The principle of this method is to evaluate, by counts exercised upon selective media, the relative importance of the different bacteria families in the flora of the coastal waters. It is important to separate the contaminant species and the commensal species in the marine environment. Ten culture media distributed into four categories (selective, non selective, saline and non saline) were examined by numeration. Production and consumption of B group vitamins in situ. ArchiMer

The changes in concentration of thiamine, biotin and vitamin B sub(12) in water of the soaked bottles for 6 h in situ in the north basin of Lake Biwa were examined successively over 30 hours. The concentrations of the vitamins changed comparatively during 6 h. soaking in situ. Generally, the concentrations of thiamine, biotin and vitamin B sub(12) were in high levels in the morning and in low levels in evening both in the soaked bottles and in the surrounding water. The fluctuation ranges in concentration of vitamins in a 5 m deep layer were greater than those in thermocline. Culture-Facilitated Comparative Genomics of the Facultative Symbiont Hamiltonella defensa ArchiMer

Many insects host facultative, bacterial symbionts that confer conditional fitness benefits to their hosts. Hamiltonella defensa is a common facultative symbiont of aphids that provides protection against parasitoid wasps. Protection levels vary among strains of H. defensa that are also differentially infected by bacteriophages named APSEs. However, little is known about trait variation among strains because only one isolate has been fully sequenced. Generating complete genomes for facultative symbionts is hindered by relatively large genome sizes but low abundances in hosts like aphids that are very small. Microbial extracellular enzyme detection on agar plates by means of fluorogenic methylum-belliferyl-substrates. ArchiMer

A rapid and sensitive method to detect the extracellular enzymatic activity of bacteria colonies grown on agar plates is described. Selective agar media supplemented with protein, starch, chitin, Tween-80, etc. are conventionally used to detect biochemical properties of bacteria. It has been experimentally demonstrated with bacteria pure cultures that fluorogenic Methylumbelliferyl (MUF)-substrates are excellent substrate analogues for normally occurring polymers. Based on MUF-substrate hydrolysis the new method provides reliable qualitative estimates of extracellular enzymatic properties of bacteria within minutes using pure cultures as well as agar plates prepared for colony counts.

During the austral summer, we examined the bacterial population along the cruise track extending from 70degrees S and 18degrees E to 30degrees S and 35degrees E. During the cruise, three distinct fractions of the bacterioplankton viz. total count, total direct viable and retrievable counts were simultaneously enumerated in these waters. In the stations south of the convergent region designated as section I the population ranged within 10(8-9) L-1 whereas in the north of the region designated as section 11 they were one order higher and ranged from 10(9-10) L-1. Rapid method for determining the sensitivity of sulphate-reducing bacteria to biocides. ArchiMer

The Minimal Inhibitory Concentration (MIC) of a biocide is usually determined by adding it in various concentrations to a suitable growth medium which is then inoculated with the test bacterium. These cultures are then observed for up to 28 days for growth of the organism as shown by its production of black ferrous sulphide. A method has been developed whereby the MIC of a biocide can be found within 4 days. Instead of noting ferrous sulphide production, this new method measures changes in: 1) optical density and 2) sulphate reduction of a logarithmic culture of the test strain exposed to the biocide.

Estimates of carbon flow through bacteriplankton in the S. Benguela upwelling region based on 3H-thymidine incorporation and predator-free incubations. ArchiMer

Spatial and temporal estimates of bacterial numbers, biomass, activity and production were measured in the S. Benguela upwelling system during the course of a phytoplankton bloom. Bacterial numbers and biomass were highest in the euphotic zone and correlated closely with particulate carbon in the water column rather than with chlorophyll a concentrations. 3H-Thymidine incorporation gave good estimates of production. Differential utilisation of photosynthetic exudates and detrital POC by bacterioplankton during the course of a bloom might account for the varying significance ascribed to exudates and POC as a source of carbon for bacterial production.

Due to technical difficulties, numbers of studies dealing with benthic bacterivory are not numerous and give conflicting results. A method using community of 15N enriched bacteria as tracer was developed in order to measure grazing of meiofauna and macrofauna. The present study concerns bacterial transit in the digestive tract of rainbow trout at 13 degree C and compares fecal excretion of 4 tracers in the alimentary bolus, an inert bacterial tracer: spores of Bacillus stearothermophilus , a bacterial marker capable of multiplying in the digestive organs: Citrobacter freundii ; a tracer of the liquid phase: polyethylene glycol (PEG), a tracer of the solid phase: Cr sub(2)O sub(3). Ballast water research in France : Current status ArchiMer

Due to important economic activities along the French oastline, the introduction of noxious organisms into French coastal waters, particularly by ship ballast water, may have serious consequences. After preliminary studies of ship movements in Charente Maritime, several samplings of ballast water and sediment were made. Samples were taken in five major harbours in France, along the Atlantic, Channel and Mediterranean coastlines. The main task of this first assessment was the research for exotic and possibly noxious phytoplancton. Clostridium and Vibrio bacteria were also tested for. The results are presented and discussed, with plans for further work.

Subseafloor archaeal communities: from the surface to a deep hot biosphere? ArchiMer

The sub-seafloor biosphere may contain two thirds of Earth's total prokaryotic biomass. The large and active microbial populations buried in the sub-seafloor play a key role in global biogeochemical cycles. However, little is known about these prokaryotic communities. The depth limit of this sub-seafloor biosphere is still unreached, and elevated temperatures as well as insufficient energy sources are the likely factors limiting life at depth.
