The Ameriorating Effects of Grapefruit Seed Extract on Testicular Morphology and Function of Varicocelized Rats International Journal of Morphology
Saalu,,L. C; Udeh,R.; Oluyemi,K. A; Jewo,P. I; Fadeyibi,L. O.
Varicoceles are abnormal tortuosity and dilatation of the veins of the pampiniform plexus that drain the testis. The pathophysiology of testicular damage in varicocele has not been completely understood. However there is an increasing body of evidence pointing towards the role of reactive oxygen species and oxidative stress in the pathogenesis of varicocele related subfertility. In the present study we examined the role of proanthocynidin-rich ethanol extract of grapefruit seed as an antioxidant in protecting the testis against damage in experimental varicocele. Three groups of rats were constructed as the first group had intact rats; experimental bilateral varicoceles were established by complete ligation of the left and right main spermatic veins in the...

Protective Effects of Melatonin on Testicular Torsion and Detorsion Damage in Sprague-Dawley Rats International Journal of Morphology
Aktas,Ayfer; Tuncer,M. Cudi; Yildirim,Ayse; Nergiz,Yusuf; Akkus,Murat.
In this study, we evaluated the ultrastructural findings of testis with systemic administration of different doses of melatonin during ischemic period in a rat model of testicular torsion/detorsion (T/D). Testis ischemia-reperfusion (I/R) injury was induced by torsion of the left testis, with a 720 degrees twisting of the spermatic cord so as to produce a total occlusion of testis for 2.5 hours. Subsequently, the same testis was then detorsioned. According to surgical procedure in each group, unilateral orchiectomies were performed for histopathologic examination. The groups were labelled as control group, torsion group (T), torsion and detorsion group (T/D), torsion-detorsion and melatonin group (T/D+20,50 and 100 mg/kg melatonin). For the histological...

Toxicological studies of Momordica charantia Linn Seed extracts in Male Mice International Journal of Morphology
Patil,Sharanabasappa A; Patil,Saraswati B.
An attempt to find out the male contraceptive molecule of plant origin, the extracts of seeds of Momordica charantia were tested in male mice. Petroleum ether, chloroform and ethanolic extracts of Momordica charantia were administered at the dose level of 25mg/100gm body weight to the albino mice for 48 days intraperitoneally. All the extracts showed antispermatogenic effect as the number of spermatogonia, spermatocytes, spermatids and spermatozoa were decreased. The increase in the weight of epididymis, prostate gland, seminal vesicle and vas deferens indicates clearly the androgenic property of these extracts. After subjecting to preliminary phytochemical screening ethanol extract showed positive tests for alkaloids, flavonoids, glycosides, phenols,...

Histopathological changes in albino mouse testis induced by the administration of Tityus n. sp. venom J. Venom. Anim. Toxins
PENNA-VIDEAÚ,S.; CERMEÑO-VIVAS,J.; MORENO-MARVAL,M.; QUIROGA-NOTTI,M..
Scorpion envenoming is a public health concern in northeastern Venezuela. Specimens of the genus Tityus are responsible for most of these. In experimental animals, Tityus venom produces histopathological changes in the skeletal muscle and pancreas, but its toxicity to the reproductive system has not been studied. The aim of this work is to describe the histopathological changes in testis and epididymis of albino mice induced by the administration of Tityus n. sp. venom. Sub-lethal doses of venom (3.75 mg/g mouse) were administered intramuscularly (IM) daily for 4 days. On the fifth day, the animals were sacrificed and the testes and epididymes were quickly removed and processed for light microscopy. The venom induced alterations in spermatogenesis. Sertoli...

A bradykinin-potentiating peptide (BPP-10c) from Bothrops jararaca induces changes in seminiferous tubules J. Venom. Anim. Toxins incl. Trop. Dis.
Gilio,Joyce M; Portaro,Fernanda CV; Borella,Maria I; Lameu,Claudiana; Camargo,Antonio CM; Alberto-Silva,Carlos.
Background The testis-specific isoform of angiotensin-converting enzyme (tACE) is exclusively expressed in germ cells during spermatogenesis. Although the exact role of tACE in male fertility is unknown, it clearly plays a critical function in spermatogenesis. The dipeptidase domain of tACE is identical to the C-terminal catalytic domain of somatic ACE (sACE). Bradykinin potentiating peptides (BPPs) from snake venoms are the first natural sACE inhibitors described and their structure-activity relationship studies were the basis for the development of antihypertensive drugs such as captopril. In recent years, it has been showed that a number of BPPs - including BPP-10c - are able to distinguish between the N- and C-active sites of sACE, what is not...

