Very low mitochondrial variability in a stingless bee endemic to cerrado Genet. Mol. Biol.
Brito,Rute Magalhães; Francisco,Flávio de Oliveira; Françoso,Elaine; Santiago,Leandro Rodrigues; Arias,Maria Cristina.
Partamona mulata is a stingless bee species endemic to cerrado, a severely threatened phytogeographical domain. Clearing for pasture without proper soil treatment in the cerrado facilitates the proliferation of termite ground nests, which are the nesting sites for P. mulata. The genetic consequences of these changes in the cerrado environment for bee populations are still understudied. In this work, we analyzed the genetic diversity of 48 colonies of P. mulata collected throughout the species' distribution range by sequencing two mitochondrial genes, cytochrome oxidase I and cytochrome B. A very low polymorphism rate was observed when compared to another Partamona species from the Atlantic forest. Exclusive haplotypes were observed in two of the five areas...

Genetic divergence among eggplant genotypes under high temperatures Horticultura Brasileira
Valadares,Ricardo de N; Nóbrega,Danieli A; Lima,Lilian B de; Mendes,Adônis Q; Silva,Fabian S; Melo,Roberto de A; Menezes,Dimas.
ABSTRACT The aim of this study was to estimate the genetic divergence among eggplant genotypes for agronomic traits in order to gather information for the selection of genotypes in eggplant breeding programs for tolerance to high temperatures. Ten traits recommended by the International Board for Plant Genetic Resources were analyzed in 24 genotypes, arranged in a randomized complete block design with four replicates. Data were submitted to analysis of variance (P<0.01) and later to the UPGMA and Tocher grouping methods, using the generalized Mahalanobis distance (D2) as dissimilarity measure. Three and six groups of similarity were obtained, respectively, for the multivariate techniques used, UPGMA and Tocher, with concordance in the grouping of 87.50%...

Biochemical descriptors: importance of the genetic divergence study in peppers Horticultura Brasileira
Araújo,Lourismar M; Neves,Leonarda G; Sousa,Demétrio A; Zeviani,Walmes M; Silva,Lucinéia da R; Marostega,Thalita N.
ABSTRACT The genetic diversity of pepper genotypes is usually evaluated based on morphological and molecular descriptors. However, studies using biochemical descriptors of peppers are still incipient. Thus, the objective of this study was identify biochemical descriptors of Capsicum spp. with antioxidant action, most responsive to the selection of superior genotypes, and evaluate the correlation between concentrations of the parameters using principal components analysis. The experiment was conducted in the city of Cáceres, southwest Mato Grosso, Brazil. Seventy-eight pepper accessions from the Active Germplasm Bank of UNEMAT were planted in randomized block design. The harvested fruits were immersed in liquid nitrogen for two minutes, and the following...

Multicategorical descriptors for creole genotypes of Amazon chicory (Eryngium foetidum) Horticultura Brasileira
Gomes,Rafaelle F; Gonçalves,Viviane P; Arruda,Renata da S; Santos,Lucas da S.
ABSTRACT The knowledge concerning genetic variability of creole genotypes by using morphological descriptors in species such as Amazon chicory is crucial to quantify local diversity and protect improved cultivars. Accordingly, this study aimed to propose multicategory leaf morphological descriptors for Amazon chicory species. To achieve that, an experiment was arranged in randomized block design with four repetitions. The treatments comprised eight creole genotypes of Amazon chicory, which were obtained from local producers’ collection. The proposed multi-categorical descriptors are based on limb shape, leaf apex shape, leaf base shape, leaf margin, bract consistency, and bract margin traits. Once the multicategorical clustered data were obtained, they...

Prediction of genetic gains through selection of sweet potato accessions Horticultura Brasileira
Vargas,Pablo F; Otoboni,Maria Eduarda F; Lopes,Beatriz G; Pavan,Bruno E.
ABSTRACT Due to the high genetic variability found in sweet potato and a low number of cultivars available on the market, there are opportunities for necessary improvements in crop breeding programs. The selection indexes are a favorable strategy to achieve higher yields through genetic gains obtained with the future population. Thus, the objective was to evaluate the selection gain of agronomic characters from sweet potato accessions for root production and dual-aptitude. 95 accessions and two commercial cultivars (Braslandia Branca and Brazlândia Roxa) were evaluated. A randomized block design with three replications of ten plants per plot was used. The index proposed by Mulamba & Mock was used to select superior individuals. The evaluated population...

