A strategy to identify genomic expression at single-cell level or a small number of cells Electron. J. Biotechnol.
Li,Biaoru.
Recent advances in functional genomics allow us to estimate the expression of several thousands of genes in the mammalian genome. Techniques such as microarrays, expressed tag sequencing (EST), serial analysis of gene expression (SAGE), subtractive cloning and differential display (DD), and two-dimensional electrophoresis gel have been extensively used to screen and analyze parallel gene expression. Some pathological processes, for example, tumorigenesis and solid tumour growth, in which the former is derived from a single-cell and the latter has a mixed-cell problem, present new challenges to the limit of these functional genomic techniques. To fully understand the functions of cells in tumorigenesis or in heterogeneous solid tumour masses, it is...

Amplification of genome-integrated BeYDV replicons by transient expression of Rep in Arabidopsis thaliana Electron. J. Biotechnol.
Moon,Yong-Sun; Hefferon,Kathleen L.
Bean yellow dwarf geminivirus (BeYDV) has been used as a potential vector to improve foreign gene expression, specifically, to achieve higher yields of vaccine proteins in plants. Previously, we have shown that when the BeYDV replication initiator protein Rep was provided in trans, replication and gene expression of GUS were enhanced enormously from a BeYDV expression vector in a transient assay system. In this paper, transgenic lines of Arabidopsis (cv. Columbia) were generated harboring the BeYDV cis-acting elements required for replication. Constructs encoding BeYDV Rep or intronless Rep open reading frames (ORFs) were transiently introduced into transgenic plants via Agrobacterium-mediated infiltration in order to examine the relative levels of...

Identification of phase relative genes in tetrasporophytes and female gametophytes of Gracilaria/Gracilariopsis lemaneiformis (Gracilariales, Rhodophyta) Electron. J. Biotechnol.
Ren,Xueying; Zhang,Xuecheng; Sui,Zhenghong.
Genes differentially expressed between tetrasporophytes and female gametophytes of Gracilaria/Gracilariopsis lemaneiformis were isolated by suppression subtractive hybridization (SSH) and screened by dot-blot macro-arrays. Different expression profiles of selected clones based on the results of dot-blot macro-arrays were verified using virtual Northern blots. Totally, 14 phase relative cDNAs had been isolated and sequence identified. Among them, seven cDNAs were respectively homologous to crucial metabolic enzymes, Rab GTPase, RP42 homolog, and two hypothetical proteins, while the rest did not have significant hits in the databases examined. The results of virtual Northern blots revealed that 11 cDNAs were differentially expressed between the two samples,...

A legume genomics resource: The Chickpea Root Expressed Sequence Tag Database Electron. J. Biotechnol.
Jayashree,B; Buhariwalla,Hutokshi K; Shinde,Sanjeev; Crouch,Jonathan H.
Chickpea, a lesser-studied grain legume, is being investigated due to its taxonomic proximity with the model legume genome Medicago truncatula and its ability to endure and grow in relatively low soil water contents making it a model legume crop for the study of agronomic response to drought stress. Public databases currently contain very few sequences from chickpea associated with expression in root tissues. However, root traits are likely to be one of the most important components of drought tolerance in chickpea. Thus, we have generated a set of over 2800 chickpea expressed sequence tags (ESTs) from a library constructed after subtractive suppressive hybridization (SSH) of root tissue from two closely related chickpea genotypes possessing different...

Effect of isomalto-oligosaccharide and gentio-oligosaccharide on the growth and fatty acid profile of Lactobacillus plantarum Electron. J. Biotechnol.
Soto,Carmen.
Background: Lactobacillus sp. are probiotic microorganisms, and some of them are able to produce conjugated linoleic acid (CLA) via the bio-hydrogenation of linoleic acid (LA). Both CLA and LA are polyunsaturated fatty acids commonly used in the prevention and control of cardiovascular disease, high cholesterol, and cancer, among other ailments. The carbon source is one variable that can affect the growth and characteristics of these bacteria. Molecules called prebiotics are known to benefit human health by stimulating the growth and activity of probiotic bacteria present in the intestinal microflora. The aim of this study was to evaluate how different oligosaccharides affect the growth and fatty acid profile of Lactobacillus plantarum (NRRL - B4496). L....

