Biofilm formation and acyl homoserine lactone production in Hafnia alvei isolated from raw milk Biol. Res.
SOUZA VIANA,ELISETH; MARTINO CAMPOS,MARIA EMILENE; REIS PONCE,ADRIANA; CUQUETTO MANTOVANI,HILÁRIO; DANTAS VANETTI,MARIA CRISTINA.
The objective of this study was to detect the presence of acyl homoserine lactones (AHLs), signal molecules of the quorum sensing system in biofilm formed by Hafnia alvei strains. It also evaluated the effect of synthetic quorum sensing inhibitors in biofilm formation. AHLs were assayed using well diffusion techniques, thin layer chromatography (TLC) and detection directly in biofilm with biomonitors. The extracts obtained from planktonic and sessile cell of H. alvei induced at least two of three monitor strains evaluated. The presence of AHLs with up to six carbon atoms was confirmed by TLC. Biofilm formation by H. alvei was inhibited by furanone, as demonstrated by 96-well assay of crystal violet in microtitre plates and by scanning electron microscopy....

Microbial succession within an anaerobic sequencing batch biofilm reactor (ASBBR) treating cane vinasse at 55ºC BABT
Ribas,Maria Magdalena Ferreira; Chinalia,Fabio Alexandre; Pozzi,Eloisa; Foresti,Eugenio.
The aim of this work was to investigate the anaerobic biomass formation capable of treating vinasse from the production of sugar cane alcohol, which was evolved within an anaerobic sequencing batch biofilm reactor (ASBBR) as immobilized biomass on cubes of polyurethane foam at the temperature of 55ºC. The reactor was inoculated with mesophilic granular sludge originally treating poultry slaughterhouse wastewater. The evolution of the biofilm in the polyurethane foam matrices was assessed during seven experimental phases which were thus characterized by the changes in the organic matter concentrations as COD (1.0 to 20.0 g/L). Biomass characterization proceeded with the examination of sludge samples under optical and scanning electron microscopy. The...

Resistance of oxidative stress in biofilm and planktonic cells BABT
Jakubowski,Witold; Walkowiak,Bogdan.
This work studied the susceptibility of biofilm produced by E. coli to oxidative stress, and compared the components of free radicals defences: level of glutathione, catalase and dismutase activities in planktonic and biofilm located cells. Results showed the diversity of responses to oxidative stress in bacterial cells in log or stationary phases in both planktonic and biofilm forms. The bacteria were exposed to free-radical donors (H2O2, tBOOH, menadione, SIN-1 or peroxynitrite) in a wide range of final concentrations, from 0.5 to 10mM. Different level of toxicity of individual donors, independence of cell type (planktonic forms or biofilm) and phases of growth were observed. The highest oxidative stress resistance was observed for the cells in...

Lima,Cláudio Antonio Andrade; Ribeiro,Rogers; Foresti,Eugenio; Zaiat,Marcelo.
This work focused on a morphological study of the microorganisms attached to polyurethane foam matrices in a horizontal-flow anaerobic immobilized biomass (HAIB) reactor treating domestic sewage. The experiments consisted of monitoring the biomass colonization process of foam matrices in terms of the amount of retained biomass and the morphological characteristics of the cells attached to the support during the start-up period. Non-fluorescent rods and cocci were found to predominate in the process of attachment to the polyurethane foam surface. From the 10th week of operation onwards, an increase was observed in the morphological diversity, mainly due to rods, cocci, and Methanosaeta-like archaeal cells. Hydrodynamic problems, such as bed clogging and...

Influence of iron deprivation on virulence traits of mycobacteria BJID
Pal,Rahul; Hameed,Saif; Sharma,Sharda; Fatima,Zeeshan.
ABSTRACT Novel strategies to combat the ever increasing burden of drug resistance in Mycobacterium tuberculosis (MTB) causing tuberculosis (TB) remains a global concern. The ability of MTB to sense and adapt to restricted iron conditions in the hostile environment is essential for their survival and confers the basis of their success as dreadful pathogen. The striking and clinically relevant virulence trait of MTB is its ability to form biofilms and adhere to the host cells. The present study elucidated the effect of iron deprivation on biofilm formation and cell adherence of Mycobacterium smegmatis, a non-pathogenic surrogate of MTB. Firstly, we showed that iron deprivation leads to enhanced cell sedimentation rate and altered colony morphology depicting...

Clonal dissemination of vancomycin-resistant Enterococcus faecium ST412 in a Brazilian region BJID
Alves,Geraldo da Silva; Pereira,Monalessa Fábia; Bride,Lais de Lima; Nunes,Ana Paula Ferreira; Schuenck,Ricardo Pinto.
ABSTRACT Vancomycin-resistant Enterococcus faecium (VREfm) has emerged as an important global nosocomial pathogen, and this trend is associated with the spread of high-risk clones. Here, we determined the genetic and phenotypic features of 93 VREfm isolates that were obtained from patients in 13 hospitals in Vitória, Espírito Santo, Brazil, during 2012-2013. All the isolates were vancomycin-resistant and harbored the vanA gene. Only 6 (6.5%) of the VREfm isolates showed the ability to form biofilm. The 93 isolates analyzed belong to a single pulsed-field gel electrophoresis lineage and presented six subtypes. MLST genotyping showed that all VREfm belonged to ST412 (the high-risk clone, hospital-adapted). The present study describes the dissemination of...

Biofilm production using distinct media and antimicrobial susceptibility profile of Pseudomonas aeruginosa BJID
Perez,Leandro Reus Rodrigues; Barth,Afonso Luís.
Biofilm production is an important mechanism for bacterial survival and its occurrence together with antimicrobial resistance represents a challenge for clinical management. Here, we evaluated the ability for biofilm production among P. aeruginosa isolates from patients with or without cystic fibrosis (CF) using two distinct media, besides determining the antimicrobial susceptibility profile of these isolates for eight antimicrobial agents. The ability for biofilm production when TSB medium was used was higher than when used CF sputum media (p = 0.0198). However, P. aeruginosa isolates from CF have demonstrated similar performance for biofilm production, independently of the medium used. Besides, among the biofilm-producing isolates, those recovered from...
