Microbial community structure in the sea surface microlayer at two contrasting coastal sites in the northwestern Mediterranean Sea ArchiMer
Joux, Fabien; Agogue, Helene; Obernosterer, Ingrid; Dupuy, Christine; Reinthaler, Thomas; Herndl, Gerhard J.; Lebaron, Philippe.
In an attempt to compare the microbial community structure between the sea surface microlayer (SML) and subsurface waters (SSW), we determined the enrichment factors (EF: the ratio of abundance or activity in the SML to abundance or activity in SSW) of 13 biological parameters. Samples were taken at 2 contrasting coastal sites in the Mediterranean Sea, corresponding to a high (Barcelona, Spain) and low (Banyuls-sur-Mer, France) urbanized area. Principal component analysis showed that temporal variability was much higher at Barcelona than at Banyuls, and that the characteristics of the SML and SSW samples were more closely related at Barcelona. At both sites, the SML was weakly enriched in heterotrophic bacteria (on average 1.1-fold), Synechococcus spp. (on...

Culture of Microalgae with Ultrafiltered Seawater: A Feasibility Study ArchiMer
Cordier, Clémence; Guyomard, Killian; Stavrakakis, Christophe; Sauvade, Patrick; Coelho, Franz; Moulin, Philippe.
The culture of microalgae is important for the production and maintenance of bivalves. One of the major challenges is to maintain the reliability of microalgae forages over the long term. The aim of this work is to use Ultrafiltered (UF) seawater to cultivate them. Thus, cultures in a volume of 300 L of 2 species of microalgae Tetraselmis and T-isochrysis, were monitored in UF water (membrane pore size: 20 nm) and in sea water usually used on the Ifremer mollusk experimental platform of Bouin (France) (Prefiltration, 3 filtrations and 2 UV). The major result is the securing of microlagae cultures with the absence of parasites in all cultures supplied with ultrafiltered water, unlike analyses of the various control cultures. In the case of T-isochrysis, 3...

Antimelanoma activity of Heterocapsa triquetra pigments ArchiMer
Haguet, Quentin; Bonnet, Antoine; Berard, Jean-baptiste; Goldberg, Joelle; Joguet, Nicolas; Fleury, Audrey; Thiery, Valerie; Picot, Laurent.
Heterocapsa triquetra (Ht), a non-toxic dinophyte, was selected to investigate the presence of pigments inhibiting melanoma cells growth. We developed an innovative microwave-assisted extraction process (MAE), determined the Ht pigment composition using Reverse Phase-High Performance Liquid Chromatography (HPLC) and high-resolution Ultra Performance Liquid Chromatography-Mass Spectrometry E (UPLC-MSE) analysis and assessed their antiproliferative activity in human invasive melanoma cells (A2058) grown in vitro. MAE allowed a fast and efficient extraction of Ht pigments as compared with conventional processes (soaking, sonication). Seventeen pigments and derivatives were unequivocally identified by UPLC-MSᴱ in the Ht extract, among which two were supposed...

Microwave-assisted extraction of phycobiliproteins from Porphyridium purpureum ArchiMer
Juin, Camille; Cherouvrier, Jean-rene; Thiery, Valerie; Gagez, Anne-laure; Berard, Jean-baptiste; Joguet, Nicolas; Kaas, Raymond; Cadoret, Jean-paul; Picot, Laurent.
In the present study, microwave-assisted extraction was first employed to extract the phycobiliproteins of Porphyridium purpureum (Pp). Freeze-dried Pp cells were subjected to microwave-assisted extraction (MAE) to extract phycoerythin (PE), phycocyanin (PC), and allophycocyanin (APC). MAE combined reproducibility and high extraction yields and allowed a 180- to 1,080-fold reduction of the extraction time compared to a conventional soaking process. The maximal PE extraction yield was obtained after 10-s MAE at 40 A degrees C, and PE was thermally damaged at temperatures higher than 40 A degrees C. In contrast, a flash irradiation for 10 s at 100 A degrees C was the best process to efficiently extract PC and APC, as it combined a high temperature necessary...

