Correlations of recognition memory performance with expression and methylation of brain-derived neurotrophic factor in rats Biol. Res.
Muñoz,Pablo C; Aspé,Mauricio A; Contreras,Luis S; Palacios,Adrián G.
Object recognition memory allows discrimination between novel and familiar objects. This kind of memory consists of two components: recollection, which depends on the hippocampus, and familiarity, which depends on the perirhinal cortex (Pcx). The importance of brain-derived neurotrophic factor (BDNF) for recognition memory has already been recognized. Recent evidence suggests that DNA methylation regulates the expression of BDNF and memory. Behavioral and molecular approaches were used to understand the potential contribution of DNA methylation to recognition memory. To that end, rats were tested for their ability to distinguish novel from familiar objects by using a spontaneous object recognition task. Furthermore, the level of DNA methylation was...

miR-22 suppresses tumorigenesis and improves radiosensitivity of breast cancer cells by targeting Sirt1 Biol. Res.
Zhang,Xia; Li,Yuehua; Wang,Dan; Wei,Xiaoer.
Abstract Background miR-22 has been shown to be frequently downregulated and act as a tumor suppressor in multiple cancers including breast cancers. However, the role of miR-22 in regulating the radioresistance of breast cancer cells, as well as its underlying mechanism is still not well understood. Methods The expressions of miR-22 and sirt1 at mRNA and protein levels were examined by qRT-PCR and Western Blot. The effects of miR-22 overexpression and sirt1 knockdown on cell viability, apoptosis, radiosensitivity, γ-H2AX foci formation were evaluated by CCK-8 assay, flow cytometry, colony formation assay, and γ-H2AX foci formation assay, respectively. Luciferase reporter assay and qRT-PCR analysis were performed to confirm the...

Candidate genes and pathogenesis investigation for sepsis-related acute respiratory distress syndrome based on gene expression profile Biol. Res.
Wang,Min; Yan,Jingjun; He,Xingxing; Zhong,Qiang; Zhan,Chengye; Li,Shusheng.
BACKGROUND: Acute respiratory distress syndrome (ARDS) is a potentially devastating form of acute inflammatory lung injury as well as a major cause of acute respiratory failure. Although researchers have made significant progresses in elucidating the pathophysiology of this complex syndrome over the years, the absence of a universal detail disease mechanism up until now has led to a series of practical problems for a definitive treatment. This study aimed to predict some genes or pathways associated with sepsis-related ARDS based on a public microarray dataset and to further explore the molecular mechanism of ARDS RESULTS: A total of 122 up-regulated DEGs and 91 down-regulated differentially expressed genes (DEGs) were obtained. The up- and down-regulated...

In vitro assessment of a computer-designed potential anticancer agent in cervical cancer cells Biol. Res.
Visagie,Michelle Helen; Jaiswal,Seema Rummurat; Joubert,Anna Margaretha.
BACKGROUND: Computer-based technology is becoming increasingly essential in biological research where drug discovery programs start with the identification of suitable drug targets. 2-Methoxyestradiol (2ME2) is a 17ß-estradiol metabolite that induces apoptosis in various cancer cell lines including cervical cancer, breast cancer and multiple myeloma. Owing to 2ME2's poor in vivo bioavailability, our laboratory in silico-designed and subsequently synthesized a novel 2ME2 analogue, 2-ethyl-3-O-sulphamoyl-estra-1,3,5(10),15-tetraen-17-ol (ESE-15-ol), using receptor- and ligand molecular modeling. In this study, the biological effects of ESE-15-ol (180 nM) and its parent molecule, 2ME2 (1 μM), were assessed on morphology and apoptosis induction in...

Relevance of a neutral cue in a two-choice detection task in the rat Biol. Res.
HAMAME,CARLOS M; DELANO,PAUL H; ROBLES,LUIS.
Relevance of a neutral cue for performance in a two-choice visuospatial detection task was examined. Nine rats were trained, 5 with short intertrial interval (ITI) and 4 with long ITI, to detect a target (lateral lights) presented after a neutral-cue (central light). The removal of the neutral-cue decreased accuracy and increased response latencies and omissions. These results demonstrate that a neutral-cue, preceding the target, is relevant for the performance, suggesting that rats are highly expectant during the neutral-cue and reallocate attentional resources during ITI. Furthermore, latencies were higher, omissions were lower and the fall of accuracy was greater for rats with long than with short ITI, which could indicate that the neutral-cue was more...

Inhibition of peroxidase activity and scavenging of reactive oxygen species by astilbin isolated from Dimorphandra mollis (Fabaceae, Caesalpinioideae) Biol. Res.
PETACCI,FERNANDO; FREITAS,SILVIA S; BRUNETTI,IGUATEMY L; KHALIL,NAJEH M.
Astilbin (5,7,3’,4’-tetrahydroxy-2,3-dihydroflavonol-3-ß-o-rhamnoside), a flavonoid with a large range of biological activities, was isolated from Dimorphandra mollis, a shrub common to the Brazilian Cerrado. The purpose of this study is to verify the effects of astilbin on myeloperoxidase (MPO) and horseradish peroxidase (HRP), and its antioxidant activity against hypochlorous acid (HOCl) and total antioxidant activity (TAC) by the 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical cation (ABTS•+). Astilbin inhibited MPO and HRP activities in a concentration-dependent relationship and effectively scavenged HOCl. The TAC by ABTS•+ of astilbin (IC50 ~ 20 mM) was higher than that of uric acid, which was used as a positive control. These data...

