Multiple pathways to sustainability in the city: the case of San Juan, Puerto Rico Ecology and Society
I examined the multiple visions of the future of the city that can emerge when city actors and organizations reconfigure themselves to address sustainability. In various cities worldwide, novel ideas, initiatives, and networks are emerging in governance to address social and ecological conditions in urban areas. However, cities can be contested spaces, bringing a plurality of actors, network configurations, preferences, and knowledge that shape the politics over desirable pathways for future development. I used the knowledge-action systems analysis (KASA) approach to examine the frames and knowledge systems influencing how different actors involved in the land governance network of the city of San Juan constructed visions for the future of the city....

Plans to solve complex environmental problems should always consider the role of surprise. Nevertheless, there is a tendency to emphasize known computable aspects of a problem while neglecting aspects that are unknown and failing to ask questions about them. The tendency to ignore the noncomputable can be countered by considering a wide range of perspectives, encouraging transparency with regard to conflicting viewpoints, stimulating a diversity of models, and managing for the emergence of new syntheses that reorganize fragmentary knowledge.

Emergent transformation games: exploring social innovation agency and activation through the case of the Belgian electricity blackout threat Ecology and Society
The persistence of current societal problems has given rise to a quest for transformative social innovations. As social innovation actors seek to become change makers, it has been suggested that they need to play into impactful macrodevelopments or “game-changers”. Here, we aim to deepen the understanding of the social innovation agency in these transformation games. We analyze assumptions about the game metaphor, invoking insights from actor-network theory. The very emergence of transformation games is identified as a crucial but easily overlooked issue. Climate change and its interactions with complex socioeconomic dynamics dictate the need for decision makers to move from incremental adaptation toward transformation as societies try to cope with unprecedented and uncertain change. Developing pathways toward transformation is especially difficult in regions with multiple contested resource uses and rights, with diverse decision makers and rules, and where high uncertainty is generated by differences in stakeholders’ values, understanding of climate change, and ways of adapting. In the present study, a 419 bp upstream sequence of the phycocyanin b subunit (cpcB) gene from Arthrospira platensis FACHB341 was fused with green fluorescent protein (gfp) gene, and a heterologous reporting system was built up to investigate the influence of light intensity on the expression of gfp gene, and the regulation function of different region of the upstream sequence of cpcB gene. An improved system for competent cell preparation and high efficiency plasmid transformation using different Escherichia coli strains Electron. J. Biotechnol.

This paper describes an efficient bacterial transformation system that was established for the preparation of competent cells, plasmid preparation, and for the storage in bacterial stocks in our laboratory. Using this method, a number of different plasmids have been amplified for further experiments. Competent cells for bacterial transformation were prepared by the calcium chloride method with an optimum concentration of 75 mM. Three different strains of Escherichia coli that were tested are DH5α, TG1 and XL1 blue, and the most efficient strain being XL1 blue. In the present study, a novel plant transformation system for Doritaenopsis and Phalaenopsis has been developed. The pollen-mediated activation tagging system was established by artificial pollination. The pollens, co-cultured with Agrobacterium tumefaciens strain EHA105 harbouring an activation tagging vector (pTAG-8), were used for pollination. In order to optimize the transformation efficiency, several factors (concentration of A. tumefaciens, concentration of acetosyringone during co-cultivation and the duration of co-cultivation) known to influence Agrobacterium-mediated DNA transfer were examined. Ethanol mediated enhancement in bacterial transformation Electron. J. Biotechnol.

In molecular biology, transformation using E. coli as a host plays a key role in synthesizing gene libraries. The present study demonstrated a new ethanol-based method for transformation of plasmid DNA to E. coli. Ethanol at 10% concentration (v/v) showed best results. Further, as compared with traditional CaCl2 method, the transformation rate, using protocol outlined in this study, was very high, suggesting amenable for further applications.

Brugmansia candida (Solanaceae) is a native tree distributed across South-American and produces the pharmacologically- important group of tropane alkaloids including scopolamine. This biocompound is synthesised from hyoscyamine by action of Hyoscyamine 6-β hydroxylase (H6H, EC 1.14.11.11) at the end of the tropane alkaloid pathway. Here are reported the tissue and organ-specific expression of h6hmRNA by RT-PCR analyses and the isolation, cloning and sequencing of the cDNA obtained from B. candida anthers and hairy root transformed cultures. The genetically transformed roots of red beet have been shown, for the first time, to produce very high levels of peroxidase (POD; EC 1.11.1.7) accounting for 1.21 x 10(6) Units L-1. Of the ten clones established using different strains of Agrobacterium rhizogenes, one was that from the strain LMG-150, three each from A 2/83, A 20/83 and A4. All the clones showed true integration of T-DNA when tested by PCR and Southern hybridization methods. The results presented in this work support the hypothesis that Agrobacterium-mediated transformation of sorghum is feasible, analogous to what has been demonstrated for other cereals such as rice, maize, barley and wheat. The four factors that we found most influenced transformation were: the sensitivity of immature sorghum embryos to Agrobacterium infection, the growth conditions of the donor plant, type of explant and co-cultivation medium. A major problem during the development of our protocol was a necrotic response which developed in explants after co-cultivation. Abstract We present a collection of minimalist binary vectors for transformation through ATMT applicable to several fungi species. pLUO plasmid binary vectors consist of a reporter module containing fluorescent proteins, mCherry or eGFP, flanked by a multiple cloning site and a transcription terminator site. Beyond the technical and technological advances in the conversion of marine by-products into useful products, the territorial, economic, social, political and environmental parameters, which govern all forms of approaches for fish by-products upgrading, must be taken in account. This is essential to understand why and how bioconversion technologies are applicable. The fishery sector is facing several issues such as fish stocks weakening, recourse to imports, aquaculture rising or consumers behaviors which complicate the approach of fish by-products upgrading as it is linked to the raw material processing. Critical steps of the process of horse mackerel surimi manufacturing were identified. This led to propose technical and chemical solutions in order to limit development of lipid oxidation reactions. In this way, analytical methods of lipids and lipid oxidation compounds were adapted to the studied products. A method to determine hydroperoxyde concentrations was developed. The front face fluorescence technique was tested to evaluate the degree of surimi alteration. Lipid oxidation kinetics were determined during horse mackerel storage on ice or at 17°C.
