Detection of siderophores in endophytic bacteria Methylobacterium spp. associated with Xylella fastidiosa subsp. pauca PAB
Lacava,Paulo Teixeira; Silva-Stenico,Maria Estela; Araújo,Welington Luiz; Simionato,Ana Valéria Colnaghi; Carrilho,Emanuel; Tsai,Siu Mui; Azevedo,João Lúcio.
The objective of this work was to study the production of siderophores by endophytic bacteria Methylobacterium spp., which occupy the same ecological niche as Xylella fastidiosa subsp. pauca (Xfp) in citrus plants. The siderophore production of Methylobacterium strains was tested according to chromeazurol agar assay test (CAS), Csáky test (hydroxamate-type) and Arnow test (catechol-type). In addition, the ability of Xfp to use siderophores, in vitro, produced by endophytic bacteria as source of iron, was evaluated. All 37 strains of Methylobacterium spp. tested were CAS-positive for siderophore production. Methylobacterium spp. produced hydroxamate-type, but not catechol-type siderophores. In vitro growth of Xfp was stimulated by the presence of...

BIOFORTIFICATION, AN ALTERNATIVE TO REDUCE FOOD INSECURITY AND ITS ADVERSE EFFECTS IN DEVELOPING COUNTRIES: A HONDURAS CASE STUDY AgEcon
Perez Suarez, Salomon.
Food insecurity can generate nutritional problems such as iron deficiency anemia, which in countries like Honduras is a moderate public health problem. The AgroSalud project is enhancing the iron content of beans and rice in order to improve the iron intake of vulnerable groups, especially children under five years and pregnant women. Iron- biofortified rice and beans together can reduce the deficit in iron intake by more than a 100% depending on the age group, farmer adoption and other variables. This, in turn, would increase the number of healthy (disability-adjusted life years, DALYs) years that the Honduran population could live.

THE IMPACT OF MICRONUTRIENTS ON LABOR PRODUCTIVITY: EVIDENCE FROM RURAL INDIA AgEcon
Weinberger, Katinka.
Micronutrient malnutrition, particularly of iron, is widespread on the South Asian subcontinent and economic losses due to reduced productivity of afflicted population groups are believed to be substantial. This paper examines the impact of micronutrient intake on productivity of house-hold engaged in agricultural labor in India by applying a two-stage least squares (2SLS) estimation technique. The micronutrient under investigation is iron. The results indicate that productivity, measured in wages is indeed affected through insufficient intake, and that wages would on average be 5 to 17.3 percent higher, if households would achieve recommended intake levels of the micronutrient discussed here. The results demonstrate that policy interventions that aim at...

Iron Uptake by Fungi Isolated from Arcelor Mittal -Annaba- in the Northeast of Algeria Rev. Bras. Ciênc. Avic.
Bourzama,G; Ouled-Haddar,H; Marrouche,M; Aliouat,A.
ABSTRACT The Metal pollution is one of the major risks in the world nowadays. Iron is an essential metal for growth and proliferation of a vast majority of organisms, but it can be toxic to human health and other living beings in the environment at high concentrations due to its increased industrial activity. Fungi have a remarkable capacity to uptake and detoxify iron metal using different mechanisms such as bioaccumulation. Thus, the aim of this work is to study the ability of iron uptake by the fungal strains isolated from Arcelor Mittal -Annaba- in Algeria. Three strains were screened at high concentration of iron (1 g/l) and their capacity to uptake iron has been studied on Czapek Yeast Agar medium. The amounts of uptaken iron ions were estimated in...

Assessment of micronutrient extractants from soils of Paraná, Brazil Rev. Bras. Ciênc. Solo
Sarto,Marcos Vinicius Mansano; Steiner,Fábio; Lana,Maria do Carmo.
In Brazil, plant-available micronutrients in the soil can be determined by several chemical extractants, the most common of which are dilute acid and chelating solutions. The purpose of this study was to assess the extractants 0.1 mol L-1 HCl, Mehlich-1, Mehlich-3 and DTPA for analysis of the micronutrients Cu, Zn, Fe, and Mn in soils from the state of Paraná. In samples from 12 soils (0-20 cm layer), wheat was planted (Triticum aestivum), grown for 42 days after emergence, and then bean (Phaseolus vulgaris) for 38 days. At the end of each planting period, the soil was sampled again. All extractants tested to assess the availability of Cu, Zn, Fe, and Mn correlated with each other. The efficiency of the extractants HCl, Mehlich-3 and DTPA in assessing...