Agronomic potential and selection of okra hybrids to obtain potential genitors Horticultura Brasileira
Maciel,Gabriel M; Finzi,Rafael R; Marra,Alexandre William C; Carvalho,Fábio J; Nogueira,Ana Paula O.
ABSTRACT Evaluation of pre-commercial hybrids in a germplasm bank is essential for determining its commercial potential or its utility as a potential genitor in a breeding program. The objective of this study was to determine genetic divergence and per se behavior of 47 pre-commercial hybrids from okra germplasm bank of the Universidade Federal de Uberlândia. Precocity index (%), number of fruits (fruits per plant), average fruit mass (g) and productivity (g per plant) were evaluated. Analysis of genetic divergence was performed by multivariate analysis using Mahalanobis distance with different clustering methods (UPGMA and canonical analysis). The performance of hybrids was compared by Scott-Knott (p= 0.05). A significant genetic variability among okra...

Analysis of phenotypes altered by temperature stress and hipermutability in Drosophila willistoni Iheringia, Sér. Zool.
D'Ávila,Marícia F.; Garcia,Rosane N.; Loreto,Elgion L. S.; Valente,Vera Lúcia da S..
Drosophila willistoni (Sturtevant, 1916) is a species of the willistoni group of Drosophila having wide distribution from the South of USA (Florida) and Mexico to the North of Argentina. It has been subject of many evolutionary studies within the group, due to its considerable ability to successfully occupy a wide range of environments and also because of its great genetic variability expressed by different markers. The D. willistoni 17A2 strain was collected in 1991 in the state of Rio Grande do Sul, Brazil (30°05'S, 51°39'W), and has been maintained since then at the Drosophila laboratory of UFRGS. Different to the other D. willistoni strains maintained in the laboratory, the 17A2 strain spontaneously produced mutant males white-like (white eyes) and...

Effect of age and environment on the summer mortality in cupped oyster Crassostrea gigas during the first two years ArchiMer
Degremont, Lionel; Boudry, Pierre; Soletchnik, Patrick; Bedier, Edouard; Ropert, Michel; Samain, Jean-francois.
Three successive generations were produced between 2001 and 2003 to assess to what extent genetic variability exists for this trait in juvenile oysters Crassostrea gigas. For each generation, two groups were selected for their high ("R" for resistant) and low ("S" for susceptible) survival. Significant differences of mortality were observed during the first year in Rivière d'Auray (RA) (Brittany-France). However, low and similar mortality were observed for both groups, with no significant difference between groups in the first and second generation in this site, where the first summer is the critical sensitive period with a clear age effect. In Baie des Veys (BDV) (Normandie-France), where mortalities usually mainly affect adults, mortality between...

Overwiew on selective breeding and genetic improvement in bivalve shellfish ArchiMer
Boudry, Pierre.
Dr. Boudry focussed his talk on oysters, which are the most important bivalve species produced world wide (more than 4 million tons). One of the main characteristic of bivalve aquaculture is that most of the juveniles ("seed or "spat") are collected from natural recruitment (e.g. 100% of the European mussel production). Hatchery propagation is used in the case of introduced species (e.g. Crassostrea gigas in USA) or limited natural recruitment. The most significant genetic improvement for the production of Pacific oyster (C. gigas) to date has been obtained through the breeding of triploids, especially since the development of tetraploids. Triploidy lead to highly reduced gametogenesis and, as a result better growth and survival. Many quantitative...

Assessment of Genetic Variability of Fish Personality Traits using Rainbow Trout Isogenic Lines ArchiMer
Millot, Sandie; Pean, Samuel; Labbe, Laurent; Kerneis, Thierry; Quillet, Edwige; Dupont-nivet, Mathilde; Begout, Marie-laure.
The study of inter-individual variability of personality in fish is a growing field of interest but the genetic basis of this complex trait is still poorly investigated due to the difficulty in controlling fish genetic origin and life history. When available, isogenic lines that allow performing independent tests on different individuals having identical genotype constitute a very relevant experimental material to disentangle the genetic and environmental components of behavioural individuality. We took advantage of heterozygous isogenic lines to investigate the personality in rainbow trout through the analysis of their reactions to different experimental situations. To this end, seven to ten rainbow trout isogenic lines were screened for their spatial...

Genetic variability and selective breeding for traits of aquacultural interest in the Pacific oyster (Crassostrea gigas) ArchiMer

The most significant genetic improvement for production of Pacific oyster (Crassastrea gigas) has been obtained through the breeding of triploids, especially since the development of tetraploids. Quantitative genetics studies suggest that significant gains, for disease resistance or for other traits of aquacultural interest, could be obtained using this approach. However, the limited extent of hatchery-propagation (versus natural recruitment) and/or various technical difficulties and biological characteristics of the species have slowed the development of selective breeding programs. Recently, in the USA, Australia and New Zealand, family-based selective breeding programs have been initiated to improve growth and yield. In Europe, where both natural and...

