A mezőgazdasági területek érzékenységének és adaptációs képességének mérési lehetőségei AgEcon
Farkas-Fekete, Maria.

Ma,Min Min; Jiang,Dan; Li,Shuang Ya; Wu,Yuan; Meng,Ju; Huang,Jiang Yao.
ABSTRACT Adulterant herbal materials are threats to import and export trade and consumer safety. In this study, we established a simple and rapid examination system for the detection of Phellodendron chinense Schneid. Two detection methods, real-time fluorescence quantitative PCR (real-time PCR) and loop-mediated isothermal amplification (LAMP), were developed for traditional Chinese medicine detection, and their specificity and sensitivity were compared. The DNA of P. chinense was extracted and its special periods amplified with designed primers. Real-time PCR and LAMP experiments were conducted to test the specificity of primers in contrast to other similar species. The template concentration was diluted from 101 ng/µL to 10-5 ng/µL in order to contrast...

ABSTRACT Introduction Latent tuberculosis infection diagnosis based on the release of interferon-gamma in cultures of peripheral blood cells stimulated with Mycobacterium tuberculosis antigens has replaced the tuberculin skin test in many countries with low tuberculosis prevalence. The IFN-γ production can be influenced by genetic polymorphisms, of which the IFNG + 874 (rs62559044) locus is the most studied. We investigated the possible influence of the IFNG + 874 A/T polymorphism on interferon-gamma test performance. Methods Patients diagnosed with pulmonary tuberculosis (75), volunteers with positive tuberculin skin test (70) and healthy volunteers with negative tuberculin skin test and no history of contact with tuberculosis (57) were evaluated...

Lectins are carbohydrate-binding proteins of non-imune origin. This group of proteins is distributed widely in nature and they have been found in viruses, microorganisms, plants and animals. Lectins of plants have been isolated and characterized according to their chemical, physical-chemical, structural and biological properties. Among their biological activities, we can stress its fungicidal action. It has been previously described the effect of the lectins Dviol, DRL, ConBr and LSL obtained from the seeds of leguminous plants on the growth of yeasts isolated from vaginal secretions. In the present work the experiments were carried out in microtiter plates and the results interpreted by both methods: visual observations and a microplate reader at 530nm....

Variation of water potential and trunk diameter answer as sensitivity to the water availability in table grapes Chilean J. Agric. Res.
Silva-Contreras,Claudio; Sellés-Von Schouwen,Gabriel; Ferreyra-Espada,Raul; Silva-Robledo,Herman.
Variation in trunk diameter (TDV) has been proposed as an indicator of the water status of grape vines, with apparently contradictory results. In Vitis vinifera L. var. Crimson Seedless we evaluated the water potential (Ψ), an indicator normally used to determine the water status of grapes, and TDV, comparing two irrigation treatments which restored 100% or 50% of the ETc, to compare the sensitivity of these two indicators to a decrease in soil water. Two evaluation periods of 23 d each were used, the first during the exponential growth phase and the second in the post-veraison period, when the trunk stops growing. In both periods TDV showed coefficients of variation greater than 25%, compared to the 7-10% recorded for water potential. However,...

Evaluation of the performance of different plastics used to seal nylon cDNA arrays Ciência e Agrotecnologia
Costa Netto,Antônio Paulino da; Drummond,Rodrigo Duarte; Felix,Juliana de Maria; Jorge,Renato Atílio; Menossi,Marcelo.
cDNA arrays are a powerful tool for discovering gene expression patterns. Nylon arrays have the advantage that they can be re-used several times. A key issue in high throughput gene expression analysis is sensitivity. In the case of nylon arrays, signal detection can be affected by the plastic bags used to keep membranes humid. In this study, we evaluated the effect of five types of plastics on the radioactive transmittance, number of genes with a signal above the background, and data variability. A polyethylene plastic bag 69 μm thick had a strong shielding effect that blocked 68.7% of the radioactive signal. The shielding effect on transmittance decreased the number of detected genes and increased the data variability. Other plastics which were thinner...
