Histological analysis of the callogenesis and organogenesis from root segments of Curcuma zedoaria Roscoe BABT
Mello,Marcia O.; Melo,Murilo; Appezzato-da-Glória,Beatriz.
Callus was induced from root segments taken from in vitro grown plants of Curcuma zedoaria Roscoe. The explants were cultured on agar-solidified Murashige and Skoog medium supplemented with 13.4muM of alpha-naphthaleneacetic acid and 2.2muM of 6-benzylaminopurine at 25ºC in the dark. Histological analysis revealed that callus was formed from the hypertrophied cortical parenchyma cells of the explant. Some of these cells underwent division while the surrounding cells accumulated starch. Callus was capable of shoot bud regeneration after 70 days when it was transfered to liquid medium of the same composition. After 30 days in liquid medium, buds developed from nodular structures. The adventitious shoots developed extensive root systems when they were placed...

Selective agent and A. tumefaciens overgrowth-control antibiotics in Eucalyptus camaldulensis Cotiledonary culture BABT
Quisen,Regina; Oliveira,Yohana de; Pileggi,Marcos; Cuquel,Francine; Quoirin,Marguerite.
The objectives of the present work were to establish the minimal lethal dose of the selective agent to determine the type and concentration of appropriate antibiotics for the elimination of Agrobacterium tumefaciens inoculated explants, without interfering with the regenerative potential of the E. camaldulensis cotyledonary explants. Non-transformed explants were cultivated in medium supplemented with kanamycin. The results showed that the antibiotic was suitable for the selection of transformed cells in the concentration of 9 mg L-1 as it inhibited the growth of non-transformed cells. Cotyledons infected with A. tumefaciens were cultivated in MS N/2 medium supplemented with BAP, ANA, Km and cefotaxime or AugmentinÒ . The highest average of regenerated...

Developmental effects of additional ultraviolet a radiation, growth regulators and tyrosine in Alternanthera brasiliana (L.) Kuntze cultured in vitro BABT
Silva,Nina Cláudia Barboza; Macedo,Andrea Furtado; Lage,Celso Luís Salgueiro; Esquibel,Maria Apparecida; Sato,Alice.
Tissue cultures of Alternanthera brasiliana (L.) Kuntze were treated with different growth regulators (Kinetin and 2,4-D), tyrosine and Ultraviolet A radiation (UV-A; 320 - 400 nm) to investigate their effects on development and pigment production. Nodal segments of plantlets grown from seeds were inoculated in all tested media and different light conditions. After eight weeks, this material was used to evaluate biomass, chlorophyll and betacyanin production. The Murashige and Skoog (MS) + kinetin medium resulted in development of approximately four shoots/explant. This medium plus white light was the best combination for micropropagation with the highest rooting percentage and betacyanin production. Plantlets grown under UV-A illumination reduced biomass...

Effect of alcohols on filamentation, growth, viability and biofilm development in Candida albicans BJM
Chauhan,Nitin M; Shinde,Ravikumar B; Karuppayil,S. Mohan.
In this study we report the potential of alcohols as morphogenetic regulators in Candida albicans. All the alcohols tested influenced various modes of growth like planktonic as well as biofilm forms. Viability was affected at high concentrations. Among the alcohols, the response of C. albicans to amyl alcohol (pentanol) was noteworthy. Amyl alcohol at a concentration 0.5% which was not inhibitory to growth and viability specifically inhibited morphogenetic switching from yeast to hyphal forms. It also inhibited normal biofilm development favoring yeast dominated biofilms. Based on this study we hypothesize that alcohols produced under anaerobic conditions may not favor biofilm development and support dissemination of yeast cells. Since anaerobic conditions...

Exploring the Potential use of Photo-Selective Nets for Fruit Growth Regulation in Apple Chilean J. Agric. Res.
Bastías,Richard M; Manfrini,Luigi; Corelli Grappadelli,Luca.
The effect of shading (i.e. reduction of sunlight availability) on fruit growth physiology has been widely studied in apple (Malus domestica Borkh.), but little knowledge exist about fruit growth responses to changes in the light spectrum. The aim of the present research was to study the effect of use of colored nets with differential sunlight transmission in the blue (B, 400-500 nm), red (R, 600-700 nm) and far-red (FR, 700-800 nm) spectra on apple fruit growth and physiological associated responses. Three year old 'Fuji' apple trees were covered with 40% photo-selective blue and red shade nets, 40% neutral grey shade net, and 20% neutral white net as control. Red and blue net reduced in the same proportion (27%) the photosynthetically active radiation...

Identification of tiller age categories based on morphogenetic responses of continuously stocked marandu palisade grass fertilised with nitrogen Ciência Rural
Paiva,Adenilson José; Pereira,Lilian Elgalise Techio; Silva,Sila Carneiro da; Dias,Raphael Antonio Prado.
Nitrogen fertilization augments tiller appearance and death, modifying the age profile of tiller population. Tillers from different age categories present distinct responses to environmental factors and management practices characterized by variation in their morphogenetic responses, which interfere in the dynamics of the herbage accumulation process. In that context, rates of leaf appearance and elongation and leaf lifespan were used as indicators to identify tiller age categories of continuously stocked marandu palisade grass managed at 30cm and subjected to nitrogen fertilization regimes using cluster analysis. Analysis was based on the average distance method using combined means of the response-variables studied, and the results used to build a...

Levels of defoliation and regrowth dynamics in elephant grass swards Ciência Rural
Rodolfo,Giselle Regina; Schmitt,Daniel; Dias,Kamila Maciel; Sbrissia,André Fischer.
The morphogenetic and structural characteristics of forage plants and the processes of growth and senescence in tissues may vary according to the grazing management strategies used. This research aimed to assess the process of regrowth in elephant grass swards Cv. 'Pioneiro' submitted to two pre-grazing heights (90 and 120cm) combined with two levels of defoliation (50 and 70%). A reduction of the leaf elongation rate (LER) was observed during the regrowth stages, in addition to a reduction in the rate of leaf appearance (LAR) and aerial tiller population density (ATPD). In contrast, an increase was observed in leaf senescence (LSR) and stem and pseudo stem elongation rates (SER). The severe defoliation (70%) combined with a pre-grazing height of 120cm...

Plant morphology and herbage accumulation of signal grass with or without fertilization, under different light regimes Ciência Rural
Lopes,Clenardo Macedo; Paciullo,Domingos Sávio Campos; Araújo,Saulo Alberto do Carmo; Morenz,Mirton José Frota; Gomide,Carlos Augusto de Miranda; Maurício,Rogério Martins; Braz,Thiago Gomes dos Santos.
ABSTRACT: The silvopastoral system has been suggested as an alternative to recover degraded pastures in tropical regions. However, trees reduce the light available for pastures, which may affect the growth and herbage accumulation. The objective of this study was to evaluate the morphogenesis, canopy structure and herbage accumulation of signalgrass ( Brachiaria decumbens ) subjected to three light regimes (0, 20 and 70% of natural shading) and two fertilization levels (presence or absence of nitrogen, phosphorus and potassium). Leaf and stem elongation rates increased under shading but did not vary with fertilization. The leaf appearance rate was greater under fertilizer treatment but was generally similar among light regimes. The tiller density was...
