A new bathyphotometer for bioluminescence measurements on the Armorican continental shelf (northeastern Atlantic) ArchiMer

A new bathyphotometer has been developed by the Oceanographic Laboratory of the French Naval Academy which has performed a series of in situ bioluminescence: measurements since 1994. It is used as an additional probe to a Seabird (R) CTD. It measures planktonic bioluminescence. It is associated with a SeaTech (R) fluorometer. The Armorican shelf has particularly been investigated since July 1994. Results obtained in January and April 1995 off Brittany (France), at the entrance to the Bay of Douarnenez and in the northern Bay of Biscay are presented as examples. Présence d'un Protogonyaulax sp. sur le littoral atlantique français pendant l'hiver 1987 ArchiMer

A dinoflagellate belonging to Protogonyaulax genera and forming more than 20 cells chains was observed from November 1987 to February 1988 in Vilaine Bay and in the west ofIle d'Yeu (France). This species was not detected in coastal phytoplankton during preceding surveys. Close to Protogonyaulax affinis it seems to be a winter developing species for this coastal area. Vertical distribution shows in december a migration of the patch towards deeper layers. Increasing occurrences of species belonging to Protogonyaulax (Alexandrium Balech) genera inshore or coastal waters need to pay more attention to the taxonomy of such organisms. Some species are effectively well known as poisons producers.

This is the 37th annual surveillance report concerning the nuclear power plant of Gravelines. Its very high power level results from six plant units located on the shore and is characterised by a cooling system with a coastal surface input and output. Dynamique des populations bactériennes au cours de deux productions expérimentales de phytoplancton marin naturel ArchiMer

Quantitative and qualitative variations of the hetero­trophic bacterial population, during two experimental induced phytoplanktonic blooms are described. When the phytoplanktonic population deceases, the bacte­rial population exhibits a significant increase. The spring microflore is quite different from the fall ones. Microbiological, physiological and biochemical fluctuations indicate qualitative fluctuations. Bacterial strains are identified as "bacterial profiles".

Blastodinium is a genus of dinoflagellates that live as parasites in the gut of marine, planktonic copepods in theWorld’s oceans and coastal waters. The taxonomy, phylogeny, and physiology of the genus have only been explored to a limited degree and, based on recent investigations, we hypothesize that the morphological and genetic diversity within this genus may be considerably larger than presently recognized. To address these issues, we obtained 18S rDNA and ITS gene sequences for Blastodinium specimens of different geographical origins, including representatives of the type species. Bacterial response to organic (amino acids) or inorganic (NH sub(4) super(+)) nitrogen was studied in 350 liter closed batch systems. The experiments demonstrate an important NH sub(4) super(+) remineralization associated with bacterial growth. The differences between substrate and bacterial C/N ratios and bacterial metabolic demands partly explain the high values of ammonia produced. Grazing by heterotrophic microflagellates resulted in an important release of NH sub(4) super(+). This suggests and important role of grazers for the nitrogen remineralization in natural waters.

Zoo/PhytoImage 4 is an « open source » software based on R and ImageJ. It processes numerical images of plankton particles digitized using a FlowCAM, a flat-bed scanner, microor macrophotos, etc. The general concept consists in the individual outlining of particles on the pictures, followed by their measurements (so-called « attributes ») such the size, the shape, transparency, textures, etc. These attributes are then used by a classification tool to automatically predict the taxonomic group the particles belong to. The classifier is obtained after a learning stage using a machine learning algorithm and a training set of pre-identified particles. Importance et role du materiel organique vivant et inerte dans les suspensions de la Baie de Seine ArchiMer

Living and inert organic matter take an important place inside suspensions in Seine Bay. There particulate matter and the organo-mineral particles can induce granulometric modes, which constitute a determinant component for dynamics characterizing of suspensions. Organic fraction, in particular, plankton (diatoms and zooplankton) is a main factor in open sea which can concentrate when coming into contact with mass water fronts. Organo-mineral aggregates are present in all estuaries. Importance and action of this organic particulate matter are studied for a best knowledge of this ecosystem.

Simplified Transformation of Ostreococcus tauri Using Polyethylene Glycol ArchiMer

Ostreococcus tauri is an easily cultured representative of unicellular algae (class Mamiellophyceae) that abound in oceans worldwide. Eight complete 13–22 Mb genomes of phylogenetically divergent species within this class are available, and their DNA sequences are nearly always present in metagenomic data produced from marine samples. Here we describe a simplified and robust transformation protocol for the smallest of these algae (O. tauri). Polyethylene glycol (PEG) treatment was much more efficient than the previously described electroporation protocol. Short (2 min or less) incubation times in PEG gave >104 transformants per microgram DNA. In French Polynesia, the black-lip pearl oyster Pinctada margaritifera has been farmed to produce pearls since the 1980s, forming the basis of a major industry. The sustainability of this activity relies on spat collection in the lagoons. However, pearl oyster spat can be difficult to identify for the evaluation of stock variations. It is especially hard to distinguish Pinctada spp. larvae at a very early stage of development. Investigations carried out in both Corsica lagoons Diana and Urbino during the period from January to August 1985, permitted to define the main characteristics of microplankton cycle. Two phytoplanktonic blooms have been observed, first in winter, second less important in July-August. Any correlation cannot be observed between phytoplanktonic cycle or one its main components, and both environmental variables temperature and salinity. However dissolved oxygen ratio may be subject to large summer variations, drifting towards until ecological desequilibrious.

Afin de fournir une meilleure idée sur la structure et la dynamique de la communauté des dinoflagellés et leurs relations avec les paramètres environnementaux dans les zones semi-fermées en communication avec le golfe de Tunis, une étude a été réalisée dans les ports puniques de Carthage afin d’estimer les densités et la dynamique des espèces de dinoflagellés ainsi que les variations des concentrations en nutriments, de janvier à juin 2006. Un nombre de 66 espèces de dinoflagellés ont été identifiés dont 14 espèces sont connues pour être nocives et responsables d’efflorescences nuisibles alors que 21 espèces sont connues pour être productrices de toxines.
