; Rochet, Marie-joelle; Compere, Chantal; Combes, D.
Aims: To develop a method to screen antifouling agents against marine bacterial adhesion as a sensitive, rapid, quantitative microplate fluorescent test. Methods and Results: Our experimental method is based on a natural biofilm formed by mono-incubation of the marine bacterium Pseudoalteromonas sp. D41 in sterile natural sea water in a 96 wells polystyrene microplate. The 4'6-diamidino-2-phenylindole (DAPI) dye was used to quantify adhered bacteria in each well. Total measured fluorescence in wells was correlated with the amount of bacteria showing a detection limit of one bacterium per 5 µm2 and quantifying 2 x 107 to 2 x 108 bacteria adhered per cm2. The antifouling properties of three commercial surface active agents and chlorine were tested by this...

Demonstrating the need for chemical exposure characterisation in a microplate test system: Toxicity screening of sixteen pesticides on two marine microalgae ArchiMer
Dupraz, Valentin; Stachowski-haberkorn, Sabine; Wicquart, Jérémy; Tapie, Nathalie; Budzinski, Hélène; Akcha, Farida.
Pesticides used in viticulture create a potential risk for the aquatic environment due to drift during application, runoff and soil leaching. The toxicity of sixteen pesticides and one metabolite were evaluated on the growth of two marine microalgae, Tisochrysis lutea and Skeletonema marinoi, in 96-h exposure assays conducted in microplates. For each substance, concentrations of stock solutions were analytically measured and abiotic assays were performed to evaluate the chemical stability of pesticides in microplates. For two chemicals, microalgae exposures were run simultaneously in microplates and culture flasks to compare EC50 calculated from the two exposure systems. Results from chemical analyses demonstrated the low stability of hydrophobic...

Screening of marine algal extracts for anti-settlement activities against microalgae and macroalgae ArchiMer
Hellio, Claire; Berge, Jean-pascal; Beaupoil, Claude; Le Gal, Yves; Bourgougnon, Nathalie.
The ban on the use of TBT-based antifouling paints for boats under 25 m in length has lead to a search for new non-toxic antifoulants. One of the most promising alternative technologies to heavy metal based antifouling paint is the development of antifouling coatings whose active ingredients are naturally occurring compounds from marine organisms. This is based on the principle that marine organisms are also facing the problem of the presence of epibionts on their own surface. In this study, the antifouling activity of a series of aqueous, ethanolic and dichloromethane extracts from 30 algae from the North East Atlantic coast was investigated. The extracts were tested in laboratory assays against species representatives of two major groups of fouling...

Photo-protective compounds in red macroalgae from Brittany: Considerable diversity in mycosporine-like amino acids (MAAs) ArchiMer
Lalegerie, Fanny; Lajili, Sirine; Bedoux, Gilles; Taupin, Laure; Stiger-pouvreau, Valerie; Connan, Solène.
To cope with the biotic and abiotic stresses experienced within their environment, marine macroalgae have developed certain defence mechanisms including the synthesis of photo-protective molecules against light and particularly harmful UV radiation. The aim of this study was to screen selected red algae, a highly diverse phylogenetic group, for the production of photo-protective molecules. The pigment content and composition (i.e. chlorophyll-a, phycobiliproteins and carotenoids) and the composition of mycosporine-like amino acids (MAAs) were studied in 40 species of red macroalgae collected in Brittany (France), at two distinct periods (i.e. February and July 2017). A high inter-specific variability was demonstrated in terms of pigment content and MAA...

Competing Screening Rules AgEcon
Hendrikse, George W.J.; Hu, Yamei.
Various studies show that agricultural cooperatives behave differently than their investor-owned counterparts. One explanation may be that the internal decision making process differs in these two governance structures. A model is developed to explore how endogenous screening rules affect efficient organizational choices and industrial structures. It is shown that screening level choice may outweigh architecture choice and that screening rules are strategic substitutes. Conditions are derived under which cooperatives are efficient organizational forms. It is also shown that competition may increase the attractiveness of investor-owned firms and circumstances are determined in which cooperatives and investor owned firms coexist in equilibrium.