Is avoiding full sib matings really worthwhile in a breeding program? ArchiMer
Dupont-nivet, Mathilde; Vandeputte, Marc.
Stochastic simulations were used to compare genetic progress and inbreeding depending on whether full sib matings were allowed or not when applying individual selection. Fifteen selection cycles were considered, using a single pair mating design. Two levels of heritability (h(2)) were studied (0.1; 0.5) as well as four different proportions of selected animals (2, 5, 10 and 20%), four population sizes (1000, 2,000, 4,000 and 10,000 candidates) and two levels of inbreeding depression (0 and 5% decrease of phenotype for 10% increase in inbreeding). Fifty replicates were generated for each set of parameters. The simulations revealed that avoiding full sib matings became less and less relevant as population size and proportion of selected animals increased and...

Residual genetic variability in domesticated populations of the Pacific blue shrimp (Litopenaeus stylirostris) of New Caledonia, French Polynesia and Hawaii and some management recommendations ArchiMer
Goyard, Emmanuel; Arnaud, Sophie; Vonau, Vincent; Bishoff, Vincent; Mouchel, Olivier; Pham, Dominique; Wyban, Jim; Boudry, Pierre.
The Latin American shrimp Litopenaeus stylirostris was introduced in three different Pacific islands (Tahiti, New Caledonia via Tahiti, and Hawaii) and hatchery-propagated for 7-25 generations to develop shrimp farming based on these domesticated stocks. Three microsatellite markers have been used in an attempt to assess the genetic bases of the populations available to start a selective breeding program. The comparison of eight hatchery stocks (five New Caledonian, two Hawaiian and one Tahitian stocks) and one wild Ecuadorian population showed a much lower variability in the domesticated stocks than in the wild population, especially in New Caledonia and Tahiti (2-3.7 vs. 14-27 alleles per locus; 20-60% vs. 90% expected heterozygosity). The Tahitian and...

Response to unintentional selection for faster development associated with inbreeding depression in Crassostrea gigas larvae, resulting from a broodstock selective breeding plan ArchiMer
Taris, Nicolas; Batista, Frederico; Marissal, Eric; Boudry, Pierre.
Direct and indirect consequences of selective breeding in marine bivalves still remain largely unexplored. For species with two-phase life cycle, like the Pacific oyster (Crassostrea gigas), most studies have focused on juvenile and adult stages, but relatively few have focused on juvenile and adult stages, but relatively few have considered the larval stage, especially in a domestication context. We assessed the impact of hatchery practices on larval traits, notably on larval growth (due to the culling of slow growing larvae), by the study of larval progenies. Larvae originating from crosses using parental oysters both from natural beds and from hatchery broodstock which had been selected for adult growth and shell shape for seven generations. A set of...

Impact of intensive larval rearing practices in oyster hatcheries : selective processes and loss of genetic variability (Poster) ArchiMer
Taris, Nicolas; Sauvage, Christopher; Boudry, Pierre.
Early-life history of oysters, like in most marine bivalves, is characterized by high fecundity and low survival rate. In hatcheries, which are ecoming more and more important for the production of juveniles for aquaculture production, rearing conditions are optimised (density, feeding, temperature...), and contrast with those observed in the wild. Additionally, slow growing larvae are culled by sieving.

Microsatellite-based parentage analysis of factorial crosses in Pacific oyster (Crassostrea gigas) larvae and spat ArchiMer
Taris, Nicolas; Sauvage, Christopher; Ernande, Bruno; Boudry, Pierre.
Several studies have suggested that significant genetic variability might exist for some early developmental traits in the Pacific oyster, C. gigas. However, most studies have examined only limited numbers of families. More critically, these families were rarely replicated, leading to possible bias due to heterogeneous larval rearing conditions among families. To overcome these potential problems, we used a mixed-family approach with subsequent parentage analysis. A recently developed set of three multiplexed microsatellite markers was used to determine parentage in a complete factorial cross between 12 males and 4 females. Larvae were reared at two different temperatures (20 and 26°C) to provide a contrast resembling wild versus hatchery conditions....

Genetic effect of domestication selective pressures on Pacific oyster at larval stage ArchiMer
Taris, Nicolas; Sauvage, Christopher; Ernande, Bruno; Boudry, Pierre.
Among bivalve species, the Pacific oyster, Crassostrea gigas, is the most economically important bivalve production over the world. Today, C. gigas is subject to an important production effort that leads to an intensive artificial selection. Larval stage is relatively unknown, specifically in a domestication context. Genetic consequence of artificial selection is still at a preliminary study. We aimed to tackle the consequence of inconscient domestication on the variance reproductive success focusing on larval stage, keystone of the life cycle. We studied two kinds of specific selective processes that common hatchery rearing practices exert : the effect of discarding the smallest larvae on genetic diversity and the artificial environment rearing effect via...
