Early selection of elite clones of an ornamental bromeliad in vitro Ciência Rural
Manfio,Candida Elisa; Motoike,Sérgio Yoshimitsu; Paula,Cláudio Coelho de; Valente,Magno Sávio; Melo,Cristiane Gamarano.
Orthophytum grossiorum is a typical bromeliad from Atlantic forestry threatened of extinction. The objectives of this research were to select O. grossiorum clones with ornamental values easy to propagate in vitro, and establish in vitro propagation protocols for these clones. The project was developed in three steps: germination and in vitro selection of seedlings responsive to BAP (6-benzylaminopurine), selection of clones with ornamental values, and establishment of protocol for in vitro propagation of the selected clones. In the first step only 18.33% of plantlets germinated in vitro were responsive to BAP. These plantlets were selected and replicated in vitro several times, each replicated plantlet constituting a clone. In the second step these clones...

Selection of mango rosa genotypes in a breeding population using the multivariate-biplot method Ciência Rural
Maia,Maria Clideana Cabral; Araújo,Lúcio Borges de; Dias,Carlos Tadeu dos Santos; Oliveira,Luís Cláudio de; Vasconcelos,Lúcio Flavo Lopes; Carvalho Júnior,José Eduardo Vasconcelos de; Simeão,Marcelo; Bastos,Yuri Gagarin Muniz.
ABSTRACT: Mango ( Mangifera indica L.) trees stand out among the main fruit trees cultivated in Brazil. The mango rosa fruit is a very popular local variety (landrace), especially because of their superior technological characteristics such as high contents of Vitamin C and soluble solids (SS), as well as attractive taste and color. The objective of this study was to select a breeding population of mango rosa (polyclonal variety; ≥5 individuals) that can simultaneously meet the fresh and processed fruit markets, using the multivariate method of principal components and the biplot graphic. The principal components, biplot graphic, and phenotype correlations were obtained using the R (2012) software. Pulp percentage and the pulp, skin, and seed mass...

Predicting the genetic gain in the Brazilian white maize landrace Ciência Rural
Souza,Ana Raquel Ribeiro e; Miranda,Glauco Vieira; Pereira,Messias Gonzaga; Souza,Leandro Vagno de.
The objectives of this research were to evaluate the genetic variability and predict genetic gain in the white maize landrace rescued in Barbacena, Minas Gerais, Brazil. The Brazilian maize landraces have economic and social importance for certain areas in Brazil, and research on this germplasm is very important for local food security. Three experiments were carried out with 100 half-sib progenies, in Campos dos Goytacazes, RJ, Coimbra and Barbacena, Minas Gerais, Brazil. These sites were chosen for their soil and climatic differences. An analysis of variance was significant among the progeny for all characters, indicating the presence of genetic variability between the three populations. In Campos dos Goytacazes and Coimbra, the genetic variation of the...

'Ponkan' mandarin (Citrus reticulata Blanco) immature fruits storage Ciência e Agrotecnologia
Pasqual,Moacir; Dutra,Leonardo Ferreira; Araujo,Aparecida Gomes de; Figueiredo,Milene Alves de.
The aim of this work was to evaluate the effect of 'Ponkan' mandarin (C. reticulata) x 'Pêra' sweet orange (C. sinensis) immature fruits storage and sucrose concentrations on embryos in vitro culture. Fruits with 3 to 4 cm in diameter were harvested and placed inside black polyethylene bags with lateral openings and stored at 5±1ºC during 135 days. Every 15 days a sample was removed, its embryos were excised and individually inoculated in test tubes containing 15 mL of MS medium (Murashige & Skoog, 1962) with sucrose (0, 1.5, 3, 6, 12, 18 and 24 g L-1) and 0.3 mg L-1 GA3 and 1 g L-1 activated charcoal. Those treatments rested 48 hours in the dark and later in a growth room at 27 ± 1ºC with a 16-h photoperiod and 32 µmol m-2 s-1 light intensity....

Resistance of potato clones to necrotic recombinant strains of potato virus y (pvy) Ciência e Agrotecnologia
Ribeiro,Silvia Regina Rodrigues de Paula; Pinto,César Augusto Brasil Pereira; Costa,Suellen Bárbara Ferreira Galvino; Menezes,Mariney de; Figueira,Antonia dos Reis.
The Ry adg allele is widely used by breeders to confer extreme resistance to all strains of PVY. However, the necrotic strain has increased recombination resulting in recent considerable losses in productivity. Thus far, not all necrotic recombinant strains of PVY have been tested for their reaction to the Ry gene. The objective of this study was to identify potato clones carrying the resistant allele and to assess their reaction to the following recombinant strains: NTN (PVY NTN), Wilga (PVY N-Wi), and "curly top" (PVY E). Advanced clones from the potato breeding program at Universidade Federal de Lavras were evaluated through a specific molecular marker for the Ry adg allele. The clones carrying the resistance allele were grafted on tobacco plants...

Resistance of tomato strains to the moth Tuta absoluta imparted by allelochemicals and trichome density Ciência e Agrotecnologia
Oliveira,Celso Mattes de; Andrade Júnior,Valter Carvalho de; Maluf,Wilson Roberto; Neiva,Irã Pinheiro; Maciel,Gabriel Mascarenhas.
We examined the resistance of improved tomato strains rich in 2-tridecanone (2-TD), zingiberene (ZGB) and acyl sugars (AA) to the tomato moth, Tuta absoluta. We also studied whether selection for strains with higher densities of glandular trichomes, and thus presumably strains with higher concentrations of 2-tridecanone, was effective in promoting greater resistance to the moth. The TOM-584 and TOM-679 strains were used as susceptible controls, which have normal concentrations of the three allelochemicals. The improved strain TOM-687, which has a high AA content, has a widely documented resistance and was used as a standard resistant strain. The wild strain PI134417, which is resistant by means of its high 2-TD content, was also used as a standard...
