Introduction: Through the combined works of research, local communities and professionals, a new activity has been created, "vénériculture", i.e. clam farming. Thanks to the knowledge acquired in research as well as production in the shellfish field, clam production results followed shortly thereafter. Rightly so, only 10 years have elapsed since the beginning of the research and the actual production phase (500 t) involving about 300 professionals. Research conducted by IFREMER positively contributes to the creation of new trades as well as diversity within existing professions. Clams are marketed nationally as well as exported. A condition in Sparus aurata larvae termed "distended gut syndrome" (DGS) has become a major problem in the past three seasons. Unable to digest rotifers and brine shrimp nauplii, the larvae develop a swollen abdomen, exhibit a disoriented spinning motion and are swept passively with the current. Bacterial or viral infections were suspected. Chloramphenicol-supplemented larval food did not affect the prevalence of DGS. Virus-like particles 80 nm in diameter were observed in necrotic cytoplasm of midgut wall cells. |

The thesis work is part of the very current issue on mass mortality outbreaks affecting Pacific oyster, C. gigas, spat and juveniles and questions about the involvement of pesticides in this phenomenon. The first part of this thesis was devoted to study the effects of pesticides on hemocyte parameters in the Pacific oyster (in vitro and in vivo). The immunomodulator effect of selected pesticides (alone or in mixture) has been explored principally through flow cytometry. The second part concerned the study of the effects of a pesticides mixture on OsHV-1 itself. No direct effects have been reported on the viral particles in presence of the pollutants. Created in 1992, the REPAMO network (Network of mollusc pathology), is a national zoosanitary surveillance network of shellfish health status along the French coasts. Its activities are in keeping with two European Directives, 91/67/EEC and 95/70/EC and are a part of the institutional tasks of Ifremer. The aims of the network are to prevent the introduction and spread of exotic pathogens and to survey the evolution ofnotifiable pathogens already present in France. The network focused on the survey of listed diseases (bonamiosis and marteiliosis of flat oyster) in two areas under agreement process (zone X and Granville bed in zone IX). The main objective of this project was to develop the tools for the in-depth understanding of mollusc biology and pathology. Such a goal has been reached in other biological systems through the development of molecular biology and cell culture. Indeed, development of a mollusc cell culture system is an essential step to approach many of the problems related to mollusc pathology, in particular to use molecular biology tools to study viral infection of these organisms. The flat oyster Ostrea edulis was originally reared in several regions of the European waters. The tidal saline ponds on the Atlantic coasts of France were highly productive before being hit by two consecutive epizooties. Parasitic diseases were described on this species, both of them leading 10 economic losses, as heavy mortalities have usually been recorded before the oysters reach the commercial size. Repeated experiments showed that the two diseases, Marteilia refringens and Bonamia ostreae are still active in this environment. The originality of this project consists in the interdisciplinary approach gathering the impact of toxic phytoplankton blooms and metallic pollution on health (behaviour, stress, immunity, genetic) and disease development of the Manila clam, an economieally important species. The Manila clam Ruditapes philippinarum is the second exploited bivalve in the world. It was introduced in Arcachon bay during the 80's and is today exploited by professional fishermen (57 licences). We describe an emerging pathology, brown muscle disease (BMD), which specifically affects the Manila clam Ruditapes philippinarum in Arcachon Bay (France). BMD induces a transformation of the posterior adductor muscle, which becomes infused by conchiolin and calcified, reducing the ability of clams to bury. The disease affects both types of muscular tissue, with striated muscle becoming affected to a higher degree than smooth muscle. The symptom which characterizes the brown ring disease (BRD) in Manilla clam Ruditapes philippinarum has been observed in other Bivalvia species (Veneridae and Pectinidae). Vibrio P1, responsible of the BRD in R. philippinarum , has been detected in other Veneridae which were carrying the symptom. In mussel and oysters which are cultivated in Europe, the symptom has never been reported and Vibrio P1 has not been detected. Experimental contaminations of bivalves of commercial interest with the pathogen showed a clear Vibrio P1 sensitivity of European clam R. decussatus ; this latter seems however to be less sensitive than the Manilla clam. |
