Genetic differentiation in Gammarus fossarum and G. caparti (Crustacea, Amphipoda) with reference to G. pulex pulex in northwestern Europe Naturalis
Scheepmaker, Maarten; Dalfsen, Jan van.
Genetic differentiation among G. fossarum Koch, 1835 from different stations in Germany, Switzerland, Belgium and northern France, and the closely related Belgian form G. caparti Pètre-Stroobants, 1980 was investigated by electrophoresis at 20 enzyme loci. Although morphologically variable, geographically distant populations of G. cf. fossarum were hitherto considered conspecific. In the present study, populations of G. cf. fossarum and G. caparti were examined with reference to G. pulex pulex as an estimate for genetic differentiation at the species level. With G. p. pulex as a standard, genetic differentiation among geographically distant populations of G. cf. fossarum is occasionally observed at species level. The populations of G. caparti studied were...

Isoenzymatic polymorphism in the leaf-cutting ant Atta capiguara Gonçalves (hymenoptera: formicidae) Neotropical Entomology
Cantagalli,Liriana B; Mangolin,Claudete A; Ruvolo-Takasusuki,Maria C C.
This study was carried out to analyze the genetic population structure of Atta capiguara from 12 nests collected in Tapejara in the state of Paraná, Brazil, using isoenzyme polymorphisms. The analyzed isoenzymes were esterases (EST - EC 3.1.1.1), acid phosphatase (ACP - EC 3.1.3.2) and carbonic anhydrase (CA - EC 4.2.1.1). Ten loci were found in A.capiguara and four polymorphic loci were detected. The observed heterozigosity (0.0296) was low when compared to the expected heterozigosity (0.1461). The high value of F IS (0.7954) shows an excess of homozygous genotypes probably caused by inbreeding.

RAPD variation within and among species of Melittobia Westwood (Hymenoptera: Eulophidae) Neotropical Entomology
Sari,Eloisa H.R.; Tavares,Mara G.; Campos,Lucio A.O..
The genus Melittobia Westwood comprises several species of microparasitoids and only two of them are know to occur in Brazil up to now: M. australica Girault and M. hawaiiensis Perkins. Nevertheless, the differentiation between these two species using traditional taxonomy is very difficult. In the present study, we used random amplified polymorphic DNA chain reaction (RAPD-PCR) to test for its ability to discriminate between these two species and to examine the genetic variation among the studied populations of M. australica. Most of the generated fragments were species-specific, occurring in all individuals of one species and absent in the individuals of the other species demonstrating the appropriateness of such technique to distinguish between both of...

Genetic similarity among Agistemus pallinii Matioli et al (Acari: Stigmaeidae) found in citrus orchards in Viçosa, Minas Gerais State, Brazil Neotropical Entomology
Matioli,André L; Pallini,Angelo; Tavares,Mara G.
Stigmaeidae are very important predators of mite and insect pests on several crops in Brazil. It is considered the second most important family of predatory mites in citrus orchards in Brazil. However, their identification, especially that of the members of the genus Agistemus, is rather difficult based only on morphology. Hence, this study describes the use of random amplified polymorphic DNA (RAPD-PCR) markers to determine the genetic similarity of an Agistemus pallinii Matioli et al population found in 2004 in a citrus orchard in Viçosa, Minas Gerais State, Brazil, preying on Panonychus citri (McGregor). Amplifi cations were performed with 12 random primers (OPAA8, OPAA19, OPAB1, OPAB5, OPAB18, OPAC9, OPAC17, OPAC19, OPAD10, OPAE9, OPAE12 and OPAE17),...

Mitochondrial DNA divergence between wild and laboratory populations of Anopheles albimanus Wiedemann (Diptera: Culicidae) Neotropical Entomology
Arias,Lida; Bejarano,Eduar E.; Márquez,Edna; Moncada,John; Vélez,Iván; Uribe,Sandra.
Studies of insect vectors may be facilitated by using laboratory colonies. However, it has been suggested that the colony insects are not representative of natural populations, sometimes yealding to erroneous interpretations of the intraspecific genetic variation between the individuals. In the present study the variability of the mitochondrial gene cytochrome b was evaluated among a closed laboratory colony of Anopheles albimanus that was founded 20 years ago and the field population from which it was derived. The analyses revealed the presence of five and three nucleotide haplotypes in the wild and colony populations, respectively. Wild individuals presented greater variability than those of the colony based on the number of polymorphic sites, haplotype...

