Adhesion, biofilm and genotypic characteristics of antimicrobial resistant Escherichia coli isolates BJM
Cergole-Novella,Maria C.; Pignatari,Antonio C.C.; Guth,Beatriz E.C..
Aggregative adherence to human epithelial cells, most to renal proximal tubular (HK-2) cells, and biofilm formation was identified among antimicrobial resistant Escherichia coli strains mainly isolated from bacteremia. The importance of these virulence properties contributing to host colonization and infection associated with multiresistant E. coli should not be neglected.

Streptomyces lunalinharesii 235 prevents the formation of a sulfate-reducing bacterial biofilm BJM
Rosa,Juliana Pacheco da; Tibúrcio,Samyra Raquel Gonçalves; Marques,Joana Montezano; Seldin,Lucy; Coelho,Rosalie Reed Rodrigues.
ABSTRACT Streptomyces lunalinharesii strain 235 produces an antimicrobial substance that is active against sulfate reducing bacteria, the major bacterial group responsible for biofilm formation and biocorrosion in petroleum reservoirs. The use of this antimicrobial substance for sulfate reducing bacteria control is therefore a promising alternative to chemical biocides. In this study the antimicrobial substance did not interfere with the biofilm stability, but the sulfate reducing bacteria biofilm formation was six-fold smaller in carbon steel coupons treated with the antimicrobial substance when compared to the untreated control. A reduction in the most probable number counts of planktonic cells of sulfate reducing bacteria was observed after treatments...

Roles of quorum sensing molecules from Rhizobium etli RT1 in bacterial motility and biofilm formation BJM
Dixit,Swarnita; Dubey,Ramesh Chand; Maheshwari,Dinesh Kumar; Seth,Prahlad Kishore; Bajpai,Vivek K..
ABSTRACT Strain RT1 was isolated from root nodules of Lens culinaris (a lentil) and characterized as Rhizobium etli (a Gram-negative soil-borne bacterium) by 16S rDNA sequencing and phylogenetic analysis. The signaling molecules produced by R. etli (RT1) were detected and identified by high-performance liquid chromatography coupled with mass spectrometry. The most abundant and biologically active N-acyl homoserine lactone molecules (3-oxo-C8-HSL and 3-OH-C14-HSL) were detected in the ethyl acetate extract of RT1. The biological role of 3-oxo-C8-HSL was evaluated in RT1. Bacterial motility and biofilm formation were affected or modified on increasing concentrations of 3-oxo-C8-HSL. Results confirmed the existence of cell communication in RT1 mediated by...

Virulence-associated characteristics of Enterococcus faecalis strains isolated from clinical sources BJM
Furumura,Márcia T.; Figueiredo,Patricia M.S.; Carbonell,Gleize V.; Darini,Ana Lucia da Costa; Yano,Tomomasa.
Thirty-two clinical isolates of Enterococcus faecalis were screened for virulence factors. Twenty-four (75%) isolates produced hemolysin on Mueller-Hinton blood agar plates with sheep erythrocytes. However, the cell free heat-stable hemolysin was detected in all isolates (100%) of E. faecalis when grown in BHI-GA (BHI medium supplemented with 1% glucose and 0.03% L-arginine), but not in BHI broth alone. Twenty-four isolates (75%) produced caseinase and 23 (71.9%) lipase, but none of the isolates produced gelatinase. Fifteen (46.9%) culture filtrates caused rounding and membrane alterations with blebbing formation followed by death in HeLa and HEp-2 cells, but not in Vero cells. Thirteen isolates (40.6%) agglutinated rabbit erythrocytes, but did not produce...

Fusarium culmorum affects expression of biofilm formation key genes in Bacillus subtilis BJM
Khezri,Maryam; Jouzani,Gholamreza Salehi; Ahmadzadeh,Masoud.
Abstract It is known that there is correlation between biofilm formation and antagonistic activities of Bacillus subtilis strains; but, the mechanism of this correlation is not clear. So, the effect of the plant pathogen (Fusarium culmorum) on the biofilm formation in a B. subtilis strain with high antagonistic and biofilm formation activities was studied. The expression of sinR and tasA genes involved in the biofilm formation was studied in both single culture of bacterium (B) and co-culture with F. culmorum (FB) using real-time PCR. The results revealed that the expression of the sinR gene in both B and FB conditions was continuously decreased during the biofilm formation period and, after 24 h (B4 and FB4), it reached 1% and 0.3% at the planktonic phase...

