Correlation between sperm characteristics and testosterone in bovine seminal plasma by direct radioimmunoassay R. Bras. Zootec.

The objectives of this study were to validate a non-extractive RIA for seminal testosterone and quantify the hormone using a solid-phase commercial kit, and study the correlation between testosterone in seminal plasma and sperm characteristics. Parallelism showed a correlation index r = 0.992 (Y = -5.47 + 1.073X; R² = 0.985), indicating that the non-extractive method presented is indicated particularly for assessment of testosterone when establishing comparisons between samples. Overall mean (±SD) of testosterone level was 0.60±0.65 ng/mL. Correlation was only found between the seminal concentrations of testosterone and pH of the semen.

Testicular shape and andrological aspects of young Nellore bulls under extensive farming R. Bras. Zootec.

Aiming to determine the relationship between the frequency of testicular shape and the andrological aspects in young Nellore bulls, 18,676 animals were assessed. All andrological examinations were performed between the years 2000 and 2008. Animals were classified as able for breeding, able for breeding in natural mating system, unable for breeding and discarded. The testicular shape was classified as long, fairly long, oval-long, spherical-oval, and spherical. The analysis of Pearson correlations was performed for testicular shape with scrotal circumference, testicular volume, progressive motility, sperm vigor, major defects, minor defects and total defects. Study on the viability of cooling canine semen using milk and egg yolk glycine extenders MV&Z

Twenty-seven ejaculates of an adult, mixed breed, medium size dog were used in this study. The semen was collected by penile stimulation and divided into three equal samples. Group I (G I): cooled without extender; Group 2 (G2): diluted with skimmed milk-based extender; Group 3 (G3): diluted with glycine egg yolk-based extender, without glycerol. Samples were evaluated after collection (MO) as to sperm motility and vigor, and integrity of sperm membranes, and then incubated at 50 C. By understanding the hormonal action behind the induction of spermiation in Leptodactylus ocellatus can help in the collection of semen for artificial insemination and cryopreservation. The experiments were conducted at the Experimental Frog Farm of the Federal University of Viçosa. Seven L. ocellatus males with the following secondary sexual characteristics were selected: black thorns, developed forelimbs, and amplexus reflex. Evaluation of semen characteristics after hormonal induction of the bullfrog could provide valuable information on the gametes of this species, which may be useful for projects related to artificial fertilization, animal improvement, and cryopreservation. Bullfrog males were induced to spermiate with buserelin acetate (GnRHa), and their semen was subsequently analyzed. Abstract: A total number of 300 (225 ducks and 75 drakes) Sudani ducks, 28-wk-old were divided into five groups to investigate the effects of dietary L-carnitine (LC) supplementation on productive, hatching and physiological performance as well as nutrients digestibility coefficients. ABSTRACT This study was designed to evaluate the possible benefits of adding xanthan gum to a standard extender for equine through in vitro analyzes of sperm quality. Semen was collected four times from five different stallions (n= 20 samples) and subjected to cooled storage under different conditions: control (only standard extender) and three different concentrations of xanthan gum (0.01%, 0.12%, and 0.25%) supplemented to the extenders. Standardized sperm concentration and volume:volume extension were compared as dilution rates for canine semen freezing. Six proven stud dogs were submitted to two seminal collections by manual stimulation. Semen was evaluated and extended in tris plus egg-yolk and glycerol according to two different dilution rates. In this experiment, it was defined a protocol of fluorescent probes combination: propidium iodide (PI), fluorescein isothiocyanate-conjugated Pisum sativum agglutinin (FITC-PSA), and JC-1. For this purpose, four ejaculates from three different rams (n=12), all showing motility &gt;80% and abnormal morphology <10%, were diluted in TALP medium and split into two aliquots. One of the aliquots was flash frozen and thawed in three continuous cycles, to induce damage in cellular membranes and to disturb mitochondrial function. Establishing the hypoosmotic swelling test for sperm analysis in collared peccaries (Pecari tajacu) Arq. Bras. Med. Vet. Zootec.

Soluções hiposmóticas com diferentes concentrações (0, 50, 100, 150, 200mOsm/L) foram testadas para a avaliação funcional da membrana espermática de catetos (n=13). Foi verificado que o número de espermatozoides reagidos diminuía (P<0,05) de acordo com o aumento da osmolaridade do meio. A maior porcentagem (71,8%) de espermatozoides reagidos, bem como a menor variação nas respostas osmóticas, foi detectada com o uso de água destilada (0mOsm/L) (P<0,05), a qual também apresentou a menor variação nos resultados, de acordo com os erros padrão verificados. Em conclusão, a água destilada aparenta ser uma solução adequada para o uso no teste hiposmótico para sêmen de catetos.

ABSTRACT The aim of this study was to evaluate the effect of pST injections on metabolism, testicular size, and sperm characteristics in young boars. Sixty 22-day old piglets were divided into two groups: pST (n=30) and Control (n=30). The pST group was submitted to pST injections (90µg/kg body weight) every three days up to 330 days of age. Considering the venereal transmission of visceral leishmaniasis from dogs to bitches, the aim of this study was to verify if the penile surface and smegma from infected dogs can be the source of parasites in bitches. Twelve Leishmania infantum infected dogs had semen and smegma samples collected for submission to PCR identification of the DNA of the parasite. Semen (41.7%) and smegma (50.0%) have similar positive incidence (P>0.05; Fisher's exact test), with 58.3% of the dogs positive for semen and/or smegma samples. The proportion of positivity for both semen and smegma was 33.3%, but 8.3% was positive only for semen, and 16.7% only for smegma, revealing a moderate agreement between tests (K=0.5; Kappa index). The genome of one equine embryo and three equine semen specimens collected from a Brazilian farm were tested by polymerase chain reaction (PCR) for the presence of EHV-1 and EHV-4-specific timidine kinase (TK) sequences. The PCR detected specific EHV-1 TK gene sequences in all samples tested. The peripheral blood leukocytes (PBL) of the embryo donor mare also was amplified by EHV-1 TK primers. Infectious virus was not recovered from any specimens. The animals did not show any clinical signal of EHV-1 or EHV-4 infections. EHV-4 was not detected in the studied specimens. The results indicate that PCR was more sensitive than virus isolation in cell culture for detecting EHV-1 in semen of carrier horses.

The effectiveness of induction of the acrosome reaction (AR) test as a parameter to in vitro estimate embryo production (IVP) in Nelore breed and the AR pattern by the Trypan Blue/Giemsa (TB) stain were evaluated. Frozen semen samples from ten Nelore bulls were submitted to AR induction and were also evaluated for cleavage and blastocyst rates. The treatments utilized for AR induction were: control (TALP medium), TH (TALP medium + 10μg heparin), TL (TALP medium + 100μg lysophosphatidylcholine) and THL (TALP medium + 10μg heparin + 100μg lysophosphatidylcholine).
