A chemically defined medium for production of actinomycin D by Streptomyces parvulus BABT

A chemically defined medium consisting of D(+)fructose, L(-)threonine, K2HPO4, MgSO4.7H2O, ZnSO4.7H2O, CaCl2.2H2O, FeSO4.7H2O and deionized water, was developed to maximize the synthesis of actinomycin D by the Streptomyces parvulus DAUFPE 3124 strain. This medium resulted in the maximum antibiotic concentration of 133mg/L while using the original medium the production of actinomycin D was poor not surpassing 43mg/L.

Root-knot nematodes are microscopic round worms, which cause severe agricultural losses. Their attacks affect the productivity by reducing the amount and the caliber of the fruits. Chemical control is widely used, but biological control appears to be a better solution, mainly using microorganisms to reduce the quantity of pests infecting crops. Biological control is developing gradually, and with time, more products are being marketed worldwide. Optimization of fibrinolytic protease production from Bacillus subtilis I-2 using agro-residues BABT

The aim of this work was to study the production of fibrinolytic protease by Bacillus subtilis I-2 on agricultural residues. Molasses substantially enhanced (63%) protease production (652.32 U/mL) than control (398.64 U/mL). Soybean meal supported maximum protease production (797.28 U/mL), followed by malt extract (770.1 U/mL), cotton cake (761.04 U/mL), gelatin (742.92 U/mL) and beef extract (724.8 U/mL). Based on the Plackett-Burman designed experiments, incubation time, soybean meal, mustard cake and molasses were identified as the significant fermentation parameters. Nutritional requirements of Zymomonas mobilis CCT 4494 for levan production BABT

The aim of this work was to study the nutritional requirements of Zymomonas mobilis CCT 4494 for levan production in three chemically defined media. During the optimization of the fermentative process for the production of the exopolysaccharide, different concentrations of glucose, fructose and sucrose as carbon source and yeast extract as vitamin source were tested. Variations of incubation temperature and initial pH of the medium were observed. The results showed that medium containing 20.0% sucrose and 0.5% yeast extract, with initial pH of 7.0, incubated at 30°C gave a 43.0% yield of the biopolymer.

Comparative study of two purified inulinases from thermophile Thielavia Terrestris NRRL 8126 and mesophile Aspergillus Foetidus NRRL 337 grown on Cichorium Intybus l BJM

Thirty fungal species grown on Cichorium intybus L. root extract as a sole carbon source, were screened for the production of exo-inulinase activities. The thermophile Thielavia terrestris NRRL 8126 and mesophile Aspergillus foetidus NRRL 337 gave the highest production levels of inulinases I & II at 50 and 24 ºC respectively. Yeast extract and peptone were the best nitrogen sources for highest production of inulinases I & II at five and seven days of incubation respectively. The two inulinases I & II were purified to homogeneity by gel-filtration and ion-exchange chromatography with 66.0 and 42.0 fold of purification respectively. Optimization of clavulanic acid production by Streptomyces daufpe 3060 by response surface methodology BJM

Clavulanic acid is a β-lactam antibiotic which has a potent β-lactamase inhibiting activity. In order to optimize its production by the new isolate Streptomyces DAUFPE 3060, the influence of two independent variables, temperature and soybean flour concentration, on clavulanic acid and biomass concentrations was investigated in 250 mL-Erlenmeyers according to a 2² central composite design. To this purpose, temperature and soybean flour (SF) concentration were varied in the ranges 26-34°C and 10-50 g/L, respectively, and the results evaluated utilizing the Response Surface Methodology. Kinetic characterization of glucose aerodehydrogenase from Aspergillus niger EMS-150-F after optimizing the dose of mutagen for enhanced production of enzyme BJM

In the present study enhanced production of glucose aerodehydrogenase from Aspergillus niger has been achieved after optimizing the dose of chemical mutagen ethyl methane sulfonate (EMS) that has not been reported earlier. Different doses of mutagen were applied and a strain was developed basing upon the best production. The selected strain Aspergillus niger EMS-150-F was optimized for nutrient requirements in order to produce enzyme through fermentation and the results showed the best yield at 2% corn steep liquor (CSL), 36 hours fermentation time, pH 5, 30°C temperature, 0.3% KH2PO4, 0.3% urea and 0.06% CaCO3. Glucose(xylose) isomerase production by Streptomyces sp. CH7 grown on agricultural residues BJM

Streptomyces sp. CH7 was found to efficiently produce glucose(xylose) isomerase when grown on either xylan or agricultural residues. This strain produced a glucose(xylose) isomerase activity of roughly 1.8 U/mg of protein when it was grown in medium containing 1% xylose as a carbon source. Maximal enzymatic activities of about 5 and 3 U/mg were obtained when 1% xylan and 2.5% corn husks were used, respectively. The enzyme was purified from a mycelial extract to 16-fold purity with only two consecutive column chromatography steps using Macro-prep DEAE and Sephacryl-300, respectively. Optimization and characterization of alkaline protease and carboxymethyl-cellulase produced by Bacillus pumillus grown on Ficus nitida wastes BJM

The potentiality of 23 bacterial isolates to produce alkaline protease and carboxymethyl-cellulase (CMCase) on Ficus nitida wastes was investigated. Bacillus pumillus ATCC7061 was selected as the most potent bacterial strain for the production of both enzymes. It was found that the optimum production of protease and CMCase were recorded at 30 °C, 5% Ficus nitida leaves and incubation period of 72 h. The best nitrogen sources for protease and CMCase production were yeast extract and casein, respectively. Also maximum protease and CMCase production were reported at pH 9 and pH 10, respectively. The results of a study of the impact of market management on the ornamental plants supply chain are presented, using Curitiba, which is considered the most expressive flower market in Paraná State, as a case study. The flower market in Paraná ranks fifth in Brazil in terms of the volume of commercialization and sixth in terms of per capita consumption. Effect of the chemical hydrogen cyanamide (CH2N2) on the thinning of flowers and fruits of the peach cultivar Coral was assessed in this study. ABSTRACT: Currently, using smaller trees is one of the most important trends in apple cultivation. It is expected that reduced size plants produce lower amount of fruit, but with high yield and yield efficiency. The aim of this study was to evaluate the yield efficiency (YE) of nine apple tree cultivars, grafted on two rootstocks, from the second to the seventh year after planting. The YE indexes oscillated from one to the other year, regardless of rootstock. Correlations between YE and yield per tree and between YE and trunk cross sectional area (TCSA) confirmed that efficiency can be increased by the production increment or by the plant's vigor reduction. Two groups of 180 Isa Brown commercial layer pullets were reared in cages (CR) or floor pens (FR) and transferred to furnished cages, where their production, egg quality and physical condition was observed throughout the laying period (18-78wks of age).
