SHARMA, R. D..

A first genetic linkage map of the European flat oyster, Ostrea edulis : towards the identification of Bonamia resistance QTLs ArchiMer
Lallias, Delphine; Lapegue, Sylvie; Haley, Chris; Heurtebise, Serge; Boudry, Pierre; Beaumont, Andy.
The flat oyster Ostrea edulis is an endemic European oyster species, found on both Atlantic and Mediterranean coasts. Its aquacultural production has dramatically decreased due to two successive diseases caused by the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer has undertaken a breeding program to produce oyster families which are tolerant to Bonamia. In this context, the next step is to identify QTLs for resistance to this parasite. A genetic map is therefore being established as a basis for QTL work. The reference family is an F2 family produced by single pair mating of two oysters from the same F1 family. The F1 family was itself produced from a bi-parental cross between a wild oyster and an individual from the...

Selective breeding to improve resistance against summer mortality in the Pacific oyster Crassostrea gigas : results after 3 generations ArchiMer
Boudry, Pierre; Degremont, Lionel; Bedier, Edouard; Samain, Jean-francois.
Summer mortality of adults and juveniles has been reported in the Pacific oyster, Crassostrea gigas, for many years and in several countries. The French multidisciplinary program "Morest" aims to investigate the causes of the summer mortality in Crassostrea gigas. Within this program, we designed multi-site field experiments to assess to what extent genetic variability exists for summer mortality in French populations of C. gigas and to determine whether selective breeding could improve survival.

Compatibility Polymorphism Based on Long-Term Host-Parasite Relationships: Cross Talking Between Biomphalaria glabrata and the Trematode Schistosoma mansoni From Endemic Areas in Brazil ArchiMer
Lima, Mariana G.; Montresor, Lângia C.; Pontes, Joana; Augusto, Ronaldo De C.; Silva, Jairo Pinheiro Da; Thiengo, Silvana C..
Sympatric snail populations have been kept in the laboratory since the isolation of the parasite from the field. To evaluate the influence of the intermediate host in the infectivity of S. mansoni, this allopatric strain was compared to two sympatric strains, from different geographical origins, and with different time of maintenance in the laboratory. Snail–trematode compatibility was accessed for a total of nine possible combinations (three snail populations, three schistosome strains), using different charges of parasite: 1, 5, 10, and 15 miracidia/snail. Each S. mansoni strain was characterized according to its infectivity phenotype that reflects the efficiency of their infection mechanism and all B. glabrata populations were characterized according to...

Greater Mitochondrial Energy Production Provides Resistance to Ocean Acidification in “Winning” Hermatypic Corals ArchiMer
Agostini, Sylvain; Houlbrèque, Fanny; Biscéré, Tom; Harvey, Ben P.; Heitzman, Joshua M.; Takimoto, Risa; Yamazaki, Wataru; Milazzo, Marco; Rodolfo-metalpa, Riccardo.
Coral communities around the world are projected to be negatively affected by ocean acidification. Not all coral species will respond in the same manner to rising CO2 levels. Evidence from naturally acidified areas such as CO2 seeps have shown that although a few species are resistant to elevated CO2, most lack sufficient resistance resulting in their decline. This has led to the simple grouping of coral species into “winners” and “losers,” but the physiological traits supporting this ecological assessment are yet to be fully understood. Here using CO2 seeps, in two biogeographically distinct regions, we investigated whether physiological traits related to energy production [mitochondrial electron transport systems (ETSAs) activities] and biomass (protein...

High temperature induces transcriptomic changes in Crassostrea gigas that hinder progress of ostreid herpesvirus (OsHV-1) and promote survival ArchiMer
Delisle, Lizenn; Pauletto, Marianna; Vidal-dupiol, Jeremie; Petton, Bruno; Bargelloni, Luca; Montagnani, Caroline; Pernet, Fabrice; Corporeau, Charlotte; Fleury, Elodie.
Of all environmental factors, seawater temperature plays a decisive role in triggering marine diseases. Like fever in vertebrates, high seawater temperature could modulate the host response to pathogens in ectothermic animals. In France, massive mortality of Pacific oysters, Crassostrea gigas, caused by the ostreid herpesvirus 1 (OsHV-1) is markedly reduced when temperatures exceed 24°C in the field. In the present study we assess how high temperature influences the host response to the pathogen by comparing transcriptomes (RNA sequencing) during the course of experimental infection at 21°C (reference) and 29°C. We show that high temperature induced host physiological processes that are unfavorable to the viral infection. Temperature influenced the...

