Diagnosis of oyster herpes-like virus: development and validation of molecular immunological and cellular tools "VINO". Second periodic progress report: 4th January 2000 at 3rd January 2001 ArchiMer
Renault, Tristan; Davison, Andrew; Xhonneux, Florence; Dorange, Germaine; Culloty, Sarah; Novoa, Beatriz; Dixon, Peter.
The aim of the programme is to develop specific tools for diagnosing herpes-like virus infections in bivalves and to validate these reagents by using them in different European laboratories involved in shellfish epidemiological surveys. This will done using teclmiques developed by the patiners to characterise viruses by studying their genome and immunologically reactive proteins, to cultivate oyster cells and vertebrate cell lines and to perform epidemiological surveys among bivalves. The programme objectives are: 1 - Obtaining the complete oyster herpesvirus (OsHV-1) DNA sequence with determination of the genome structure. 2 - Comparing OsHV -1 with viruses belonging to the Herpesviridae family on the basis of sequence data and genome structure. 3 -...

Rickettsia-like organism infecting juvenile sea-bass Dicentrarchus labrax ArchiMer
Comps, Michel; Raymond, J. C.; Plassiart, G. N..
High levels of mortalities (20%) associated with abnormal swimming have been observed in farmed juveniles of seabass, Dicentrarchus labrax, in Mediterranean coast, southern France. Histopathology was obvious in the brain with nervOUS tissue necrosis and the presence of ceUs containing basophilic granules corresponding to smaU coccoid organisms. Ultrastructural examination revealed these organisms were rickettsia-like organisms morphologicaUy close to Piscirickettsia salmonis

Le réseau de pathologie des mollusques. REPAMO ArchiMer
Grizel, Henri.
The creation of a mollusc pathology network was in effect and made official during 1992. The subject of this document is to present this network, which has not yet achieved its definitive size. Why this network? The creation of this network responds to several institutional and scientific needs. Indeed, the setting up of control measures aimed at regulating mollusc exchanges notably via European Directive 91/67 and its transcription into French law has, as a corollary, caused the recognition of IFREMER as the agency charged with performing the analyses necessary for responding to the needs of this Directive, as far as marine invertebrates are concerned. Moreover, France being among the primary producers of shellfish in the world, the setting up of a...

Histological study of a cellular reaction in Ruditpaes decussatus infected by a protozoan ArchiMer
Chagot, Dominique; Comps, Michel; Boulo, Viviane; Ruano, Francisco; Grizel, Henri.
An infection caused by a Perkinsus -like parasite has been observed in Ruditapes decussatus from Portugal (Comps and Chagot, 1987). Following a pathological study carried out on clams affected by mortalities in the Algarve area, this parasite was found again, inducing a reaction process in the host. Histological examination reveals the parasite in the connective tissue of different organs. The adjoining tissues, particularly the epithelia, are not affected.

Prophylactic strategies and zootechnic measures, recent advances ArchiMer
Grizel, Henri.
The spread of major diseases of molluscs around the world and the increase of transfers need the development of adapted prophylactic strategies and the increase of research in epidemiology, immunology and genetics. The prophylactic strategies must be based on common laws with the establishment of a list of declarable pathogens, with basic sampling rules (sample size, frequency, definition of geographical areas) and with reglementation of the internal transfers. Moreover, specific adapted diagnosis should be developed to increase the sanitary control performance (e.g. ELISA test, DNA probe, cell culture). In the case of an infectious disease the epidemiology studies should be concerned, the relationship between the culture techniques and the disease, the...

Some regulatory issues related to perkinsosis ArchiMer
Berthe, Franck.
Perkinsosis is one of the OIE listed diseases of molluscs caused by Perkinsus marinus and P. olseni. Consequently, molluscs originating from endemic areas should not be transferred into areas free of the disease. Impediments to the implementation of international zoosanitary regulation will be reviewed.

Fleury, Pierre-gildas; Langlade, Aime; Le Coguic, Marie-jo; Thebault, Anne; Chellet, B.; Ponthoreau, C.; Devulder, G..

Doan, Quoc Khanh.
European seabass is one of the most economic species in aquaculture in Mediterranean areas. Viral nervous necrosis (VNN), a disease affecting at least 70 aquatic species, has become the most serious threat to seabass cultured industry. While numerous studies have been performed in order to control this disease, no simple and effective procedures are available. In this thesis, we question genetic variability and the potential of selective breeding as an opportunity to address thwart this threat. After a general introduction (first chapter) and a deep literature review of nodavirus in aquaculture (second chapter), we explore in the third chapter the genetic variability of resistance of different wild populations of European seabass, namely Northern Atlantic...

Among the marine molluscs, Pacific oyster, Crassostrea gigas, is actually the most reared species in the world. Noticeably in France, where it was imported between 1960 and 1970, and replaced Portuguese oyster, Crassostrea angulata. Intensification of breeding and the associated introductions of new species may be hazardous. Indeed, they might be responsible for the appearance and the dissemination of new diseases in bivalve molluscs. In addition to these observations, the quasi monobreeding of C. gigas in France enhances the necessity to develop research concerning the diagnosis of pathologies affecting this species, namely the diagnosis of viral diseases. Several families of viruses have been described within marine bivalves molluscs. The purpose of this...

