Micropropagation of Ilex dumosa (Aquifoliaceae) from nodal segments in a tissue culture system Biocell

Micropropagation of Ilex dumosa var. dumosa R. ("yerba señorita") from nodal segments containing one axillary bud was investigated. Shoot regeneration from explants of six-year-old plants was readily achieved in ¹/4 strength Murashige and Skoog medium (¹/4 MS) plus 30 gr·L-1 sucrose and supplemented with 4.4 mM BA. Further multiplication and elongation of the regenerated shoots were obtained by subculture in a fresh medium of similar composition with 1.5 gr·L-1 sucrose. Behavior and preservation of an in vitro collection of European aspen in Spain Biocell

An in vitro collection has been established with selected European aspen from Palencia province (Spain). Currently, this collection includes 32 high quality clones, selected for their good bearing and healthy state. Most of them belong to different discrete local populations. Populus tremula L. was propagated in proliferation Aspen Culture Medium; they required subculture every 3 months. Anogramma chaerophylla belongs to a fern genus with annual sporophytes and potentially perennating gametophytes. In the studied area, Natural Reserve Punta Lara, the plants grow stressed mainly by pollution and the invasion of exotic species. As a part of a project on conservation of ferns inhabiting Buenos Aires province, the objectives of this work were to evaluate in Anogramma chaerophylla the conditions for in-vitro spore germination and to analyse the different stages of its reproductive cycle. Effect of 6-BA on nodal explant bud sproutings of Coffea arabica cv. Mundo Novo Bragantia

Coffee plants can be micropropagated by nodal bud sprouting using the 6-benzylaminopurine (6-BA) hormone. However, literature reports the use of a wide range of 6-BA, from 0.5 to 88.8 µM L-1. So, this study was performed to narrow that range. Nodal explants of Coffea arabica cv Mundo Novo obtained from in vitro plantlets were inoculated on gelled-MS medium supplemented with different concentrations of 6-BA. Two assays were carried out: in the first one, 6-BA was used at concentrations of 0, 5, 25, 50, and 100 µM L-1, being evaluated at 43 and 123 days. Production of pre-basic potato seed by polyvinyl chloride PVC: articulate gutters hydroponic system BABT

The development of more efficient and productive systems for pre-basic seed potato production would improve the quality of the propagative material used by the potato growers, directly affecting the crop yields. A two-year experiment was carried out to evaluate the potato pre-basic seed production by two types of hydroponic systems (fibrocement tiles and articulated PVC gutters), two cultivars (`Baronesa` and `Eliza`) and two types of propagative material (plants coming from in vitro culture and minitubers). The PVC gutters system was highly efficient. When using minitubers, this system reached multiplication rates up to 74 tubers per plant. Minitubers were more productive than in vitro plants, independent of cultivar and hydroponic system utilized.

In vitro establishment and multiplication of the Normania triphylla (Lowe) Lowe BABT

Normania triphylla is an endemic species from Madeira island (Portugal) extinct in the wild since 1991. The aim of this work was to culture the meristems of this species in vitro and to multiply its shoots in order to preserve this endangered species. The best results in terms of multiplication were obtained in Murashige and Skoog medium supplemented with 10 µM 6‑benzylaminopurine (BAP). The number of shoots, the number of nodes and the number of leaves were the most important in this medium. Induction of leafy galls in Acacia mearnsii De Wild seedlings infected by Rhodococcus fascians BABT

Plantlets of blackwattle (Acacia mearnsii De Wild) were inoculated with the bacterium Rhodococcus fascians and cultured in vitro. Leafy galls appeared at the cotyledonary nodes in 75% of the infected plants. The galls were separated from the plants and cultured on a medium containing three-quarters-strength MS salts (Murashige and Skoog, 1962), MS vitamins, 2% sucrose and an antibiotic (cephalothin), supplemented with or without 0.2% activated charcoal. Plant regeneration through somatic embryogenesis of yacón [smallanthus sonchifolius (poepp. and endl.) H. Robinson] BABT

Smallanthus sonchifolius has tuberous roots containing large amounts of fructo-oligosaccharides and its medicinal use has increased due to the hypoglycemic properties reported for this species. An efficient system for propagation via somatic embryogenesis is reported using petiole segments cultivated on MS medium supplemented with combinations of BA, kinetin and 2,4-D, under light and darkness conditions. Embryogenic callus was formed in most of the treatments; however, somatic embryogenesis was promoted by the presence of light. This work aimed at studying the effects of different sources and cytokinin concentrations, as well as explants orientation and time of their permanence in shoots induction medium to obtain high multiplication rate. Four experiments were carried out to evaluate the sprout percentage mean, shoots number per explant, elongated shoots percentage and shoots length. The treatments with 2iP and zeatin did not promote in vitro plant multiplication. ABSTRACT Nidularium procerum and Nidularium innocentii (Bromeliaceae) were cultivated in vitro on media supplemented with different sources and levels of GA3 (gibberellic acid). These sources were the commercial powder (analytical degree) and fermented extract obtained by Fusarium moniliforme via solid state fermentation. The in vitro elongation and rooting of these plants were evaluated after 50 days of cultivation. In this work, plant regeneration via somatic embryogenesis was achieved from leaf and internode derived callus of Wedelia calendulacea, an endangered medicinal plant. Primary callus was induced by culturing leaf disc and internode explant on Murashige and Skoog medium supplemented with 2.0 mg L-1 of 2,4-D under light condition. Transfer of embryogenic callus on a reduced concentration of 2,4-D facilitated somatic embryo development while calluses remained unorganized at the same 2,4-D level. Micropropagation of an elite F1 watermelon (Citrullus lanatus) hybrid from the shoot tip of field grown plants BABT

The aim of this work was to develop a protocol for rapid micropropagation of an elite F1 hybrid watermelon cultivar using shoot tip of field-grown plants. Maximum frequency (73%) of shoot tip showed growth response in MS medium supplemented with 5 mg l-1 benzyl adenine (BA) and 0.1 mg l-1 indole-3 acetic acid (IAA). Upon transfer to cytokinin-enriched medium, the cultures produced multiple shoots and 2.0 mg l-1 BA was optimum in this respect. Addition of gibberellic acid (GA3) in the multiplication medium resulted in better growth of shoots. Antimicrobial activity of the essential oil (EO) of Lippia gracilis Schauer over fungi and heliconia endophytic bacteria was evaluated. For the fungi Geotrichum candidum; Trichoderma viride; Torula herbarum; Paecillomyces sp.; Fusicoccum sp.; P. aeruginens; Curvularia lunata; Aspergillus nidulans; A. flavus; and A. niger mycelial discs were inoculated in PDA medium with oil (0; 420; 440; 460; 480 e 500 µL L-1).