Modélisation de la résistance aux chocs de salinité des postlarves de Penaeus vannamei ArchiMer
Aquacop, Aquacop; Le Moullac, Gilles; Damez, D.
The notion of penaeid shrimp postlarvae quality, which implies a survival potential, is still a subjective parameter. The zootechnical criteria and the visual observations are not sufficient to evaluate the physiological condition of the hatchery reared postlarvae. The resistance of postlarvae to salinity shocks is a representative criteria of their condition. The goal of this study is to present a non-linear regression model at 2 variables (size, salinity) for the P. vannamei postlarvae resistance to salinity shocks. Between the second and the twentieth day after metamorphosis; 5 salinity shocks (20, 15, 10, 5 and 20/00) were tested daily, each one with 50 postlarvae. Survival rates were recorded after 2 hrs. Concurrently, the postlarvae growth in the...

Etude des interactions hôte/parasite chez l'huître plate Ostrea edulis et son parasite Bonamia ostreae ArchiMer
Morga, Benjamin.
The history of the French oyster production highlights the fragility of this production against overexploitation and disease outbreaks. In particular, the production of flot oyster, Ostrea edulis, has decreased following the emergence of two parasitic diseases including bonamiosis. The means to fight against bonamiosis are relatively limited. They are mainly based on oyster health surveillance to limit the spread of the disease. However, the use of predictive models of disease progression in infected area would help to improve stock management and minimize the impact pathogens. Moreover the development of resistant animais could help to revive this production. These different approaches require appropriate diagnostic tools, a good knowledge of the life...

Symbiodinium clades A and D differentially predispose Acropora cytherea to disease and Vibrio spp. colonization ArchiMer
Rouze, Heloise; Lecellier, Gael; Saulnier, Denis; Berteaux-lecellier, Veronique.
Coral disease outbreaks have increased over the last three decades, but their causal agents remain mostly unclear (e.g., bacteria, viruses, fungi, protists). This study details a 14-month-long survey of coral colonies in which observations of the development of disease was observed in nearly half of the sampled colonies. A bimonthly qPCR method was used to quantitatively and qualitatively evaluate Symbiodinium assemblages of tagged colonies, and to detect the presence of Vibrio spp. Firstly, our data showed that predisposition to disease development in general, and, more specifically, infection by Vibrio spp. in Acropora cytherea depended on which clades of Symbiodinium were harbored. In both cases, harboring clade D rather than A was beneficial to the...

Contribution of in Vivo Experimental Challenges to Understanding Flat Oyster Ostrea edulis Resistance to Bonamia ostreae ArchiMer
Morga, Benjamin; Renault, Tristan; Faury, Nicole; Lerond, Sophie; Garcia, Celine; Chollet, Bruno; Joly, Jean-pierre; Lapegue, Sylvie; Harrang, Estelle; Arzul, Isabelle.
Bonamiosis due to the parasite Bonamia ostreae has been associated with massive mortality outbreaks in European flat oyster stocks in Europe. As eradication and treatment are not possible, the control of the disease mainly relies on transfer restriction. Moreover, selection has been applied to produce resistant flat oyster families, which present better survival and lower prevalence than non-selected oysters. In order to better understand the mechanisms involved in resistance to bonamiosis, cellular and molecular responses of 2 oyster groups (selected oysters and wild-type oysters) were analyzed in the context of experimental injection and cohabitation infections. Cellular responses including non-specific esterases detection, ROS production and...

Bonamia ostreae, parasite de l'huître plate, Ostrea edulis: sa position taxonomique parmi les parasites du groupe "microcell", analyses des interactions hôte/parasite chez plusieurs populations d'huîtres plates ArchiMer
Besnard-cochennec, Nathalie.
Bonamia ostreae is the causative agent of hemocyte disease. It has been detected first during mortalities occured in Franc in 1979. Bonamiosis spread rapidly among oysters farming in France and Europ. Another species, Bonamia sp. has been detected in New Zealand and Australia. These paasites were both included in the "microcell" group. This group contains 2 others parasites, Mikrocytos mackini affecting Crassostrea gigas in British Columbia (Canada) and M. roughleyi affecing Saccostrea glomerata in Australia. Structural and molecular caracterisation of all these parasites were realised. Results show tha B. ostreae, B. sp., B. exitiosus. Molecular tools are available for genus detection, Bonamia spp. and Mikrocytos and species identification (B. ostreae, B....

A proteomic study of resistance to Brown Ring disease in the Manila clam, Ruditapes philippinarum ArchiMer
Smits, M.; Artigaud, Sebastien; Bernay, B.; Pichereau, Vianney; Bargelloni, L.; Paillard, Christine.
Marine mollusk aquaculture has more than doubled over the past twenty years, accounting for over 15% of total aquaculture production in 2016. Infectious disease is one of the main limiting factors to the development of mollusk aquaculture, and the difficulties inherent to combating pathogens through antibiotic therapies or disinfection have led to extensive research on host defense mechanisms and host-pathogen relationships. It has become increasingly clear that characterizing the functional profiles of response to a disease is an essential step in understanding resistance mechanisms and moving towards more effective disease control. The Manila clam, Ruditapes philippinarum, is a main cultured bivalve species of economic importance which is affected by...