Oscillatory fluid flow elicits changes in morphology, cytoskeleton and integrin-associated molecules in MLO-Y4 cells, but not in MC3T3-E1 cells Biol. Res.
Xu,Huiyun; Zhang,Jian; Wu,Jiawei; Guan,Ying; Weng,Yuanyuan; Shang,Peng.
Interstitial fluid flow stress is one of the most important mechanical stimulations of bone cells under physiological conditions. Osteocytes and osteoblasts act as primary mechanosensors within bones, and in vitro are able to respond to fluid shear stress, both morphologically and functionally. However, there is little information about the response of integrin-associated molecules using both osteoblasts and osteocytes. In this study, we investigated the changes in response to 2 hours of oscillatory fluid flow stress in the MLO-Y4 osteocyte-like cell line and the MC3T3-E1 osteoblast-like cell line. MLO-Y4 cells exhibited a significant increase in the expression of integrin-associated molecules, including OPN, CD44, vinculin and integrin avp3. However,...

Biophilosophical and epistemological problems in the study of living beings: Reflections on the views of Humberto Maturana Biol. Res.
SERANI-MERLO,ALEJANDRO.
Theories on the nature of living beings have been present in our culture since the beginnings of science and philosophy in ancient Greece. The two major theoretical approaches to living beings, philosophical mechanism and Aristotelian realism, appear today with renewed force in almost every confrontation concerning the theoretical considerations of life. In recent times a strong and prolific school of thought has risen, headed by the Chilean neurobiologist Humberto Maturana. This author and his school have developed a complex and articulated theoretical system beginning with a theory of living beings and a `biology of cognition,' and extending to ethical, political, and even metaphysical considerations. This work is one of the first efforts to perform a...

Antioxidant activity of extract from Polygonum cuspidatum Biol. Res.
HSU,CHIN-YUAN; CHAN,YU-PEI; CHANG,JELI.
Numerous diseases are induced by free radicals via lipid peroxidation, protein peroxidation and DNA damage. It has been known that a variety of plant extracts have antioxidant activities to scavenge free radicals. Whether Polygonum cuspidatum Sieb. et Zuce has antioxidant activity is unknown. In this study, dried roots of Polygonum cuspidatum were extracted by ethanol and the extract was lyophilized. Free radical scavenging assays, superoxide radical scavenging assays, lipid peroxidation assays and hydroxyl radical-induced DNA strand scission assays were employed to study antioxidant activities. The results indicate that the IC50 value oí Polygonum cuspidatum extract is 110 µg/ml in free radical scavenging assays, 3.2 µg/ml in superoxide radical scavenging...

In vivo expression of ß-galactosidase by rat oviduct exposed to naked DNA or messenger RNA Biol. Res.
RIOS,MARIANA; VENEGAS,ALEJANDRO; CROXATTO,HORACIO B.
Intra-oviductal administration of RNA obtained from oviducts of estradiol-treated rats resulted in accelerated egg transport (<A HREF="#rios97">Ríos et al., 1997</A>). It is probable that estradiol-induced messenger RNA (mRNA) entered oviductal cells and was translated into the proteins involved in accelerated egg transport. In order to test this interpretation we deposited in vivo 50 µg of pure ß-galactosidase (ß-gal) mRNA, 50 µg of pure DNA from the reporter gene ß-gal under SV40 promoter or the vehicle (control oviducts) into the oviductal lumen of rats. Twenty four hours later the ß-gal activity was assayed in oviductal tissue homogenates using o-nitrophenyl-ß-D-galactopyranoside as a substrate. The administration of ß-gal mRNA and pSVBgal...

Progress in tear microdesiccate analysis by combining various transmitted-light microscope techniques Biol. Res.
Traipe-Salas,Felipe; Traipe-Castro,Leonidas; Salinas-Toro,Daniela; López,Daniela; Valenzuela,Felipe; Cartes,Christian; Toledo-Araya,Héctor; Pérez,Claudio; Solís,Remigio López.
BACKGROUND: Tear desiccation on a glass surface followed by transmitted-light microscopy has served as diagnostic test for dry eye. Four distinctive morphological domains (zones I, II, III and transition band) have been recently recognized in tear microdesiccates. Physicochemical dissimilarities among those domains hamper comprehensive microscopic examination of tear microdesiccates. Optimal observation conditions of entire tear microdesiccates are now investigated. One-&#956;! aliquots of tear collected from individual healthy eyes were dried at ambient conditions on microscope slides. Tear microdesiccates were examined by combining low-magnification objective lenses with transmitted-light microscopy (brightfield, phase contrasts Ph1,2,3 and...

