Genetic consequences of production of Pacific oyster larval in hatchery: drift and selective pressures related to rearing practices. In order to study the genetic consequences of production of oyster larvae in hatcheries, two factors were examined: the effects of discarding the smallest larvae (i.e. culling) and temperature effects. A mixed-family approach was used in order to infer the genetic composition of the larval population. The results show that high polymorphic microsatellite-based family assignment is a powerful tool for the study of bivalve larvae genetics. Impact of temperature on larval development and evolution of physiological indices in Crassostrea gigas ArchiMer

The effect of four temperatures, ranging between 17 degrees C and 32 degrees C, was studied on development and lipid reserve management of Crassostrea gigas larvae. No effect of temperature was found on larval mortality, as high survival (>90%) was recorded before competence at all temperatures studied. Temperature did, nonetheless, have a strong effect on growth and settlement success. At low temperature (17 degrees C), larvae competent to metamorphose were only observed from day 23 and only a low percentage finally achieved metamorphosis (12%). La conchyliculture, et principalement l’élevage de l’huître creuse, Crassostrea gigas, constitue la principale activité aquacole française. Activity of digestive enzymes (trypsin, chymotrypsin and amylase) was studied in relation with food in Penaeus vannamei larvae first stages. Microparticles containing casein involved the decrease of soluble protein content and enzyme activities. Specific trypsin activity but not chymotrypsin was correlated (p<1%) to casein level between 10 and 60% in microparticles. Cellulose fibres in food seemed to have little effect on digestive enzymes. Starch, between 1 and 20% in microparticles, had no influence on specific amylase activity. The effect of different feeding conditions were also tested on growth. Effects of a remarkably high overall lipid Tisochrysis lutea strain (T+) upon gross biochemical composition, fatty acid (FA), sterol and lipid class composition of Crassostrea gigas larvae were evaluated and compared with a normal strain of Tisochrysis lutea (T) and the diatom Chaetoceros neogracile (Cg). Detection of ostreid herpesvirus-1 (OsHV-1) by PCR using a rapid and simple method of DNA extraction from oyster larvae ArchiMer

A DNA extraction procedure was developed for the detection of ostreid herpesvirus-1 (OsHV-1) using the polymerase chain reaction (PCR) in oyster larvae. The DNA extraction procedure developed was tested on 8 larval samples. Abnormal nuclei with characteristic features associated with OsHV-1 infections were only observed in samples in which the viral DNA was detected by PCR. A previously described competitive PCR method was applied to detect inhibition during PCR reactions. The results show that the method can be used on small amounts of oyster larvae (3 mg) for the detection of OsHV-1 DNA by PCR.

Scallop Pecten maximus larvae have been cultured at the Argenton and Tinduff (Brittany, France) hatcheries with antibiotic treatment (chloramphenicol at 8 ppm) for 15 yr. Without treatment, outbreak of disease has normally occurred between Day 12 and Day 19 or sometimes earlier. A bacteriological study of larvae reared with and without antibiotic was performed over a 4 yr period. The application of a simple, fast and reliable volumetric microrespirometric method to assess respiration of bivalve larvae is discussed. As a model, C. gigas larvae of various sizes were used. Metabolic activity of veliger larvae was assessed by measuring respiratory rate for use in ecophysiological modelling. The want of C. gigas spat-catching in the Marennes-oleron basin in 1981 can be due to various different reasons which are analysed here. Physico -chemical and trophic factors that can induce variations in the quality of gametogenesis, are successively in comparison with the year 1979 when reproduction was good. The biochemical evolution and a study of gonads slow a lack in maturation in 1981 even during the summer period. Late maturation of gonads bring about physiological consequences which induced mortality of cultivated oysters at the end of winter. A feeble recruit and a decrease of the cultivated stock have economic consequences which here sketched up.

The events occurring between artificial induction of spawning and normal fertilization were studied experimentally in order to explain this difference. Fertilization and rearing trials were performed on oocytes obtained from the gonads of both species by scarification, by aspiration from genital ducts (of P. maximus only) and after spawning. Every batch of oocytes was fixed for histological study.