Anti-Müllerian hormone as an indicator of hemi-castrated unilateral cryptorchid horses OAK
MURASE, Harutaka; SAITO, Shigeaki; AMAYA, Tomohiko; SATO, Fumio; BALL, Barry Allen; NAMBO, Yasuo.
Anti-Müllerian hormone (AMH), a glycoprotein secreted from the fetal testis, is responsible for regression of the Müllerian duct in the male fetus. The aim of this study was to evaluate the usefulness of serum AMH as a biomarker for diagnosis of cryptorchidism in horses. Serum AMH concentrations were measured in intact stallions, hemi-castrated unilateral cryptorchid stallions, and geldings. In addition, expression of AMH was characterized in cryptorchid testes by immunohistochemistry. Serum AMH was detected in intact stallions (n=11, 13.3 ± 1.8 ng/ml) and in hemi-castrated cryptorchid stallions (n=8, 17.6 ± 3.0 ng/ml), but not in geldings (n=6, all data were below the limit of detection). Immunolabeling for AMH was detected in Sertoli cells of undescended...

ABSTRACT Cottonseed coproducts contain gossypol which can have specific effects upon the male reproductive system. We evaluated the reproductive development of male lambs close to puberty fed on three cotton coproducts. Twenty-four 5-month old male lambs received four diets: 20 % of dry matter intake of whole cottonseed (WCS), high oil cottonseed meal (CSC), cottonseed meal (CSM), and a control group (CTL) without any cottonseed coproducts. Free gossypol intake was 16.32, 6.98, 5.47 and 0 mg kg−1 BW−1 d−1 for WCS, CSC, CSM and CTL, respectively. Every 15 days, the animals were weighted and serum and semen samples were collected. After 95 days, testis samples were collected for analysis under light and transmission electron microscopes. The CTL group had...

Morphological study of the testes of the dove Columba livia (Gmelin) (Columbidae, Columbiformes) Zoologia
Maruch,Sandra Maria das Graças; Teles,Maria Eloiza de Oliveira; Ribeiro,Maria das Graças.
Known as "domestic dove", the Columba livia (Gmelin, 1789) is a columbidae species widely distributed in Brazil, whose reproductive biology has been studied by many researchers. The testes of 12 Columba livia males were collected and prepared for histologic examination under an optical microscope, the results of which were analysed and photographed. The tunica albuginea that covers the testes consists of a thick, not very cellular layer of dense connective tissue. Groups of interstitial cells with typical morphological appearance and surrounded by loose, well vascularized connective tissue are observable within the organ, between the seminiferous tubules. The seminiferous tubules are thick, intensely wound and, when seen in cross section, show Sertoli...

HISTOLOGICAL DESCRIPTION OF THE REPRODUCTIVE SYSTEM OF MALE AND FEMALE HATCHLINGS OF THE MAGDALENA RIVER TURTLE (Podocnemis lewyana) Acta biol.Colomb.
SÁNCHEZ-OSPINA,ANDRÉS C; RODRIGUEZ,BERARDO; CEBALLOS,CLAUDIA P..
In this study we described the macroscopic and microscopic histology of the reproductive system of male and female Podocnemis lewyana neonates (3.5 months old). We found macroscopic differences in the morphology of the gonads between the sexes, with ovaries being twice as long as testes, and testes being twice as wide as ovaries. Microscopically, we identified several immature elements, such as the lack of a muscle layer in the oviduct of females, and simple epithelia instead of pseudostratified epithelia in the oviduct and epididymis described in reptile adults. We also found a black pigment observed macroscopically in the mesovarium, and macroscopically and histologically in the epididymis. This pigment is consistent with the center of melano-macrophages...

Expression of caveolin-1 in rat Leydig cells Biocell
Casanova,Marta B..
Caveolin-1, the first member of caveolin family reported, is recognized as the structural component of caveola, a plasma membrane invagination or vesicles that are a subcompartment distinct from clathrin-coated pits. This protein is also known to be involved in cholesterol trafficking. The aim of this study was to determine the expression of caveolin-1 in adult rat Leydig cells. Testis sections incubated with an antibody to caveolin-1 showed, by immunohistochemistry, a moderate number of Leydig cells with different degrees of immunoreaction and a strong reaction in endothelial cells and in the lamina propia of seminiferous tubules. Caveolin-1 was detected in the cell cytoplasm with a granular pattern and on the cell surface of Leydig cells cultured 24 h on...

