Gene expression profiling of clinical stages II and III breast cancer BJMBR
Folgueira,M.A.A.K.; Brentani,H.; Katayama,M.L.H.; Patrão,D.F.C.; Carraro,D.M.; Mourão Netto,M.; Barbosa,E.M.; Caldeira,J.R.F.; Abreu,A.P.S.; Lyra,E.C.; Kaiano,J.H.L.; Mota,L.D.; Campos,A.H.J.F.M.; Maciel,M.S.; Dellamano,M.; Caballero,O.L.S.D.; Brentani,M.M..
Clinical stage (CS) is an established indicator of breast cancer outcome. In the present study, a cDNA microarray platform containing 692 genes was used to identify molecular differences between CSII and CSIII disease. Tumor samples were collected from patients with CSII or CSIII breast cancer, and normal breast tissue was collected from women without invasive cancer. Seventy-eight genes were deregulated in CSIII tumors and 22 in CSII tumors when compared to normal tissue, and 20 of them were differentially expressed in both CSII and CSIII tumors. In addition, 58 genes were specifically altered in CSIII and expression of 6 of them was tested by real time RT-PCR in another cohort of patients with CSII or CSIII breast cancer and in women without cancer....

Transcriptional up-regulation of PHLDA1 by 17β-estradiol in MCF-7 breast cancer cells BJMBR
Marchiori,A.C.; Casolari,D.A.; Nagai,M.A..
Most breast cancer risk factors are associated with prolonged exposure of the mammary gland to high levels of estrogens. The actions of estrogens are predominantly mediated by two receptors, ERα and ERβ, which act as transcription factors binding with high affinity to estrogen response elements in the promoter region of target genes. However, most target genes do not contain the consensus estrogen response elements, but rather degenerated palindromic sequences showing one or more mutations and other ER-binding sites such as AP-1 and SP-1. Using the differential display reverse transcription-polymerase chain reaction technique, our group identified several genes differentially expressed in normal tissue and in ER-positive and ER-negative primary breast...

Effect of exercise on the caloric intake of breast cancer patients undergoing treatment BJMBR
Battaglini,C.L.; Mihalik,J.P.; Bottaro,M.; Dennehy,C.; Petschauer,M.A.; Hairston,L.S.; Shields,E.W..
The purpose of this study was to examine the effects of an exercise intervention on the total caloric intake (TCI) of breast cancer patients undergoing treatment. A secondary purpose was to determine whether or not a relationship existed between changes in TCI, body fat composition (%BF), and fatigue during the study, which lasted 6 months. Twenty females recently diagnosed with breast cancer, scheduled to undergo chemotherapy or radiation, were assigned randomly to an experimental (N = 10) or control group (N = 10). Outcome measures included TCI (3-day food diary), %BF (skinfolds), and fatigue (revised Piper Fatigue Scale). Each exercise session was conducted as follows: initial cardiovascular activity (6-12 min), followed by stretching (5-10 min),...

Chromogenic in situ hybridization compared with other approaches to evaluate HER2/neu status in breast carcinomas BJMBR
Rosa,F.E.; Santos,R.M.; Rogatto,S.R.; Domingues,M.A.C..
Human epidermal growth factor receptor 2 (HER2) has been evaluated in breast cancer patients to identify those most likely to benefit from herceptin-targeted therapy. HER2 amplification, detected in 20-30% of invasive breast tumors, is associated with reduced survival and metastasis. The most frequently used technique for evaluating HER2 protein status as a routine procedure is immunohistochemistry (IHC). HER2 copy number alterations have also been evaluated by fluorescence in situ hybridization (FISH) in moderate immunoexpression (IHC 2+) cases. An alternative procedure to evaluate gene amplification is chromogenic in situhybridization (CISH), which has some advantages over FISH, including the correlation between HER2 status and morphological features....

Women cancer prevention and pharmaceutical contribution BJPS
Lourenço,Andrezza Viviany.
In Brazil, many cases of breast and cervical cancers are only diagnosed in advanced stages. Information on prevention of cancer in women is increasingly available. However, prevention or early treatment alternatives are often not practiced. This study investigated the issues hindering the practice of prevention against cancer in women. A qualitative method was employed in this exploratory and descriptive study. The sample included thirty-three randomly selected women undergoing treatment. The survey data was collected at the South Parana Institute of Oncology, Ponta Grossa - PR in September 2007 using a semi-structured individual interview after approval by the Research Ethics Committee of the Brazilian College of Systemic Studies - CBES, Curitiba - PR,...

Gd-GLU toward NMR imaging: synthesis, characterization and breast cell uptake assay BJPS
Campos,Tarcisio Passos Ribeiro de; Dalmazio,Ilza; Augusti,Rodiney; Almeida,Iassudara Garcia.
Breast cancer cell uptake of Gd-metal is investigated based on the formation of coordinate compounds of gadolinium and glucose (Glu) molecules in solution. The hypothesis is that glucose helps Gd-internalization by complex formations constituted of Gd3+ coordinate to m-glucose molecules, whose valence was complemented by Cl- anions. Such a proposal is an insight toward a metabolic-dependent contrast-agent for cancer and inflammation in magnetic resonance image. A solution was prepared based on anhydrous d-glucose and gadolinium chloride (Gd-Glu). Uptake assays for MDA-MB-231(c231) cells were elaborated collecting incubated c231-cells with Gd-Glu and measuring metal-uptake and their concentrations by Nuclear Activation Analysis (NAA). The ionic solution was...

The improvement of anti-HER2 scFv soluble expression in Escherichia coli BJPS
Farshdari,Farzaneh; Ahmadzadeh,Maryam; Nematollahi,Leila; Mohit,Elham.
The relationship between the expression of HER2 and malignity of breast tumors has led to the generation of antibodies targeting HER2+ tumors. In addition, the expression of scFvs, as the smallest antigen-binding region of antibody containing two disulfide bonds in Escherichia coli often results in accumulating non-functional protein in the cytoplasm. A redox-modified strain of E. coli such as Origami (DE3) may facilitate the formation of proper disulfide bond in cytoplasm. The present study aimed to optimize the expression of anti-HER2 scFv in Origami and evaluate the influence of induction temperature, and host strain on the solubility of the protein. To this aim, chemicallysynthesized anti-HER2 scFv of Trastuzumab was cloned in pET-22b (+). The results...
