Population genetics of the European flat oyster (Ostrea edulis) : from larvae to populations ArchiMer

Nowadays, PCR-based DNA markers allow the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European flat oyster (Ostrea edulis) populations has aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Results were congruent between allozymes, microsatellites and mitochondrial DNA and support an isolation by distance model. High variability was observed in the breadth of diversity between populations using a mitochondrial marker, this highlights the small effective population sizes in some locations. Enrichment of live foods. ArchiMer

Live foods such rotifers and Artemia are still indispensable for mass propagation of larval fish. At present, more than 20 fish and crustacean species are mass produced in Japanese governmental fish farming centres and private hatcheries. Fry are either released into the coastal areas or used for commercial cultivation. This culture technology is rapidly developing with new species being introduced each year and larval rearing is increasingly identified as the number one constraint. Artemia is used most extensively, particularly for the mass production of Kuruma prawn, Penaeus japonicus .

Variability in the onset of hatching of Maja brachydactyla Balss, 1922 (Brachyura: Majidae) in the English Channel in relation to sea temperature ArchiMer

Long-term monitoring of zooplankton and temperature has been carried out at the nuclear power station of Flamanville (west coast of Cotentin, English Channel, France) since 1977. Zooplankton sampling and temperature measurements took place at fortnightly intervals, particularly during the spring and summer. In addition, daily records of water temperature were recorded since 1986. The seasonal appearance of Maja brachydactyla Balss, 1922 first stage zoeae in the plankton outside the thermal plume near to the power station can greatly vary from year-to-year. Export of deep-sea hydrothermal particles, indigenous thermophilic microorganisms and larvae to the surrounding Ocean ArchiMer

To assess the production and the export of particulate and biological material in deep-sea hydrothermal vents, four moorings with sediment traps and current meters were deployed for 24 days in the Azores Triple Junction (ATJ) region. They were deployed along an axis starting at the base of a hydrothermal vent chimney and ending 1000 m away. The particles sampled at the base of the chimney were characterized by high concentrations in total sulfur (16%) and iron (4%). The particle composition changed drastically with distance from the vent: the sulfur concentration decreased with an increase in clay components. Influence of phytoplankton diet mixtures on microalgae consumption, larval development and settlement of the Pacific oyster Crassostrea gigas (Thunberg) ArchiMer

Microalgae commonly used as feed for bivalves, Pavlova lutheri (P), Isochrisis affinis galbana (T) and Chaetoceros calcitrans forma pumilum (Cp), were fed to Pacific oyster Crassostrea gigas to assess their nutritional value for larval development and metamorphosis during two experiments. Monospecific, bispecific and trispecific diets were firstly evaluated during 3 weeks from D larvae to young postlarvae. Microplastics are present in marine habitats worldwide and may be ingested by low trophic organisms such as fish larvae, with uncertain physiological consequences. The present study aims at assessing the impact of polyethylene (PE 10-45µM) microbeads ingestion in European sea bass (Dicentrarchus labrax) larvae. Fish were fed an inert diet including 0, 104 and 105 fluorescent microbeads per gram from 7 until 43 days post-hatching (dph). Microbeads were detected in the gastrointestinal tract in all fish fed diet incorporating PE. Our data revealed an efficient elimination of PE beads from the gut since no fluorescent was observed in the larvae after 48h depuration. Shrimp aquaculture in New Caledonia ArchiMer

In New Caledonia, there is no indigenous shrimp species for commercial aquaculture and it is necessary to control the complete cycle in captivity. Since 1973 and the creation of the Station d'Aquaculture de Saint-Vincent (S.A.S.V.), a joint project between IFREMER, France-Aquaculture and the territory of New Caledonia, nine species have been tested and for one of these, P. stylirostris , (introduced in 1980), we are now in a commercial scale production with the tenth generation obtained in captivity.

Nutrional inadequacy of Nannochloris atomus and Stichoccocucs bacillaris for the oyster Crassostre gigas (Thunberg) larvae ArchiMer

The evaluation of Nannochloris atomliS and StichoccocllS bacillaris as nutrient sources for oyster larvae has been attempted in the present work. After a study of their grawth and size distributions, these two species were used as food for Crassostrea gigas larvae. Despite praper sizes (2.5 to 20 µm3) the larval development of Crassostrea gigas was poor. Evaluation of ingestion and digestion of both algae, by means of epifluorescence microscopy, on 2 day and 6 day old larvae showed that they were normally ingested but weakly digested. Consequently, their nutritional inadequacy for larval molluscs may be also related to their indigestibility.

