Toxic factors of Vibrio strains pathogenic to shrimp ArchiMer

Vibriosis is a major disease problem in shrimp aquaculture. 'Syndrome 93' is a seasonal juvenile vibriosis caused by Vibrio penaeicida which affects Litopenaeus stylirostris in grow-out ponds in New Caledonia. This study assessed the toxic activities of extracellular products (ECPs) from V: penaeicida, V. alginolyticus and V, nigripulchritudo using in vivo injections in healthy juvenile L. stylirostris (= Penaeus stylirostris) and in vitro assays on shrimp primary cell cultures and the fish cell line epithelioma papulosum cyprini (EPC). Toxic effects of ECPs were demonstrated for all pathogenic Vibrio strains tested both in vivo and in vitro, but for shrimp only; no effect was observed on the fish cell line. Etude ecologique des vibrions du bassin d'Arcachon ArchiMer

Water and sediment samples monthly collected from Arcachon Bay (Gironde, France) were examined during three years for the presence of marine vibrios. V. cholerae serotype 01 was never detected. On the other hand, the Eyre River and mouth contain non-agglutinating V. cholerae (127 strains) principally in water. The low temperatures slow their development down without stopping it completely. V. parahaemolyticus (257 strains) present in the same areas is particularly isolated from water when the aquatic medium temperature rises to 15 degree C. V. alginolyticus (638 strains) settles the whole Bay, except the river, in any season. The low temperatures promote growth of V. metschnikovii (203 strains) very tolerant towards salinity. Highly diverse recombining populations of Vibrio cholerae and Vibrio parahaemolyticus in French Mediterranean coastal lagoons ArchiMer

Vibrio parahaemolyticus and Vibrio cholerae are ubiquitous to estuarine and marine environments. Thes two species found in Mediterranean coastal systems can induce infection in humans. Environmental isolates of V. cholerae (n = 109) and V. parahaemotycius (n = 89) sampled at different dates, stations and water salinities were investigated for virulence genes and by a mutilocus sequence-based analysis (MLSA). V. cholerae isolate were all ctxA negative and only one isolate of V. parahaemolyticus displayed trh2 gene. Most Sequence Types (ST) corresponded to uique ST isolated at one date or one station. Phenoloxidases (POs) are a group of copper proteins including tyrosinase, catecholase and laccase. In several insects and crustaceans, antibacterial substances are produced through the PO cascade, participating in the direct killing of invading microorganisms. However, although POs are widely recognised as an integral part of the invertebrate immune defence system, experimental evidence is lacking that these properties are conserved in molluscs, and more particularly in the Pacific oyster Crassostrea gigas. First description of French V. tubiashii strains pathogenic to mollusk: II. Characterization of properties of the proteolytic fraction of extracellular products ArchiMer

Extracellular products (ECPs) of the French V. tubiashii strain 07/118 T2 were previously reported to be toxic for the Pacific oyster Crassostrea gigas. In this study we now assessed host cellular immune responses and bacterial potential effectors by which these ECPs can be associated with host damages. The adhesion capacity (28% inhibition) and phagocytosis ability (56% inhibition) of oyster hemocytes were the main functions affected following in vitro contact between hemocytes and V. tubiashii ECPs. This may be linked to the demonstration of the capability of ECPs to cleave various cellular substrates as oyster collagen. The presence of Vibrio bacteria in raw shellfish poses a significant health hazard to the consuming public. These human pathogens are natural, seasonal components of the microbial flora of shellfish which can result in primary septicemic or diarrheal fatal infections. Existing shellfish depuration neither targets nor eliminates the Vibrio bacteria. For this reason, additional purification processes are necessary. Uso de probióticos na produção de pós-larvas de camarão-rosa PAB

O objetivo deste trabalho foi avaliar o uso de probióticos na produção de pós-larvas do camarão-rosa (Farfantepenaeus brasiliensis). Utilizou-se o delineamento experimental inteiramente casualizado, com dois tratamentos - uso de probióticos (Bacillus spp.) e controle com uso de antibiótico (eritromicina) - e quatro repetições. Pós-larvas no estágio PL1 foram estocadas em cada parcela experimental (3,0 L) e cultivadas até o estágio PL10. As variáveis de qualidade de água e o desenvolvimento de bactérias do gênero Vibrio, na água de cultivo, foram monitorados ao longo do experimento. Abstract The objective of this work was to evaluate the effect of Lactobacillus plantarum on the preservation of fresh mussels and its effect on the attractiveness, consumption, and midgut bacterial microbiota of Pacific white shrimp broodstock. The experiment evaluated mussels stored with L. plantarum at 4ºC. The controls were: mussels stored at -18ºC without L. plantarum; mussels stored at -18ºC with L. plantarum; and mussels stored at 4ºC without L. plantarum. This study evaluated the pathogenic characteristics of bacteria isolated from Litopenaeus vannamei during an outbreak at the Laboratory of Marine Shrimp, UFSC, Santa Catarina State, Brazil. Their virulence potential in larvae and juvenile shrimp and the effects on the total haemocyte count, phenoloxidase activity and serum agglutinate titre were examined after experimental infection. Bacterial strains were isolated from larvae and adult shrimps, identified by the AP120E biochemical system as: two strains of Vibrio alginolyticus, three of Aeromonas salmonicida and one of Pasteurella multocida sp. and Pasteurella sp. Adherence assays and Slime production of Vibrio alginolyticus and Vibrio parahaemolyticus BJM

In this study we investigated the phenotypic slime production of Vibrio alginolyticus and Vibrio parahaemolyticus strains, food-borne pathogens, using a Congo red agar plate assay. Furthermore, we studied their ability to adhere to abiotic surfaces and Vero cells line. Our results showed that only V. alginolyticus ATCC 17749 was a slime-producer developing almost black colonies on Congo red agar plate. Adherence to glace tube showed that all V. alginolyticus strains were more adherent than V. parahaemolyticus. Only V. alginolyticus ATCC 17749 was found to be able to form biofilm on polystyrene microplate wells (OD570 = 0.532). Adherence to Vero cells showed that all tested strains were non adherent after 30 min, however after 60 min all the studied strains...