Azospirillum lipoferum and nitrogen fertilization effect on chlorophyll content, nutrients uptake and biometric properties of Zea mays L. Agrociencia
Davaran-Hagh,Elnaz; Mirshekari,Bahram; Reza-Ardakani,Mohammad; Farahvash,Farhad; Rejali,Farhad.
Integrated nutrient management is a major agronomic practice which brings together all methods to support plants nutritional requirement. However, the interaction of these methods together is very complex and needs to be studied in different climatic conditions. So, the effect of nitrogen fertilizer and Azospirillum lipoferum inoculation on maize (Zea mays L.) growth, yield and nutrients uptake was determined in this study. The hypothesis was that the bacterial inoculation decreases the need for N fertilizer application without reducing plant yield. Therefore, we carried out a factorial experiment in a randomized complete block design with three replications. Nitrogen at 0, 25, 50, 75 and 100 % concentrations of the recommended dose, i.e. 150 kg N ha-1...

Hepcidin is a useful biomarker to evaluate hyperferritinemia associated with metabolic syndrome Anais da ABC (AABC)
RAUBER,MARIANA R.; PILGER,DIOGO A.; CECCONELLO,DAIANE K.; FALCETTA,FREDERICO S.; MARCONDES,NATÁLIA A.; FAULHABER,GUSTAVO A.M..
Abstract: Investigation of hyperferritinemia in metabolic syndrome patients represents a diagnostic challenge, but it is essential for the identification of individuals with iron overload. Hepcidin negatively regulates iron absorption and release. An increase in hepcidin occurs when iron levels are sufficient or in inflammatory states, conditions often associated with hyperferritinemia. Hemochromatosis causes hyperferritinemia due to iron overload, but frequently has low hepcidin levels. Our aim was to evaluate biochemical and molecular parameters related to iron metabolism in patients with metabolic syndrome. We evaluated 94 patients with metabolic syndrome according to the International Diabetes Federation criteria in a cross-sectional study....

Mitochondrial DNA damage associated with lipid peroxidation of the mitochondrial membrane induced by Fe2+-citrate Anais da ABC (AABC)
Almeida,Andréa M.; Bertoncini,Clélia R.A.; Borecký,Jiri; Souza-Pinto,Nadja C.; Vercesi,Aníbal E..
Iron imbalance/accumulation has been implicated in oxidative injury associated with many degenerative diseases such as hereditary hemochromatosis, beta-thalassemia, and Friedreich's ataxia. Mitochondria are particularly sensitive to iron-induced oxidative stress - high loads of iron cause extensive lipid peroxidation and membrane permeabilization in isolated mitochondria. Here we detected and characterized mitochondrial DNA damage in isolated rat liver mitochondria exposed to a Fe2+-citrate complex, a small molecular weight complex. Intense DNA fragmentation was induced after the incubation of mitochondria with the iron complex. The detection of 3' phosphoglycolate ends at the mtDNA strand breaks by a 32P-postlabeling assay, suggested the involvement of...

Iron transportation across the placenta Anais da ABC (AABC)
Oliveira,Claudia M. de; Rodrigues,Marcio N.; Miglino,Maria Angélica.
According to the classification of placental types among animals, the transfer of iron through the placenta can occur via: absorption connected to transferin through the outer surface of the trophoblast in direct contact with circulating maternal blood; absorption of the erythrocytes by the chorionic epithelium in direct contact with accumulation of blood extravased from haemotophagous areas; absorption by the chorionic epithelium in direct contact with iron enriched secretions from the endometrial glands and absorption by extravasations of the blood in the maternal-fetal surface and the subsequent phagocytosis of the erythrocytes by trophoblast cells described in bovine, small ruminants, canine and feline. Iron restriction increases myoglobin gene and protein expression in Soleus muscle of rats Anais da ABC (AABC)
SOUZA,JANAINA S. DE; BRUNETTO,ERIKA L.; NUNES,MARIA TEREZA.
ABSTRACT Iron is an important trace element for proper cell functioning. It is present in cytochromes, hemoglobin and myoglobin (Mb), where it binds to oxygen. It is also an electron acceptor in the respiratory chain. Mb is an 18 kDa heme-protein, highly expressed in skeletal muscle and heart. The expression of several genes involved in the metabolism of iron is post-transcriptionally regulated by this element. Iron was shown to interfere with the polyadenylation step, modifying their poly (A) tail length and, as a consequence, their stability and translation rate. The aim of this study was to investigate whether iron supplementation or long and short-term restriction affects Mb gene and protein expression, as well as Mb mRNA poly(A) tail length, in...

