Nowadays, PCR-based DNA markers allow the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European flat oyster (Ostrea edulis) populations has aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Results were congruent between allozymes, microsatellites and mitochondrial DNA and support an isolation by distance model. High variability was observed in the breadth of diversity between populations using a mitochondrial marker, this highlights the small effective population sizes in some locations. A 10-fold quantitative difference was observed in Fst... |

A comparative study of the growth of the oysters Crassostrea gigas, Crassostrea angulata and their hybrids was carried out in 1999 in system controlled with contribution of the micro alga Skeletonema costatum. The results showed significant differences of the pondéral growth of the four studied progenies. The animals C. gigas had growth rates 1.4 times greater than C. angulata oysters. The hybrid progenies were shown more powerful than Portuguese oysters but are not different from the Japanese oysters. The ecophysiologic study showed that the averages of filtration and oxygen uptake, obtained by means of a three hours recording, cannot differentiate the four studied progenies. On the other hand, the calculation of the FTA (Feeding Time Activity) allowed to... |

Oysters, like many other marine species, are characterized by very high fertilities. Natural or hatchery populations, often present very low Ne/N ratios (Hedgecock, 1994). It can be explained by very high variances of their breeding success (Li and Hedgecock, 1998), the effective population size being estimated through the temporal variance of the allelic frequencies in natural populations (Hedgecock et al., 1992) or, more directly, through the estimated number of founders in hatchery populations (Launey, 1998). This document is a presentation of the results of a number of experiments, realised in the scope of the European program "GENEPHYS" (FAIR 95-421), and using microsatellite markers to analyse the parental contributions in the flat oyster Crassostrea... |

Background: Bacterial infections are common in bivalve larvae and can lead to significant mortality, notably in hatcheries. Numerous studies have identified the pathogenic bacteria involved in such mortalities, but physiological changes associated with pathogen exposure at larval stage are still poorly understood. In the present study, we used an integrative approach including physiological, enzymatic, biochemical, and molecular analyses to investigate changes in energy metabolism, lipid remodelling, cellular stress, and immune status of Crassostrea gigas larvae subjected to experimental infection with the pathogenic bacteria Vibrio coralliilyticus. Findings: Our results showed that V. coralliilyticus exposure induced (1) limited but significant increase... |

Pathogenic bacteria are known to be one of the main factors affecting Pecten maximus larval survival in hatcheries. As a result, juvenile production often relies on the use of antibiotics during larval culture. However, limitations of the usage of chemicals such as chloramphenicol in aquaculture have been reinforced due to their negative environmental impact and alternatives are accordingly needed. Thus, the importance of bacterial transfer from oocytes to larvae has been studied here as well as procedures to limit larval mortality in P. maximus. In order to reduce bacterial contamination during larval development, we focused on two periods, broodstock conditioning and post-fertilization. The animals were conditioned for 2 months with two erythromycin... |

The taxonomic status of the cupped oysters Crassostrea angulata and C. gigas has received considerable attention in the last decades. Based on larval shell morphology, experimental hybridization, allozymes and nuclear DNA studies several authors have considered these two taxa as being synonymous. However, mitochondrial data showed clear genetic differences between the two taxa. In addition, microsatellite-based studies and cytogenetic studies have also provided evidence that supports their differentiation. Considerable differences have also been observed at the phenotypic level in terms of growth rate and ecophysiological parameters. In the present study, C. angulata from Sado estuary (Portugal) and C. gigas from Seudre estuary (France) were collected and... |

A comparative study of the growth performances in cupped oysters Crassostrea gigas, Crassostrea angulata and their hybrids was conducted from March to October 1999 in a controlled farm with phytoplankton (Skeletonema costatum) input. The individuals, of the same age, were from genitors which had been identified through a marker analysis of the mitochondrial genome (Boudry et al, 1998). As a consequence, two pure offspring were obtained through the crossing of Portuguese oyster C. angulata (AA) genitors and the crossing of Japanese oyster C. gigas (GG) genitors. Two other offsprings were produced through the hybrid crossing of C. gigas female genitors with C. angulata (GA) male genitors on one hand, and C. angulata female genitors with C. gigas (AG) male... |

To date, the most significant genetic improvement for the production of Pacific oyster (Crassostrea gigas) has been obtained through the breeding of triploids, especially since the development of tetraploids. Quantitative genetics studies suggest that significant gains, for disease resistance or for other traits of aquacultural interest, could be obtained in diploids using this approach. However, the limited extent of hatchery-propagation (versus natural recruitment) and/or various technical difficulties and biological characteristics of the species have retarded the development of selective breeding programs. Recently, in the U.S.A., Australia and New Zealand, countries where hatcheries are a major source of C. gigas juveniles, family-based selective... |