Effect of lipid level in a compound diet on the development of red drum (Sciaenops ocellatus) larvae ArchiMer

Red drum larvae were fed from day 11 to day 24 post-hatch, with Artemia (LP group) or two isoproteinic-formulated diets with varying lipid levels, 15% (L15 group) and 30% (L30 group). The lipid fraction was composed of a mixture of cod liver oil and soy lecithin. Larvae fed Artemia exhibited the greatest length, L30 diet induced a larvae length significantly greater than that obtained with L15 diet (P < 0,05). No difference was observed in larvae survival among the three dietary groups. The increase in dietary lipid concentration led to an increase in amylase and trypsin secretion levels in 24-day-old larvae. The food value of starch rich flagellates for Pecten maximus (Linné) larvae. Preliminary results ArchiMer

Pecten maximus larvae exhibit amylase as a main digestive enzyme indicating the possibility of starch hydrolysis. The food value of Rhodomonas salina and Porphyridium cruentum, both rich in starch, on the development of Pecten maximus larvae was investigated. The larval development was studied by means of an image analysis technique and epifluorescence microscopy was used for detecting ingestion and digestion of each phytoplankton species. The biochemical contents of algae and larvae were also assessed. The best growth of Pecten maximus larvae was observed when fed with the mixture PTS (plurispecific control). Dietary TAG source and level affect performance and lipase expression in larval sea bass (Dicentrarchus labrax) ArchiMer

The influence of dietary TAG source (fish oil, triolein, and coconut oil) and level (7.5 and 15% of the diet) on growth, lipase activity, and mRNA level was studied in sea bass larvae, from mouth opening until day 24 and from day 37 to 52. Fish oil and triolein induced better growth in both experiments, this being significant at a higher dietary level. Coconut oil significantly decreased growth at the higher level, possibly as the result of an excessive supply of medium-chain TAG. Growth was not related to lipase specific activity, suggesting a production in excess to dietary needs. Protein requirements of Penaeid shrimp. ArchiMer

Proteins are indispensable nutrients for growth and maintenance of live of all animals. The optimum protein levels in diets for shrimps are different among the various species. Squid meal is an effective protein source for many penaeids. The effects of dietary protein, lipid, and carbohydrate levels on the growth and survival of larvae of Penaeus japonicus were examined by feeding trials using purified diet with carrageenan as a binder. As a result, the effects of protein levels on growth and survival of P. japonicus larvae varied with dietary carbohydrate levels but not dietary lipid levels.

Ontogenic changes in the contents of dopamine, norepinephrine and serotonin in larvae and postlarvae of the bivalve Pecten maximus ArchiMer

In the present study, norepinephrine (NE), epinephrine (E), dopamine (DA) and 5-hydroxy-tryptamine (5HT) have been quantified by high performance liquid chromatography (HPLC) in Pecten maximus during larval and post-larval development. With average values ranging from 2 to 5 pg mug(-1) of protein, NE remained low through the whole larval life, while epinephrine (E) was undetected. DA and 5HT were the most abundant monoamines with significant variations between larval, pre-metamorphic and post-larval stages. During the first 20 days, corresponding to D larval and umboned larval stages, levels of DA and 5HT increased from 15 to 30 and 10 to 15 pg mug(-1) of protein, respectively. La conchyliculture, et principalement l’élevage de l’huître creuse, Crassostrea gigas, constitue la principale activité aquacole française. Cette activité repose, en grande partie, sur le recrutement naturel de l’espèce qui assure 60 à 70% des besoins en jeunes huîtres (naissain) : cette activité de collecte s’appelle le captage. Les deux principaux centres de captage en France sont les bassins d’Arcachon et de Marennes-Oléron. Or, depuis une quinzaine d'années, sur le bassin d'Arcachon, le captage devient très variable: à des années de captage nul (par exemple 2002, 2005, 2007) ou faible (2009, 2010, 2011) succèdent des années excellentes voire pléthoriques (2003, 2006, 2008, 2012, 2014). The quality of the scallop embryos is determined by the spawners characteristics as well as by the external factors they support during the maturation and the spawning phases. All these parameters are not still well controlled during the artificial conditioning in the hatcheries. As a consequence, the quality of the embryos is drastically variable and with them, the larvae performances in terms of survival and growth rates. It would be interesting to get a simple and quick test which would allow the estimation of the spawns quality. The problem of the clogging up of sea water pipes by Mytilus edulis larvae is a very real problem. A study has been made of the ways to destroy these larvae, (1) by electricity (publication in preparation), (2) by ultrasound. This last technique has given interesting data, but exploitation of results on a large scale presents many difficulties. An interesting general phenomenon is to be underlined. For ultrasound treatments of short duration (10s), many larvae are simply stressed and recover later on. For longer treatments, there is actual dislocation of the larvae and instant mortality, this mortality increases with time.

Using a continuous breeding of T. brevicornis the authors try to approach an experimental procedure in order to estimated the effects of thermal stresses on larvae produced by ovigerous females. In spite of the main factor driving to successful breeding was the healthiness of algal food, sublethalt or inhibitory effects were observed with duration were only observed to LT 50 level. On opposite, decrease in nauplii production is noted just after ovigerous females are exposed to 35 degree C of final temperature.
