Anogramma chaerophylla belongs to a fern genus with annual sporophytes and potentially perennating gametophytes. In the studied area, Natural Reserve Punta Lara, the plants grow stressed mainly by pollution and the invasion of exotic species. As a part of a project on conservation of ferns inhabiting Buenos Aires province, the objectives of this work were to evaluate in Anogramma chaerophylla the conditions for in-vitro spore germination and to analyse the different stages of its reproductive cycle. Spores were sterilized in an aqueous solution 10 % of NaCIO during different times and then sown in Petri dishes with Murashige & Skoog medium, without the addition of sucrose. The dishes were kept under laboratory conditions, at 12 h light/ darkness... |

Coffee plants can be micropropagated by nodal bud sprouting using the 6-benzylaminopurine (6-BA) hormone. However, literature reports the use of a wide range of 6-BA, from 0.5 to 88.8 µM L-1. So, this study was performed to narrow that range. Nodal explants of Coffea arabica cv Mundo Novo obtained from in vitro plantlets were inoculated on gelled-MS medium supplemented with different concentrations of 6-BA. Two assays were carried out: in the first one, 6-BA was used at concentrations of 0, 5, 25, 50, and 100 µM L-1, being evaluated at 43 and 123 days. In the second experiment, dosis of 10, 20 and 30 µM L-1, have evaluated at 65 and 100 days. Treatments with 6-BA induced multiple sprouting from the nodal explants, which were best characterized around 100... |

Plantlets of blackwattle (Acacia mearnsii De Wild) were inoculated with the bacterium Rhodococcus fascians and cultured in vitro. Leafy galls appeared at the cotyledonary nodes in 75% of the infected plants. The galls were separated from the plants and cultured on a medium containing three-quarters-strength MS salts (Murashige and Skoog, 1962), MS vitamins, 2% sucrose and an antibiotic (cephalothin), supplemented with or without 0.2% activated charcoal. Histological studies conducted from the sixth to the twenty-second day after plant infection revealed the presence of newly formed meristematic centers, first in the axillary region, then on the petioles and lamina of the leaflets around the apical meristem. Approximately 37% of the galls developed one... |

This work aimed at studying the effects of different sources and cytokinin concentrations, as well as explants orientation and time of their permanence in shoots induction medium to obtain high multiplication rate. Four experiments were carried out to evaluate the sprout percentage mean, shoots number per explant, elongated shoots percentage and shoots length. The treatments with 2iP and zeatin did not promote in vitro plant multiplication. When different BAP concentrations were tested, an increase in sprout percentage was obtained using BAP concentration up to 3.66 mg L-1 (80%), and for shoots number per explant a linear behavior was observed, showing a mean of four shoots per explant with 4.0 mg L-1 BAP. For the 'Flordaguard' rootstocks, the explants... |

ABSTRACT Nidularium procerum and Nidularium innocentii (Bromeliaceae) were cultivated in vitro on media supplemented with different sources and levels of GA3 (gibberellic acid). These sources were the commercial powder (analytical degree) and fermented extract obtained by Fusarium moniliforme via solid state fermentation. The in vitro elongation and rooting of these plants were evaluated after 50 days of cultivation. The GA3 present in the fermented extract possess the same effect of purified GA3 (analytical degree) for the increase of the height of aerial part of shoots of N. innocentii, but not for the N. procerum being the GA3 fermented extract in a lesser degree. The GA3 fermented extract influences negatively the rooting in N. innocentii, while GA3... |

In this work, plant regeneration via somatic embryogenesis was achieved from leaf and internode derived callus of Wedelia calendulacea, an endangered medicinal plant. Primary callus was induced by culturing leaf disc and internode explant on Murashige and Skoog medium supplemented with 2.0 mg L-1 of 2,4-D under light condition. Transfer of embryogenic callus on a reduced concentration of 2,4-D facilitated somatic embryo development while calluses remained unorganized at the same 2,4-D level. A histological analysis confirmed somatic embryo by revealing the presence of a closed vascular system in the developing embryos and lack of a vascularconnection with surrounding callus tissues. Somatic embryos germinated into plantlets upon transfer on MS medium... |

Celosia argentea (Var.) cristata (Amaranthaceae) is a widely cultivated ornamental plant, which has antibacterial, astringent, haemostatic, hypertensive, ophthalmic, and parasitic significance. This study describes a protocol for in vitro callus induction and plant regeneration from leaf and stem explants of C. argentea using Murashige and Skoog (MS) medium. Callus culture was initiated and established from seedling, leaf, and stem explants. Explants were cultured on MS medium supplemented with auxin alone (0.5 mg/L Naphthaleneacetic acid (NAA), 2, 4-Dicholorophenoxyacetic acid (2, 4-D). Green and red compact callus (98%) were induced using MS medium supplemented with 0.5 mg/L NAA and 1.0 mg/L Benzyladenine (BA). Two different concentrations (1.0 mg/L BA +... |

ABSTRACT Multipurpose use of bamboo in rural life makes it as poor man’s timber in Asian countries. Deforestation and industrialization leads to destruction of natural forest. To replenish, a rapid plantation of bamboo could be one of the possible solutions. Bamboo is propagated mainly by vegetative methods though it is not suitable for large scale plantation because of several limitations. Micropropagation is gaining importance for large scale propagation because of its capability in raising huge number of true to type propagules in a limited space in very short span of time. Like any other plant, the chief constraint of bamboo micropropagtion is in vitro contamination arises from several sources including explants. Most of the contaminants are reduced by... |

Nutrients media (Murashige & Skoog, Hoagland & Arnon and White's media) supplemented or not with sucrose and substrates (vermiculite, agar and natrosol) were tested for their effects on plant development and root colonization of micropropagated sweet potato, cv. White Star, by Glomus etunicatum Becker & Gerdemann (isolate INVAM FL S329). Addition of sucrose (3%) did not affect plant development. However, hyphal growth and root colonization were depressed. Contrasting responses to media nutrient concentration were observed for plant height, root colonization, and hyphal growth. The highest concentration of nutrients in Murashige & Skoog medium improved plant development, but this medium decreased hypha growth and inhibited... |

Rhodophiala C. Presl (Amaryllidaceae) is a genus of attractive flowering geophytes native to South America. They have ornamental value, but most species are little known and have conservation problems. The objective of this study was to optimize a micropropagation process to support the use and preservation of three Chilean native species, R. montana (Phil.) Traub, R. splendens (Rengifo) Traub., and R. ananuca (Phil.) Traub. The research evaluated the feasibility of implementing liquid medium culture and assessed the influence of different tissue culture systems on the shoot production and biomass increment of small bulbs. Three experiments were carried out. The first one determined the influences of flask size and volume of media; the second compared... |
