AMP-deaminase in elasmobranch fish: A comparative histochemical and enzymatic study ArchiMer

AMP-deaminase activity was measured in white muscle from a wide range of fish, including one cyclostome, 13 chondrosteans, and one teleost to elucidate the pattern of the AMP-deaminase activity in white muscle of fish. Compared to a mammalian (rat) muscle extract, low enzyme activities are found in the cyclostome and two elasmobranchs from two families (Scyliorhinidae, Hexanchidae). In contrast, higher AMP-deaminase activities, similar to mammals, are expressed in Squalidae, all families of skates, Chimaeridae and in the teleostean fish. A polyclonal rabbit anti-human lysozyme (Dako) was applied on histological sections from bivalve mollusc and revealed with a mouse anti-rabbit IgG fluorescein conjugated antibody to localise cellular lysozyme in tissues where its activity had previously detected by enzymatic techniques. Labelling occurred in the cytoplasm of epithelial cells covering the body. This is in agreement with the intracellular distribution of this enzyme. In the Lucinid Lucinella divaricata gills, the cytoplasm of bacteriocytes was also labelled. This suggests that antibacterial activity occurred in these cells. Effects of an environmentally relevant concentration of diuron on oyster genitors during gametogenesis: responses of early molecular and cellular markers and physiological impacts ArchiMer

Genitors of the Pacific oyster Crassostrea gigas were submitted during gametogenesis to a short pulse exposure to the herbicide diuron at a realistic environmental concentration. Histological analysis showed no effect of diuron on gametogenesis course, sex ratio and reproductive effort. A non-significant increase in testosterone and progesterone levels was observed in genitors exposed to the herbicide. At cell level, diuron exposure was shown to modulate the phagocytic activity of circulating hemocytes. Evaluating the histological-based condition of wild collected larval fish: A synthetic approach applied to common sole (Solea solea) ArchiMer

In the present study, the histological-based condition of common sole (Solea solea) larvae in the English Channel was evaluated to assess the relevance of experimentally-calibrated indices for wild collected specimens. Based on topographical observation of foregut, midgut, hindgut, liver and pancreas, each of the 202 larvae analysed was attributed one of the six grades of a condition index that was developed for sole in the nineties. In parallel, a synthetic table of 20 criteria historically used as condition indicators was created from an extensive review of the literature. The Y organ appears during the embryonic development of the prawn (P. serratus ) at a stage approximately C, according to Richard's definition (1974). 3 weeks separate this stage from hatching at a temperature of 20 degree C. The organ formation is realized over 48 hours and is marked by an histological modification and a small mitotic crisis in the epidermal tissue from which it differentiates. Its metameric localization is mainly maxillular.

We evaluated the effects of dietary lipid class (phospholipid vs. neutral lipid) and level of n-3 long-chain PUFA (LC-PUFA) on the growth, digestive enzymatic activity, and histological organization of the intestine and liver in European sea bass larvae. Fish were fed from the onset of exogenous feeding at 7 to 37 d post-hatch with five isoproteic and isolipidic compound diets with different levels of EPA and DHA. The ontogenetic development of the gut and accessory organs in large yellow croaker Pseudosciaena crocea was investigated using light microscopy from hatching up to the juvenile stage (40 days post hatch, dph). At 3 dph (mean +/- S.D., 4.1 +/- 0.1 mm total length, L-T), coinciding with the buccopharynx opening, larvae started to feed exogenously, and the gut consisted of a well-developed buccopharynx, a partially-differentiated oesophagus and an intestine divided in three regions (anterior intestine, intermediate intestine and rectum). Here, the relevance of the widely used gonadosomatic index (GSI) as proxy of giant clam gonad maturation is tested with a large dataset (n = 265). Azadinium spinosum (Elbrächter and Tillmann), a small marine dinoflagellate, has been recently described as a de novo producer of azaspiracid-1 and -2 (AZA1 and -2) diarrhoeic toxins. A culture of A. spinosum was established in our laboratory and optimised for pilot-scale production of this organism, to evaluate and understand AZA1 and -2 accumulation and biotransformation in blue mussels (Mytilus edulis) fed with A. spinosum. We evaluated differences in productive traits and disease susceptibility among Ostrea edulis stocks. We produced 4 to 5 families from each of 4 oyster populations (Irish, Greek and 2 Galician) in a hatchery. Spat corresponding to 19 different families were transferred to a raft in the Ria de Arousa (Galicia, Spain) for grow-out. Samples of each family were histologically processed every month for 2 yr. Neoplasia detection in Macoma balthica from the Gulf of Gdansk: comparison of flow cytometry, histology and chromosome analysis ArchiMer

A flow cytometry protocol was applied for the detection of neoplasia in Macoma balthica L. from the Gulf of Gdansk (Baltic Sea, Poland). A simple method, based on an osmotic shock, was used to permeabilise gill cells. The cytometric pattern of normal clams consisted of 2 peaks, a major peak B and a smaller peak C. The cytometric pattern of affected clams consisted of 2 peaks named B' and C'.