Tissue mRNA expression in rat of newly described matrix metalloproteinases Biol. Res.
BERNAL,FABIÁN; HARTUNG,HANS-PETER; KIESEIER,BERND C.
Matrix metalloproteinases (MMPs) comprise a large group of endoproteinases that degrade all protein components of the extracellular matrix. Functionally, MMPs contribute to several different physiological as well as pathological conditions. The number of newly described MMPs has increased in recent years, although current knowledge about their expression pattern in various tissues remains incomplete. Here we analyzed the relative mRNA expression of the most recently described MMPs _ MT5-MMP (MMP-24), MT6-MMP (MMP-25), MMP-27 and epilysin (MMP-28) _ in a broad selection of rat tissues using real time-PCR. MMP-24 mRNA was found to be widely expressed with predominance in the central nervous system. MMP-25 mRNA, in contrast, exhibited peak expression levels...

Abstract Background The muskrat is a seasonal breeder. Males secrete musk to attract females during the breeding season. The testosterone binding to the androgen receptor (AR) in musk glands of muskrat may play an important role conducting the musk secretion process. Methods The musk gland, testis and blood samples of musk rats are collected in both breeding and non-breeding seasons. Some part of the samples are kept in liquid nitrogen for transcriptome analysis and Western blotting test. Some part of the samples are kept in 70% alcohol for histology experiment, blood samples are kept at −20 °C for the serum testosterone measurement experiment. Results This study demonstrates that the quantity of secreted musk, the volume of the musk...

Differential expression and localization of ADAM10 and ADAM17 during rat spermatogenesis suggest a role in germ cell differentiation Biol. Res.
Urriola-Muñoz,Paulina; Lizama,Carlos; Lagos-Cabré,Raúl; Reyes,Juan G; Moreno,Ricardo D.
BACKGROUND: Extracellular metolloproteases have been implied in different process such as cell death, differentiation and migration. Membrane-bound metalloproteases of the ADAM family shed the extracellular domain of many cytokines and receptor controlling auto and para/juxtacrine cell signaling in different tissues. ADAM17 and ADAM10 are two members of this family surface metalloproteases involved in germ cell apoptosis during the first wave of spermatogenesis in the rat, but they have other signaling functions in somatic tissues. RESULTS: In an attempt to further study these two enzymes, we describe the presence and localization in adult male rats. Results showed that both enzymes are detected in germ and Sertoli cells during all the stages of...

Contribution of environmental pollutants to male infertily: A working model of germ cell apoptosis induced by plasticizers Biol. Res.
Lagos-Cabré,Raúl; Moreno,Ricardo D.
Bisphenol A [2,2-bis(4-hydroxyphenyl)propane] (BPA), 4-nonylphenol (NP) and di(2-ethylhexyl)phthalate (DEHP), and its metabolite mono-2-ethylhexyl phthalate (MEHP) are chemicals found in plastics, which act as endocrine disruptors (EDs) in animals, including human. EDs act like hormones in the endocrine system, and disrupt the physiologic function of endogenous hormones. Most people are exposed to different endocrine disruptors and concern has been raised about their true effect on reproductive organs. In the testis, they seem to preferentially attack developing testis during puberty rather than adult organs. However, the lack of information about the molecular mechanism, and the apparently controversial effect observed in different models has hampered the...

Human testis-expressed sequence 101 is limitedly distributed in germinal epithelium of testis and disappears in seminoma Biol. Res.
Shen,Cong-Cong; Kang,Yu-Huan; Yu,Lin; Cui,Dan-Dan; He,Yi; Yang,Jin-Liang; Gou,Lan-Tu.
BACKGROUND: Testis-expressed sequence 101 (TEX101) was found to be highly expressed in testis and involved in acrosome reaction in previous studies. Recently, the metastasis suppressor function of TEX101 in cancer was disclosed, but the comprehensive investigation of its expression has rarely been reported. In this study, the expression features of TEX101 in normal human organs and seminoma were systematically analyzed. RESULTS: Immunohistochemistry demonstrated intense staining of TEX101 in human testis tissues; however, its expression in 27 other types of normal human organs, including the ovary, was negligible. Higher expression of TEX101 was observed in the spermatocytes and spermatids of the testis, but relatively lower staining was detected in...