SSR genetic diversity assessment of popular pigeonpea varieties in Malawi reveals unique fingerprints Electron. J. Biotechnol.
Njung'e,Vincent; Deshpande,Santosh; Siambi,Moses; Jones,Richard; Silim,Said; De Villiers,Santie.
Background: Pigeonpea (Cajanus cajan (L.) Millsp.) is a drought tolerant legume of the Fabaceae family and the only cultivated species in the genus Cajanus. It is mainly cultivated in the semi-arid tropics of Asia and Oceania, Africa and America. In Malawi, it is grown as a source of food and income and for soil improvement in intercropping systems. However, varietal contamination due to natural outcrossing causes significant quality reduction and yield losses. In this study, 48 polymorphic SSR markers were used to assess the diversity among all pigeonpea varieties cultivated in Malawi to determine if a genetic fingerprint could be identified to distinguish the popular varieties. Results: A total of 212 alleles were observed with an average of 5.58 alleles...

Production of phenolic metabolites by Deschampsia antarctica shoots using UV-B treatments during cultivation in a photobioreactor Electron. J. Biotechnol.
Sequeida,Álvaro; Tapia,Eduardo; Ortega,Marcelo; Zamora,Pablo; Castro,Álvaro; Montes,Christian; Zúñiga,Gustavo E; Prieto,Humberto.
Deschampsia antarctica (DA), the only species in the Gramineae family endemic to the Antarctic territory, is characterized by a combination of high levels of free endogenous phenylpropanoid compounds under normal in situ and in vitro growth conditions. In this article, we describe the design and use of a specific temporary immersion photobioreactor to produce both increased DA biomass and secondary metabolite accumulation by UV-B elicitation during cultivation. Three min-long immersions in an induction medium applied every 4 hrs at 14ºC ± 1 and 20/4 hrs light/darkness photoperiod increased DA biomass production over previous in vitro reports. Biomass duplication was obtained at day 10.7 of culturing, and maximum total phenolics and antioxidant activity...

Multiple gene knock-down by a single lentiviral vector expressing an array of short hairpin RNAs Electron. J. Biotechnol.
Stove,Veronique; Smits,Kaatje; Naessens,Evelien; Plum,Jean; Verhasselt,Bruno.
RNA interference (RNAi), mediated by short double-stranded RNAs, is a powerful mechanism for posttranscriptional gene silencing. Sustained expression of short hairpin RNA (shRNA) can be accomplished in mammalian cells by viral delivery systems. Using lentiviral constructs, stable gene silencing is established both in dividing and non-dividing cells. Targeting one single gene can lead to the development of escape mutants or may be insufficient to silence redundant pathways. Therefore, simultaneous targeting of multiple genes may be necessary. We have generated a lentiviral vector-based system for expression of multiple shRNAs from a single viral vector, which also encodes an EGFP reporter protein. We show that knock-down of each single gene from multiple...

Sorption kinetics of Pb2+ and Cu2+ ions from aqueous solution by Nipah palm (Nypa fruticans Wurmb) shoot biomass Electron. J. Biotechnol.
Wankasi,Donbebe; Horsfall Jnr,Michael; Ibuteme Spiff,Ayabaemi.
The sorption kinetics of Pb2+ and Cu2+ ions in aqueous solution by unmodified and mercaptoacetic acid modified biomass of Nypa fruticans shoot has been investigated. The equilibrium sorption capacity of Pb2+ and Cu2+ was determined from the Langmuir equation and found to be 15.59 mg/g and 21.85 mg/g for unmodified biomass and 52.86 mg/g and 66.71 mg/g for modified biomass at 30ºC. The time dependent studies showed relatively rapid sorption of 5 - 10 min to reach equilibrium for both metal ions. A batch sorption model, based on the assumption of the pseudo-second-order mechanism, was applied to predict the rate constants (g mg-1 min-1), which were found to be 3.59 x 10-4 (Pb2+) and 1.85 x 10-4 (Cu2+) and 2.82 x 10-2 (Pb2+) and 3.67 x 10-2 (Cu2+) on...