In vitro approach to the chemical drive of breathing Biol. Res.
EUGENIN,JAIME; LLONA,ISABEL; INFANTE,CLAUDIA D; AMPUERO,ESTIBALIZ.
Since its introduction two decades ago, the isolated brain stem-spinal cord preparation of neonatal rodents has been the preferred method used to reveal the mystery underlying the genesis of the respiratory rhythm. Little research using this in vitro approach has focused on the study of the central respiratory chemosensitivity. Some unexpected findings obtained with the brain stem-spinal cord preparation have added new questions that challenge our previous theoretic framework. Some of these findings are addressed here.

Li,Pengfei; Yu,Xuejing; Xie,Jianshan; Yao,Xiaolei; Liu,Wenzhong; Yao,Jianbo; Zhu,Zhiwei; Lyu,Lihua.
Abstract Background: Cocaine- and amphetamine-regulated transcript (CART), discovered initially by via differential display RT-PCR analysis of brains of rats administered cocaine, is expressed mainly in central nervous system or neuronal origin cells, and is involved in a wide range of behaviors, such as regulation of food intake, energy homeostasis, and reproduction. The hens egg-laying rate mainly depends on the developmental status of follicles, expression of CART have not been identified from hen follicles, the regulatory mechanisms of CART biological activities are still unknown. The objective of this study was to characterize the mRNA expression of CART in hen follicular granulosa cells and determine CART peptide localization and regulatory role...

Colorectal cancer DNA methylation patterns from patients in Manaus, Brazil Biol. Res.
Almeida,Fabiana Greyce Oliveira; Aquino,Priscila Ferreira de; Souza,Afonso Duarte Leäo de; Souza,Antonia Queiroz Lima de; Vinhote,Sonia do Carmo; Mac-Cormick,Thais Messias; Silva,Marcelo Soares da Mota; Chalub,Sidney Raimundo Silva; Fischer,Juliana de Saldanha da Gama; Carvalho,Paulo Costa; Carvalho,Maria da Gloria da Costa.
BACKGROUND: DNA methylation is commonly linked with the silencing of the gene expression for many tumor suppressor genes. As such, determining DNA methylation patterns should aid, in times to come, in the diagnosis and personal treatment for various types of cancers. Here, we analyzed the methylation pattern from five colorectal cancer patients from the Amazon state in Brazil for four tumor suppressor genes, viz.: DAPK, CDH1, CDKN2A, and TIMP2 by employing a polymerase chain reaction (PCR) specific to methylation. Efforts in the study of colorectal cancer are fundamental as it is the third most of highest incidence in the world. RESULTS: Tumor biopsies were methylated in 1/5 (20 %), 2/5 (40 %), 4/5 (80 %), and 4/5 (80 %) for CDH1, CDKN2A, DAPK, and TIMP2...

Essential oils from two Eucalyptus from Tunisia and their insecticidal action on Orgyia trigotephras (Lepidotera, Lymantriidae) Biol. Res.
Slimane,Badreddine Ben; Ezzine,Olfa; Dhahri,Samir; Jamaa,Mohamed Lahbib Ben.
BACKGROUND: Essential oils extracted from aromatic and medicinal plants have many biological properties and are therefore an alternative to the use of synthetic products. The chemical composition of essential oils from two medicinal plants (Eucalyptus globulus and E. lehmannii) was determined and, their insecticidal effects on the third and fourth larval stages of Orgyia trigotephras were assessed. RESULTS: Larvae were collected from Jebel Abderrahmane (North-East of Tunisia), conserved in groups of 50/box (21 × 10 × 10 cm) at a temperature of 25°C. Larvae were tested for larvicidal activities of essential oils. Each oil was diluted in ethanol (96%) to prepare 3 test solutions (S1 = 0.05%, S2 = 0.10% and S3 = 0.50%). Essential oils were used for contact,...

Study of the expression of carotenoid biosynthesis genes in wild-type and deregulated strains of Xanthophyllomyces dendrorhous (Ex.: Phaffia rhodozyma) Biol. Res.
LODATO,PATRICIA; ALCAÍNO,JENNIFER; BARAHONA,SALVADOR; RETAMALES,PATRICIO; JIMÉNEZ,ANTONIO; CIFUENTES,VÍCTOR.
The expression, at the mRNA level, of carotenoid biosynthetic genes from Xanthophyllomyces dendrorhous was studied by RT-PCR. The experimental conditions for the RT-PCR assay were standardized to quantify the relative transcript levels of idi, crtE, crtYB and crtI genes. This work attempted to correlate astaxanthin production with the transcript levels of carotenogenic genes in a wild-type strain (UCD 67-385) and two overproducer deregulated strains (atxS1 and atxS2). At 3 day cultures, the wild-type strain contained higher transcript levels from the crtE and crtYB genes on minimal medium than on rich medium. Similarly, carotenoid production was higher on minimal medium than on rich medium. However, carotenoid production in the atxS1 and atxS2 strains was...
