Since there is no test that alone can predict sperm fertility, as more sperm parameters are analyzed during evaluation, more acurated will the result. Celulas meioticas de lirio, microsporocitos, tem sido amplamente utilizadas para estudos bioquimicos combinados. O sincronismo natural das celulas meioticas, a duracao arelativamente longa da profase (2 a 5 dias) e a habilidade das celulas desenvolverem-se in vitro possibilitou analisar cada intervalo da sintese de DNA, separadamente.

O Prosa Rural desta semana vai abordar o problema da resistência de lagartas à Soja-BT, aquela soja que expressa a proteína-inseticida em seu DNA, como forma de controle de algumas espécies de lagartas que atacam essa cultura. Apesar da área com resistência ainda ser pequena no Brasil, a Embrapa faz um alerta para que os produtores atentem para o manejo adequado dessas pragas, para evitar que o problema atinja mais áreas em pouco espaço de tempo.

Detection of oyster herpesvirus DNA and proteins in asymptomatic Crassostrea gigas adults ArchiMer

Since 1972, several herpes-like virus infections have been reported among different bivalve species around the world. Most of these reports involved larvae or juveniles presenting high mortalities. Two case reports of herpes-like viruses concerned adult oysters, Crassostrea virginica in USA and Ostrea angasi in Australia. Molecular techniques including PCR and in situ hybridization (ISH) have been recently developed to detect the oyster herpesvirus genome. In the present study, 30 Pacific oyster, Crassostrea gigas, adults have been analyzed using three different techniques: PCR, ISH and immunochemistry, in order to detect herpesviruses in asymptomatic individuals. Development of genomic probes for detection of hepatitis A virus and enteroviruses in shellfish ArchiMer

The development of molecular biology techniques has modified research procedures for certain viruses, particularly hepatitis A virus (HAV) and enteroviruses. Accordingly, we designed cDNA and RNA genomic probes to detect viruses in shellfish in the environment. Better sensitivity was noted with RNA than cDNA probes: only 17/83 shellfish samples were positive with HAV and enterovirus DNA probes, where as 42/83 were positive with HAV and enterovirus RNA probes.

A rare and unusual new bittiine genus with two new species from the South Pacific (Cerithiidae, Gastropoda) ArchiMer

A new genus, Limatium gen, n., and two new species, L. pagodula sp, n, and L. aureum sp, n. are described, found on outer slopes of barrier reefs and fringing reefs in the South Pacific. They are rare for cerithiids, which typically occur in large populations. The two new species arc represented by 108 specimens sampled over a period of 30 years, only 16 of which were collected alive. Three subadults from the Philippines and Vanuatu likely represent a third species. In addition to their rarity, Limatium species arc atypical for cerithiids in their smooth, polished, honey to golden brown shells with distinctive white fascioles extending suture to suture. Recent mass mortality outbreaks around the world in Pacific oysters, Crassostrea gigas, have seriously affected the aquaculture economy. Although the causes for these mortality outbreaks appear complex, infectious agents are involved. Two pathogens are associated with mass mortality outbreaks, the virus ostreid herpesvirus 1 (OsHV-1) and the bacterium Vibrio aestuarianus. Here we describe the interactions between these 2 pathogens and autophagy, a conserved intracellular pathway playing a key role in innate immunity. We show for the first time that autophagy pathway is present and functional in Pacific oysters and plays an important role to protect animals from infections. The scientific question addressed in this work is: what hides beneath first order kinetic constant k (s(-1)) measured for hybridization of a DNA target on a biosensor surface. Kinetics hybridization curves were established with a 27 MHz quartz microbalance (9 MHz, third harmonic) biosensor, constituted of a 20-base probe monolayer deposited on a gold covered quartz surface. Oligonucleotide quartz crystal microbalance sensor for the microalgae Alexandrium minutum (Dinophyceae) ArchiMer

We report the immobilization on a gold surface of a 20-base DNA probe labeled with disulfide group and on the selective hybridization with the complementary 20-base DNA strand. The oligonucleotide probe is the complementary strand of a partial sequence of the gene encoding for a large ribosomal RNA sub-unit which is a coding sequence of Alexandrium minutum DNA, a microalgae that produces neurotoxins responsible for paralytic shellfish poisoning on European and Asian coasts. Identification of skipjack tuna juveniles based on DNA control region sequences and potential spawning area around reunion island ArchiMer

In this study, we confirmed that DNA control region can be used to identify potential spawning area from pelagic juveniles species. We used the mitochondrial DNA control region to identify tuna juveniles predated by common dolphinfish and sampled in surface waters around Reunion Island. These were skipjack juveniles at an early stage (small length and young age). The biological results confirmed the species identification based on DNA analysis. Catch data by longline reveal a higher number of skipjack from October to December around Reunion Island corresponding to the period of our skipjack juvenile sampling

Antimicrobial histones and DNA traps in invertebrate immunity: evidences in Crassostrea gigas. ArchiMer

Although antimicrobial histones have been isolated from multiple metazoan species, their role in host defense has long remained unanswered. We found here that the hemocytes of the oyster Crassostrea gigas release antimicrobial H1-like and H5-like histones in response to tissue damage and infection. These antimicrobial histones were shown to be associated with extracellular DNA networks released by hemocytes, the circulating immune cells of invertebrates, in response to immune challenge. The hemocyte-released DNA was found to surround and entangle vibrios. This defense mechanism is reminiscent of the neutrophil extracellular traps (ETs) recently described in vertebrates. We report on a probe modification of a quartz crystal microbalance (QCM) DNA-biosensor that permits to reversibly change the DNA sequence detected. A QCM DNA-biosensor was designed by immobilization of a 20-base DNA-disulfide probe on the gold-covered quartz surface of a 27 MHz microbalance (9 MHz, third overtone). After immobilization on the gold covered quartz surface, this probe was modified by hybridization with a 45-base DNA that includes the complementary 20-base sequence, a 5-base spacer and a non-complementary 20-base sequence. Viral particles, associated with high mortality rates, have been reported among larvae and juveniles in different marine bivalve specie and characterized as member of the Hespesviridae family. Most studies focused on the detection of such viral infections among economically important species. However, the persistence of bivalve herpes viruses in marine environment is poorly documented. This topic appears as an important research domain in order to clarify the natural infection cycle of the herpesvirus of marine bivalves.

Morphoquantitative Analysis of the Intraepithelial Neoplasic Uterine Cells by Computer Image System International Journal of Morphology

The purpose of the present research was to detect the DNA variations by Feulgen stained chromatin and the packing degree of the cervical chromatin in the clinical cases of human uterine carcinomas. Scanning microphotometry was carried out on slides prepared from block Feulgen-stained. Accurate control procedures were performed using normal human cell nuclei. The cervical cells was morphometric studied using the Image Lab System for citologic classification. The overall diagnostic accuracy for neoplasia type I (CIN I) and type II (CIN II), demonstrated by stain interpetration values of a sensitive detection of DNA neuroploidy and a significant difference between the two groups in scanning static cytophotometry.