Valeur alimentaire de Tetraselmis striata et T. chui pour les larves de Crassostrea gigas ArchiMer
Ponis, Emanuele; Parisi, Guilina; Robert, Rene.
In an attempt to simplify mollusc hatchery-nursery procedures and to provide cost-effective alternative diets, the use of concentrated and preserved microalgae is an alternative to the use of fresh microalgae. Because of their ability for preservation, Prasinophytes appears to be good candidates for remote algal production. In the present work, the food value of fresh and preserved Tetraselmis chui and T. striata for umboned Crassostrea gigas larvae was studied. During the first experiment, fresh T. striata led to the best larval development, but these performances were not confirmed subsequently. When used as monospecific diets the food value of both Prasinophytes is unsufficient to support a good C. gigas larval development. However in bispecific diets a...

Zeaxanthin, an abundant carotenoid present in fruits, vegetables and algae was reported to exert antiproliferative activity and induce apoptosis in human uveal melanoma cells. It also inhibited uveal melanoma tumor growth and cell migration in nude mice xenograft models. Here we report that zeaxanthin purified from the rhodophyte Porphyridium purpureum (Bory) K.M.Drew & R.Ross, Porphyridiaceae, promotes apoptosis in the A2058 human melanoma cell line expressing the oncogenic BRAF V600E mutation. Zeaxanthin 40 μM (IC50) induced chromatin condensation, nuclear blebbing, hypodiploidy, accumulation of cells in sub-G1 phase, DNA internucleosomal fragmentation and activation of caspase-3. Western blot analysis revealed that zeaxanthin induced up-regulation...

The use of Arthrospira platensis in rearing Nile tilapia (Oreochromis niloticus) in salt water Rev. Ciênc. Agron.
Leite,Lorena Alves; Quaresma,Fabrízia da Silva; Ribeiro,Pedro Florêncio; Farias,Wladimir Ronald Lobo; Souza,Bartolomeu Warlene Silva de.
ABSTRACT Nile tilapia (Oreochromis niloticus) is a euryhaline species which, in marine systems, can provide animal protein where fresh water is scarce. As a result, adequate water management to maintain sustainability is essential, and the use of a salt water recirculating system would be an advantage at the present time. In addition, Arthrospira platensis is used in the diet of aquatic organisms, as it is rich in protein. The aim of this study, therefore, was to evaluate the use of A. platensis as a food supplement in the performance of Nile tilapia in a salt water recirculating system. The experimental model included two groups, with four replications. Each crop had a duration of 45 days (control and test), giving a total of 90 days, with the control...

Influence of nutrients on biomass and oil yield from microalgae Chlorella vulgaris for biodiesel production Rev. Ciênc. Agron.
Araujo,Glacio Souza; Lopes,Dilliani Naiane Mascena; Santiago,Clarice da Silva; Silva,José William Alves da; Fernandes,Fabiano André Narciso.
ABSTRACT Microalgae are commonly used in aquiculture as feed for postlarval mollusks, fish and crustaceans because they are easy to grow, small in size, grow rapidly and have high levels of fatty acids. These microorganisms also accumulate high amounts of oil, which can be extracted and converted into biodiesel using chemical processes. In this work, freshwater microalgae Chlorella vulgaris was grown in the Live Food Production Laboratory (LABPAV/IFCE Aracati Campus), with urea (stock solution 1), triple superphosphate (stock solution 2) and vitamins (stock solution 3), in growth medium, in triplicate, using three different quantities of stock solutions 1 and 2, but with a constant amount of vitamins. The quantities of 0.5, 1 and 2 mL (T0.5, T1 and T2,...