Green speciation of iron using aqueous two-phase system Anais da ABC (AABC)
CAMPOS,RAQUEL A.; PATRÍCIO,PAMELA R.; VARGAS,SILVIA JULIANA R.; SILVA,LUIS HENRIQUE M. DA; HESPANHOL,MARIA C..
Abstract Fe(II) and Fe(III) have distinct chemical and biological functions. Consequently, it is more important to determine the fraction of both oxidation state that knowing the total iron concentration in a sample. However, green methods for iron speciation are still limited. This work uses aqueous two-phase system, a safe alternative to liquid–liquid extraction, to perform the chemical speciation of iron. This method is based on the reaction of Fe(II) with 1,10-phenanthroline extractant, forming a complex of Fe(II)-phenanthroline that concentrates in the top phase of the system. The Fe(III) specie concentrated in the bottom phase of the system. Iron speciation was affected by the electrolyte nature, macromolecule type, quantity of phenanthroline added,...

Zinc in the prevention of Fe2initiated lipid and protein oxidation Biol. Res.
ZAGO,M. PAOLA; VERSTRAETEN,SANDRA V.; OTEIZA,PATRICIA I..
In the present study we characterized the capacity of zinc to protect lipids and proteins from Fe2+-initiated oxidative damage. The effects of zinc on lipid oxidation were investigated in liposomes composed of brain phosphatidylcholine (PC) and phosphatidylserine (PS) at a molar relationship of 60:40 (PC:PS, 60:40). Lipid oxidation was evaluated as the oxidation of cis-parinaric acid or as the formation of 2-thiobarbituric acid-reactive substances (TBARS). Zinc protected liposomes from Fe2+ (2.5-50 muM)-supported lipid oxidation. However, zinc (50 muM) did not prevent the oxidative inactivation of glutamine synthelase and glucose 6-phosphate dehydrogenase when rat brain superntants were oxidized in the presence of 5 muM Fe2+ and 0.5 mM H2O2 .We also...

Molecular and pathological basis of aceruloplasminemia Biol. Res.
KONO,SATOSHI; MIYAJIMA,HIROAKI.
Aceruloplasminemia is an autosomal recessive neurodegenerative disease characterized by iron accumulation in the brain as well as visceral organs. It is a loss-of-function disorder caused by mutations in the ceruloplasmin gene. Clinically, this disease consists of the triad of adult-onset neurological disease, retinal degeneration and diabetes mellitus. Massive iron accumulation and extensive loss of neurons are observed in the basal ganglia. The elevated iron concentration is associated with increased lipid peroxidation in the brains of aceruloplasminemia patients. Enlarged or deformed astrocytes and spheroid-like globular structures are characteristic neuropathological findings in aceruloplasminemia. Moreover, deformed astrocytes and globular structures...

Effect of iron on the activation of the MAPK/ERK pathway in PC12 neuroblastoma cells Biol. Res.
MUÑOZ,PABLO; ZAVALA,GABRIELA; CASTILLO,KAREN; AGUIRRE,PABLA; HIDALGO,CECILIA; NÚÑEZ,MARCO T.
Recent evidence suggests that reactive oxygen species function as second messenger molecules in normal physiological processes. For example, activation of N-Methyl-D-Aspartate receptor results in the production of ROS, which appears to be critical for synaptic plasticity, one of the cellular mechanisms that underlie learning and memory. In this work, we studied the effect of iron in the activation of MAPK/ERK pathway and on Ca2+ signaling in neuronal PC12 cells. We found that iron-dependent generation of hydroxyl radicals is likely to modulate Ca2+ signaling through RyR calcium channel activation, which, in turn, activates the MAPK/ERK pathway. These findings underline the relevance of iron in normal neuronal function.

Hereditary hemochromatosis: An opportunity for gene therapy Biol. Res.
EZQUER,FERNANDO; NÚÑEZ,MARCO T; ROJAS,ALEJANDRO; ASENJO,JUAN; ISRAEL,YEDY.
Levels of body iron should be tightly controlled to prevent the formation of oxygen radicals, lipoperoxidation, genotoxicity, and the production of cytotoxic cytokines, which result in damage to a number of organs. Enterocytes in the intestinal villae are involved in the apical uptake of iron from the intestinal lumen; iron is further exported from the cells into the circulation. The apical divalent metal transporter-1 (DMT1) transports ferrous iron from the lumen into the cells, while the basolateral transporter ferroportin extrudes iron from the enterocytes into the circulation. Patients with hereditary hemochromatosis display an accelerated transepithelial uptake of iron, which leads to body iron accumulation that results in cirrhosis, hepatocellular...