Parmi...

Temperature influence on pathogen transmission and subsequent mortalities in juvenile Pacific oysters Crassostrea gigas ArchiMer
Petton, Bruno; Pernet, Fabrice; Robert, Rene; Boudry, Pierre.
Since 2008, mass mortalities of 1-yr-old Crassostrea gigas associated with the ostreid herpesvirus OsHV-1 μVar have occurred along all the coasts of France when seawater temperature reaches 16 to 17°C. The present study aimed to characterize the effect of temperature on oyster survival in combination with OsHV-1 DNA quantification by standard real-time PCR and total vibrio population levels in oyster tissues. To examine the effect of seawater temperature on disease transmission and related mortality of oysters, cohabitation experiments were conducted between healthy naïve oysters and oysters previously exposed to field conditions in areas where mortalities were occurring. Oysters initially maintained in controlled conditions (free of mortality and negative...

Marteiliosis in molluscs: A review ArchiMer
Berthe, Franck; Leroux, Frederique; Adlard, Robert; Figueras, Antonio.
Among mollusc pathogens, paramyxeans are an important group of protistan parasites belonging to the genera Marteilia and Marteilioides. Martelia refringens and M. sydneyi are of particular concern given their potential impact on mollusc aquaculture world-wide. Aber disease and QX disease are currently listed by the OIE, the World Organisation for Animal Health. After more than thirty years of existence in the scientific literature, these organisms still pose questions and research challenges to investigators. This paper reviews current knowledge of the group and key references. The review was focused on taxonomy, epidemiology, pathology and potential control methods for these organisms in order to enhance understanding of paramyxean infection issues in...

The velvet swimming crab Necora puber, commonly found on the French coast was an important resource for inshore fishermen of the Mor-Braz sea in Southern Brittany before the production collapsed in 1985. Although there are still many uncertainties as to the reasons for this, we note that the collapse took place after a significant ecological imbalance which locally caused massive mortality of many marine organisms in 1982. Without being obviously linked to the former point, mortality due to pathological causes was observed in velvet swimming crabs. Seeing the lack of general knowledge about these crabs, this study was designed to acquire data about its fishery and its biology. The first part focused on fleet, fishing effort and yield trends in the...

ArchiMer
Comps, Michel; Duthoit, Jean-louis.
Two viral infections associated to some epizooties have been observed in Crassostrea angulata Lmk. The first one concerns the Gill disease in which the viral damages go hand in hand with a cellular hypertrophy, while, in the second one, the contaminated cells are associated with a deterioration of the connective tissue. These branchial necrosis associated to a virus have been observed again recently in some Portuguese oysters from the natural deposits of the Cadix (Spain) area. Moreover, the study of the summer mortalities (July 1977) in the oyster Crassostrea gigas Th. demonstrated the presence in this species of a virus of the same type as the one associated to the massive mortalities of the Portuguese oyster. In both cases, the viral morphogenesis takes...

Combined oral toxicity of azaspiracid-1 and yessotoxin in female NMRI mice ArchiMer
Aasen, John A. B.; Espenes, Arild; Miles, Christopher O.; Samdal, Ingunn A.; Hess, Philipp; Aune, Tore.
For many years, the presence of yessotoxins (YTXs) in shellfish has contributed to the outcome of the traditional mouse bioassay and has on many occasions caused closure of shellfisheries. Since YTXs do not appear to cause diarrhoea in man and exert low oral toxicity in animal experiments, it has been suggested that they should be removed from regulation. Before doing so, it is important to determine whether the oral toxicity of YTXs is enhanced when present together with shellfish toxins known to cause damage to the gastrointestinal tract. Consequently, mice were given high doses of YTX, at 1 or 5 mg/kg body weight, either alone or together with azaspiracid-1 (AZA1) at 200 μg/kg. The latter has been shown to induce damage to the small intestine at this...

The REPAMO is a French Network for the surveillance of Mollusc diseases that notably meets the obligations of the European Directive 2006/088/EC. It is entirely funded by the French State. The coordination of this network entrusted to the Institut Français de Recherche pour l`Exploitation de la Mer since 1986 aims at monitoring the notifiable endemic diseases, detecting the emergence of exotic or new pathogens and ensuring a minimum level of surveillance of the general health condition of molluscs. Beside IFREMER, different partners are involved in the surveillance of mollusc health at the national level: the Departmental direction for territories and sea who are the local representatives of the competent authority and the producers who have the obligation...

Latrouite, Daniel; Morizur, Yvon; Noel, Philippe; Chagot, Dominique; Wilhelm, G.
Unusural crab mortality in winter led us to identify a Hematodinium like parasitic dinoflagellate in the blood of edible crabs Cancer pagurus. Crabs from several fishing areas (the English Channel, the Iroise sea, the bay of Biscay, off Scotland) and from one area of the Channel shore were examined. The crab parasit was found everywhere we investigated and throughout the year. The prevalence which was found independant of the intermolt stage, sex and size of crabs was higher inshore than offshore. The pathogenicity is assumed to differ according to season.