The UE project "GENEPHYS", initiated by the IFREMER laboratory GAP-La Tremblade (Génétique Aquaculture et Pathologie) and CREMA-L'Houmeau (Centre de Recherche en Ecologie marine et Aquaculture), aims to establish the relationships between physiological traits involved in growth (oxygen consumption, absorption efficiency, scope for growth) and their genetic bases (determinism, variability within and among populations) in cupped oyster Crassostrea gigas. The main objectives are : 1. To express the maximum variability of growth, both at the larval stage and at the adult stage, in a oyster population originating from parents of various origins. 2. To check the persistence over the time, of growth performance and related physiological functions. 3. To explain... |

Twenty male and 20 female parental oysters, originating from four sites located along the French Atlantic coast, were crossed together. The 400 crosses were performed separately and then pooled to give a batch of larvae with a large genetic base. Successive sieving after 17 days at 23°C enabled the separation of the largest larvae from the batch. These larvae (i.e., sieving groups) were left to metamorphose and fix onto flat PVC collectors changed daily. Four groups of larvae representing distinctly different growth rates were successively separated during the attachment and metamorphosis of the whole population. This lasted 12 days, from the 17th day to the 29th after fertilisation. A part of each sieving group was settled separately as replicates on... |

Background: Triploidy can occur in many animal species but is often lethal. Among invertebrates, amphibians and fishes, triploids are viable although often sterile or infertile. Most triploids of the Pacific oyster Crassostrea gigas are almost sterile (named "3n beta'') yet a low but significant proportion show an advanced gametogenesis (named "3n alpha''). These oysters thus constitute an interesting model to study the effect of triploidy on germ cell development. We used microarrays to compare the gonad transcriptomes of diploid 2n and the abovementioned triploid 3n beta and 3n alpha male and female oysters throughout gametogenesis. Results: All triploids displayed an upregulation of genes related to DNA repair and apoptosis and a downregulation of genes... |

We describe a simple protocol to reduce the number of cloning reactions of nuclear DNA sequences in population genetic studies of diploid organisms. Cloning is a necessary step to obtain correct haplotypes in such organisms, and, while traditional methods are efficient at cloning together many genes of a single individual, population geneticists rather need to clone the same locus in many individuals. Our method consists of marking individual sequences during the polymerase chain reaction (PCR) using 5'-tailed primers with small polynucleotide tags. PCR products are mixed together before the cloning reaction and clones are sequenced with universal plasmid primers. The individual from which a sequence comes from is identified by the tag sequences upstream... |

Background Originating from Northeast Asia, the Pacific oyster Crassostrea gigas has been introduced into a large number of countries for aquaculture purpose. Following introduction, the Pacific oyster has turned into an invasive species in an increasing number of coastal areas, notably recently in Northern Europe. Methods To explore potential adaptation of reproductive traits in populations with different histories, we set up a common garden experiment based on the comparison of progenies from two populations of Pacific oyster sampled in France and Denmark and their hybrids. Sex ratio, condition index and microarray gene expression in gonads, were analyzed in each progeny (n = 60). Results A female-biased sex-ratio and a higher condition index were... |

Reproduction was studied in triploid Crassostrea gigas, by crossing gametes from diploid and triploid parents.Triploids are able to produce viable gametes and their resulting progeny consisted in diploids, aneuploids and polyploids. Gametes from triploid parents frequently led to abnormal polar body formation, even before caryogamy occurred. A high frequency of aneuploids was observed, related to the missegregation of chromosomes during meiosis in triploids. This might be due to the observed incomplete synapsis in triploid eggs during Prophase I. The proportion of aneuploids decreased during larval development, reflecting differential mortality between ploidy levels. Due to limited gonadic development and high larval mortality rates, the reproductive... |

Herpes-like viral infections have been reported in different bivalve mollusc species throughout the world. High mortalities among hatchery-reared larvae and juveniles of different bivalve species have been associated often with such infections. The diagnosis of herpes-like viruses in bivalve molluscs has been performed traditionally by light and transmission electron microscopy. The genome sequencing of one of these viruses, oyster herpesvirus 1 (OsHV-1), allowed the development of DNA-based diagnostic techniques. The polymerase chain reaction (PCR) has been used for the detection of OsHV-1 DNA in bivalve molluscs at different development stages. In addition, the PCR used for detection of OsHV-1 has also allowed the amplification of DNA from an OsHV-1... |