Use of Spirulina platensis in treatment of fish farming wastewater Rev. Ciênc. Agron.
Nogueira,Sara Monaliza Sousa; Souza Junior,José; Maia,Hudson Damasceno; Saboya,Jefferson Pablo Sousa; Farias,Wladimir Ronald Lobo.
ABSTRACT Each year, the amount of fish produced around the world increases, which contributes to several environmental impacts such as the disposal of effluents without treatment in the environment. This scientific work had as main objective the development of Spirulina platensis in fish effluents, a low cost medium for the production of biomass, in order to reduce the levels of some inorganic nutrients to reach the allowed parameters by the Brazilian environmental standards for effluent disposal and to enable reuse of the water. Nile tilapia (Oreochromis niloticus) fingerlings were produced in fresh water. The effluent generated by the culture was transferred to a pool where seawater was added until the salinity of 10 ‰ was reached. A strain of...

Neves,Cristina; Maroneze,Mariana Manzoni; Santos,Aline Meireles dos; Francisco,Erika Cristina; Wagner,Roger; Zepka,Leila Queiroz; Jacob-Lopes,Eduardo.
ABSTRACT This study aimed to evaluate third generation biodiesel production by microalgae Phormidium autumnale using cassava processing wastewater as a platform. Experiments were performed in a heterotrophic bubble column bioreactor. The study focused on the evaluation of the bioreactor (batch and fed-batch) of different operational modes and the analysis of biofuel quality. Results indicate that fed-batch cultivations improved system performance, elevating biomass and oil productions to 12.0 g L−1 and 1.19 g L−1, respectively. The composition of this oil is predominantly saturated (60 %) and monounsaturated (39 %), resulting in a biodiesel that complys with U.S., European and Brazilian standards. The technological route developed indicates potential for...

Digestibility and pricing of Chlorella sorokiniana meal for use in tilapia feeds Scientia Agricola
Barone,Rafael Simões Coelho; Sonoda,Daniel Yokoyama; Lorenz,Evandro Kleber; Cyrino,José Eurico Possebon.
ABSTRACT: Several microalgae contain in excess of 50 % crude protein with amino acid profile comparable to that of fish meal. In addition, high polyunsaturated fatty acid contents encourage their use in animal feeding and nutrition, particularly in the formulation and processing of aquafeeds. This study aims at estimating the feasibility of Chlorella meal as feed ingredient for the feeding and nutrition of farmed tilapia based upon digestibility data. Juvenile tilapia were stocked in conical-bottomed tanks (200 L) with superficial, continuous water flow, and fed to apparent satiation in three daily meals with a reference diet and a test diet containing 30 % lyophilized Chlorella sorokiniana added of an inert marker. Feces were collected overnight by...

Diets of tadpoles from a temporary pond in southeastern Brazil (Amphibia, Anura) Zoologia
Rossa-Feres,Denise de C.; Jim,Jorge; Fonseca,Mariluce Gonçalves.
The diet of tadpoles of 13 anuran species was determined to verify whether food resource partitioning occurs and whether the degree of diet similarity is related to taxonomic affinity. Tadpoles of all species studied were mainly herbivorous, except for these of Leptodactylus fuscus (Schneider, 1799) which were mycophagous. Although some species had exclusive items in their diet, most tadpole species ingested the same items, but differed in the amount of each item consumed. Two guilds were found: tadpoles that feed on diatoms on the pond bottom, and tadpoles that feed on Oedogonium Link, 1820 algae in midwater. Diet similarity was related to the taxonomic relationship, microhabitat and feeding behavior of tadpoles indicating that the community organization...

Chia,Mathias Ahii; Akinsanmi,Jamiu Taiwo; Tanimu,Yahuza; Ladan,Zakari.
The application of synthetic algaecides for the control of algae produces by-products that are sometimes toxic to the environment. There is a need for natural and cheap alternatives to synthetic algaecides. In the present study, we investigated the potential of aqueous crude extract of Azadirachta indica to inhibit the growth of Scenedesmus quadricauda. Phytochemical screening of the extract revealed the presence of groups of bioactive compounds that are capable of inhibiting microalgal growth. Chlorophyll a concentration, dry weight production and cell density of microalga decreased with increasing crude extract concentration. After three days of exposure, the 1000 mg/L extract concentration resulted in complete growth inhibition and cell lysis....
