A l'heure actuelle, l'objectif des éc10series d'huîtres est d'atteindre une autonomie d'approvisionnement en larves quelle que soit la saison et donc d'étaler les émissions de gamètes sur toute l'année. A la suite de problèmes rencontrés pour atteindre cet objectif, il s'est avéré nécessaire d'entreprendre des études sur les méthodes à mettre en oeuvre pour contrôler la qualité de la reproduction en écloserie et notamment l'état de maturité des géniteurs. Growth of the japanese oyster, Crassostrea gigas was monitored during 4 years, by following the recruitment of the year 1978, held in rearing conditions. After three and half years, the average length was la cm and the average total weight was 80 g. Clear seasonal changes were observed on the growth in length and weight. An important loss in dry weight occured in July, during the release of gametes. Such losses constituted 29 %, 43 % and 60 % of the dry weight, respectively for individuals which were 2 years, 3 years and 4 years old. Real-time PCR assay for rapid detection and quantification of Vibrio aestuarianus in oyster and seawater: A useful tool for epidemiologic studies ArchiMer

Because Vibrio aestuarianus is known to cause serious infections in Pacific oyster Crassostrea gigas, a real-time PCR assay was developed targeting the dnaJ gene of this bacterium. Only V. aestuarianus strains isolated from C. gigas mortality events in different geographic areas and the reference strain tested positive, whereas no amplification products was obtained with type strains belonging to 23 other species of Vibrio. Sensitivity and reproducibility of the method were assessed using either seawater or oyster homogenate samples spiked with one V aestuarianus strain. Effect of age and environment on the summer mortality in cupped oyster Crassostrea gigas during the first two years ArchiMer

Three successive generations were produced between 2001 and 2003 to assess to what extent genetic variability exists for this trait in juvenile oysters Crassostrea gigas. For each generation, two groups were selected for their high ("R" for resistant) and low ("S" for susceptible) survival. Significant differences of mortality were observed during the first year in Rivière d'Auray (RA) (Brittany-France). Relationships between oyster mortality patterns and environmental data from monitoring databases along the coasts of France ArchiMer

Oyster mortality was monitored at multiple sites along the French coastline (around 5 500 km) between 1993 and 2005. Mortality data for one- and two-year-old oysters were collected over 10–12 years in 39 oyster culture sites bordering 3 different "seas": the English Channel, Bay of Biscaye and Mediterranean. Combining these data with records from environmental monitoring databases, 11 of these sites had consistent chronological data sets including both environmental data and mortality records. Mortality in one-year-old oysters was clearly summer mortality (49% of their annual mortality) whereas mortality of two-year-olds occurred mostly in spring (51%). Les conditions météorologiques semblaient cette année à priori favorables à la reproduction des moules. Cependant, le maturation des gonades a été très lente. Ainsi, les premières pontes, de faible importance, ont eu lieu début Avril. Plusieurs émissions tardives sont apparues fin Mai et se sont poursuivies jusqu'au début du mois de Juillet. Elles n'ont pas été importantes et ont conduit à une faible quantité de larves au stade fixation. Un captage "moyen" a été observé début Mai. Herpesviruses associated with mortalities among Pacific oyster, Crassostrea gigas, in France-Comparative study ArchiMer

Sporadic mortalities were reported in june and july of 1992 and 1993 among batches of hatchery-reared larval Pacifie oyster, Crassastrea gigas, and at the beginning of July and in August of 1993 among five batches of 3-7 month old young spats, C. gigas. Observations with transmission electron microscope showed the presence of herpes-like virus particles in infected larvae and young spats. The viruses observed in diseased larvae and affected young spat oysters were very similar. Elevated temperature and crowding may increase susceptibility of oysters to these herpes-like virus infections.

Analyse historique de la production conchylicole du Bassin de Marennes-Oléron et essai d'estimation empirique du potentiel conchylicole ArchiMer

The analysis of the official statistics of commercialised production of oysters (.Crassostrea angutata and Crassostrea gigas ) is achieved from 1866 to 1984 in term of their numbers and weights for the bay of Marennes-Oleron. The number of sanitary labels, multiplied by the mean weight of a crate can also give out the commercialised production provided it is increased by the bias and decreased by the exterior production of the bay . Thus the production of Portuguese oysters is only 45 000 tons from 1948 to 1957 with a maximum of 52, 000 tons for respectively 70 000 tons and 280 000 tons for the statistics . AIso, after the epizooties of Crassostrea angulata the production, fell down to 25 000 tons in 1971 according to the number of sanitary labels...

In Vivo RNA Interference of a Gonad-Specific Transforming Growth Factor-beta in the Pacific Oyster Crassostrea gigas ArchiMer

We investigated the role of oyster gonadal TGF beta (og-TGF beta) in the reproduction of Crassostrea gigas, using an in vivo RNA interference approach. We designed double-stranded RNA targeting og-TGF beta, which is specifically expressed in the somatic cells surrounding germ cells in the gonad of both male and female oysters. In vivo injection of this og-TGF beta dsRNA into the gonad led to knock-down phenotypes for both sexes, with significant reduction (77.52% relative to controls) of the gonad area, lowered reproductive effort and germ cell under-proliferation. Starch supplementation modulates amylase enzymatic properties and amylase B mRNA level in the digestive gland of the Pacific oyster Crassostrea gigas ArchiMer

In the oyster Crassostrea gigas consumption-related traits, amylase properties and growth were found to be linked through genotypes that differed for polymorphism in the two amylase genes AMYA and AMYB. Modulation of AMYA mRNA level had already been observed in response to food availability, whereas the functional role of AMYB was still unknown. To improve knowledge about the regulation of amylase expression in C. gigas and the respective roles of the two genes, we made an assay of amylase expression at mRNA and enzymatic levels in the digestive gland of oysters that had received dietary supplements of starch. To gain information on oxidative phosphorylation (OXPHOS) functionality during the motility phase of Pacific oyster spermatozoa, we studied 1) changes in spermatozoal mitochondrial activity, that is, mitochondrial membrane potential (MMP), and intracellular ATP content in relation to motion parameters and 2) the involvement of OXPHOS for spermatozoal movement using carbonyl cyanide m-chlorophenyl hydrazone (CCCP).
