Induction of somatic embryogenesis and plant regeneration of sugarcane cv. RB925345 from somatic embryos Sci. Agrar.Paran. / SAP

The aim of this study was to establish a protocol for induction of somatic embryogenesis and regeneration of somatic embryos of sugarcane (cultivar RB925345). Best results was obtained using the concentration of 13.5 µM L-1 2,4-dichlorophenoxyacetic acid, 45 days of incubation in induction medium, one subculture and 15 days of incubation in regeneration medium, providing 47.5% of embryogenic calli and 38.9% of plants regenerated. Adjust are necessary in this protocol to increase the rate of embryogenic calli production and regeneration of somatic embryos.

ABSTRACT: Plants of Coffea arabica Icatu IAC 925 germplasm show resistance to Meloidogyne paranaensis under field conditions, but with variable rates of segregation. One mother plant, IAC 3, of Icatu IAC 925 was evaluated under controlled conditions through the reaction of its clones, obtained by somatic embryogenesis. Reproduction of nematodes in clones were compared with an open-pollinated progeny from the same mother plant, C. arabica cultivar Mundo Novo IAC 515-20, susceptible to nematodes, and Clone IAC 1, also susceptible and obtained by somatic embryogenesis. Regeneration of soybean via embryogenic suspension culture Scientia Agricola

In an attempt to establish an alternative plant regeneration system for soybean [Glycine max (L.) Merrill] cultivars used in Brazilian breeding programs, ten genotypes were tested for their embryogenic potential. Cotyledons were removed as explants from immature seeds harvested from field-grown plants. After 45 days on induction medium, the number of responding cotyledons and the number of somatic embryos per immature cotyledon were evaluated. The percentage of explants that produced somatic embryos varied from 1 to 70% among cultivars. The first step in plant transformation using somatic embryogenesis or organogenesis for plant regeneration in vitro has been the identification of genotypes with the best totipotency. Thus, the objective of this experiment was to evaluate the totipotency of ten cultivars and one soybean line from the Coodetec Gene Bank. The somatic embryos developed from immature cotyledons measuring from 4 to 6 mm in length, and the organogenesis was evaluated from mature soybean seeds. Induction of cassava somatic embryogenesis in liquid medium associated to floating membrane rafts BABT

The objective of this study was to examine the effect of two culture systems, liquid medium associated to floating membranes and solid medium, both supplemented with different concentrations of 2,4-D, in the induction of somatic embryogenesis of cassava (Manihot esculenta Crantz). Only 28% of the young leaf lobes (with 9 µM 2,4-D) were induced to form organized embryogenic structures (OES) with membrane rafts, compared to 50% of the explants presenting this type of tissue in solid medium with 36 µM of 2,4-D. Biochemical and morphological changes during the growth kinetics of Araucaria angustifolia suspension cultures BABT

Embryogenic cultures of Araucaria angustifolia were established in a BM liquid medium supplemented with 2 µM 2,4-dichlorophenoxyacetic acid, 1 µM 6-benzylaminopurine and 1 µM kinetin (BM2) and in a BM medium free of growth regulators (BM0). During 42 days in culture, the cell growth pattern of both cultures was similar. The pH of the culture medium of both BM0 and BM2 underwent progressive reduction during culture time. A method for the induction of somatic embryogenesis in eight cassava genotypes from northeastern Brazil is described. The explants used were shoot apexes isolated both from in vitro grown plants and from shoots that sprouted from stem cuttings. Somatic embryogenesis was achieved in high frequencies by the addition in the induction medium of the auxin picloram over a wide range of concentrations. Effect of carbon source on morphology and histodifferentiation of Araucaria angustifolia embryogenic cultures BABT

The aim of the present work was to establish in vitro conditions for the induction, stabilization and proliferation of embryogenic cultures of A. angustifolia. Pre-cotyledonary staged zygotic embryos inoculated BM medium supplemented with 5 µM 2,4- D, 2 µM BAP and Kin, and 3% maltose or sucrose resulted in 66.7% induction rate. The rate of induction of embryogenic cultures was affected by the carbon source, as well the multiplication and morphology of the embryogenic cultures. In vitro techniques have been applied not only for coffee improvement through genetic transformation but also to study various aspects in coffee cells such as chemical (caffeine synthesis and the production of coffee aroma), physiological and more recently, biochemical aspects. The most important advances obtained to date on in vitro coffee techniques in fields like biochemistry, physiology, regeneration systems and genetic engineering, are presented and discussed.