Elastin increases biofilm and extracellular matrix production of Aspergillus fumigatus BJM
Brandão,Ildnay de Souza Lima; Oliveira-Moraes,Heloiza Maria da Silva; Motta,Cristina Maria de Souza; Oliveira,Neiva Tinti de; Magalhães,Oliane Maria Correia.
Abstract Aspergillus fumigatus is an opportunistic saprobe fungus that accounts for 90% of cases of pulmonary aspergillosis in immunosuppressed patients and is known for its angiotropism. When it reaches the respiratory tract, A. fumigatus interacts with structural components and blood vessels of the lungs, such as elastin. To understand the effect of this structural component, we examined the effect of elastin on the production and development of the biofilm of A. fumigatus. In RPMI containing 10 mg/mL of elastin, a significant increase (absorbance p < 0.0001; dry weight p < 0.0001) in the production of biofilm was observed in comparison to when RPMI was used alone, reaching a maximum growth of 18.8 mg (dry weight) of biofilm in 72 h. In addition,...

Virulence factors and biofilm production by isolates of Bacteroides fragilis recovered from dog intestinal tracts BJM
Reis,Ana Catarina M.; Silva,Janice O.; Laranjeira,Bruno J.; Pinheiro,Adriana Q.; Carvalho,Cibele B.M..
Bacteroides fragilis colonizes dog guts both as a commensal and as an opportunistic pathogen. This study aims to evaluate virulence factors of 13 B. fragilis strains isolated from dog intestinal tracts and their ability for biofilm formation. Capsules were detected in all the evaluated strains. A total of 61.5% of all strains were biofilm producers. These attributes most likely play an important role in B. fragilis persistent colonization in the gut.

Marques,Viviane Figueira; Motta,Cássia Couto da; Soares,Bianca da Silva; Melo,Dayanne Araújo de; Coelho,Shana de Mattos de Oliveira; Coelho,Irene da Silva; Barbosa,Helene Santos; Souza,Miliane Moreira Soares de.
Abstract Staphylococcus spp. play an important role in the etiology of bovine mastitis. Staphylococcus aureus is considered the most relevant species due to the production of virulence factors such as slime, which is required for biofilm formation. This study aimed to evaluate biofilm production and its possible relation to beta-lactamic resistance in 20 S. aureus isolates from bovine mastitic milk. The isolates were characterized by pheno-genotypic and MALDI TOF-MS assays and tested for genes such as icaA, icaD, bap, agr RNAIII, agr I, agr II, agr III, and agr IV, which are related to slime production and its regulation. Biofilm production in microplates was evaluated considering the intervals determined along the bacterial growth curve. In addition, to...

Bruder-Nascimento,Ariane; Camargo,Carlos Henrique; Mondelli,Alessandro Lia; Sugizaki,Maria Fátima; Sadatsune,Terue; Bagagli,Eduardo.
Over the last decades, there have been important changes in the epidemiology of Candida infections. In recent years, Candida species have emerged as important causes of invasive infections mainly among immunocompromised patients. This study analyzed Candida spp. isolates and compared the frequency and biofilm production of different species among the different sources of isolation: blood, urine, vulvovaginal secretions and peritoneal dialysis fluid. Biofilm production was quantified in 327 Candida isolates obtained from patients attended at a Brazilian tertiary public hospital (Botucatu, Sao Paulo). C. albicans ALS3 gene polymorphism was also evaluated by determining the number of repeated motifs in the central domain. Of the 198 total biofilm-positive...

2(5H)-Furanone: a prospective strategy for biofouling-control in membrane biofilm bacteria by quorum sensing inhibition BJM
Ponnusamy,Kannan; Paul,Diby; Kim,Young Sam; Kweon,Ji Hyang.
Biofouling of membranes demands costly periodic cleaning and membrane replacement. A sustainable and environmentally friendly solution for maintenance is not available and would be of great interest for many purposes including economical. As complex biofilm formation by environmental strains is the major cause of biofouling and biofilm formation in most cases are controlled by N-Acylhomoserine lactone (AHL)mediated Quorum Sensing (QS). An effort was made to understand the appropriateness of 2(5H)-furanone, to use against biofouling of membranes. QS inhibition activity by 2(5H)-furanone was studied using bioindicator strains and known AHLs of different acyl chain lengths. The biofilm inhibition was studied by growth analysis on polystyrene plate of...

Multidrug-resistant Candida glabrata strains obtained by induction of anidulafungin resistance in planktonic and biofilm cells BJPS
Hatwig,Camila; Balbueno,Eduardo A.; Bergamo,Vanessa Z.; Pippi,Bruna; Fuentefria,Alexandre M.; Silveira,Gustavo P..
Candida glabrata has emerged as a common cause of serious life-threatening fungal infections, largely owing to their low susceptibility to azole antifungals. Recent guidance indicates the use of echinocandins as the first-choice drug for the treatment of systemic infections of C. glabrata; however, C. glabrata resistance to echinocandins is reportedly increasing. Herein, we present the induction of anidulafungin resistance in planktonic and sessile cells of C. glabrata and the development of fluconazole cross-resistance. MICs of 21 clinical C. glabrata strains were determined by a broth microdilution method using anidulafungin and fluconazole. Biofilm formation on a tracheal catheter was determined using 1- × 1-cm2 polyvinyl polychloride catheter...