Characterization and genetic relationships among Brazilian biotypes of Schizaphis graminum (Rondani) (Hemiptera: Aphididae) using RAPD markers Neotropical Entomology
Lopes-da-Silva,Marcelo; Tonet,Gabriela E.L.; Vieira,Luiz G.E..
The greenbug Schizaphis graminum (Rondani) is one of the most important cereals pests in the world. Within populations of this species, several biotypes, which are clones that share same virulence relationships with cultivated plants, can be distinguished. Molecular markers have been used to genetically characterize insect populations because they are fast and cost effective. In order to find RAPD markers to identify Brazilian S. graminum biotypes, nineteen clonal populations of three biotypes (B, C and E) from Brazil and three clonal populations from the U.S. were examined. Eighteen primers were used to analyze the material, but only six primers revealed polymorphisms and among those, three produced diagnostic band profiles that allowed biotype...

Bernardo,Karla Annielle da Silva; Freire Filho,Francisco Rodrigues; Ribeiro,Valdenir Queiroz; Vieira,Paulo Fernando de Melo Jorge; Lopes,Ângela Celis de Almeida; Oliveira,Rosana Mendes de Moura.
Abstract: The objective of this work was to obtain cowpea (Vigna unguiculata) progenies with compound inflorescences, short peduncles, early cycle, good plant architecture, and high-yielding potential. The initial genetic material consisted of 453 F2:3 progenies, selected from ten crosses and evaluated using Federer’s augmented block design, with two controls. Of these, 60 progenies from the F4:5 and F4:6 generations were selected and assessed in two consecutive experiments, both in a 8x8 lattice design with four controls and four replicates. Early selection was efficient at fixing the evaluated traits. A wide genetic variability was detected among and within progenies. The estimated and obtained heritability and genetic gains in the F4:5 and F4:6...

Carvalho,Jane Fiuza Rodrigues Portela de; Robinson,Ingrid Peters; Alfenas,Acelino Couto.
The objectives of this work were to optimize the isozyme electrophoresis technique for Bixa orellana, and use isozyme markers for a preliminary survey on the genetic variability in Brazilian annatto germplasm accessions. Collection consisted of seed samples from sixty open pollinated trees, representing two Northern and four Southern geographic provenances. The extraction, electrophoresis, and interpretation of annatto isozymes are described. Three out of the twenty-one identified isozyme loci were polymorphic in the collection. The percentage of polymorphic loci (P = 21.05) and the expected heterozygosity in annatto (H T = 0.064) were low, compared to other tropical woody species. A UPGMA phenogram, constructed with Nei's genetic distances, clearly...

Transgene inheritances and genetic similarities of near isogenic lines of genetically modified common beans PAB
Pinheiro,Patrícia Valle; Faria,Josias Corrêa de; Nogueira,Elsa Oliveira Paranaguá e Lago; Aragão,Francisco José Lima.
The objective of the present work was to determine the inheritance and stability of transgenes of a transgenic bean line expressing the genes rep-trap-ren from Bean golden mosaic virus and the bar gene. Crosses were done between the transgenic line and four commercial bean cultivars, followed by four backcrosses to the commercial cultivars. Progenies from each cross were evaluated for the presence of the transgenes by brushing the leaves with glufosinate ammonium and by polymerase chain reaction using specific oligonucleotides. Advanced generations were rub-inoculated with an isolate of Bean common mosaic necrosis virus (BCMNV). The transgenes were inherited consistently in a Mendelian pattern in the four crosses studied. The analyzed lines recovered close...

Genetic Dissimilarity in Conyza sumatrensis Revealed by Simple Sequence Repeat (SSR) Markers Planta Daninha
SCHNEIDER,T.; RIZZARDI,M.A.; BRAMMER,S.P.; SCHEFFER-BASSO,S.M.; NUNES,A.L..
ABSTRACT: In view of the rapid evolution of Conyza sumatrensis populations resistant to glyphosate, it is necessary to understand the genetic diversity aimed to improve strategies for managing this weed. We investigated the genetic dissimilarity among 15 biotypes of C. sumatrensis from different geographic regions using microsatellite loci. The biotypes, were cultivated in a greenhouse to obtain vegetal material for DNA extraction. Nineteen microsatellite markers (SSR), were developed for C. sumatrensis biotypes. The genetic dissimilarity was estimated by the Jaccard coefficient (JC) and the biotypes grouped by the UPGMA method. The results demonstrated a high dissimilarity (JC = 7.14 to 82.62) of the analyzed material, with the biotypes forming five...
