Seedling length in wheat determined by image processing using mathematical tools Rev. Ciênc. Agron.
Brunes,André Pich; Araújo,Ádamo de Souza; Dias,Letícia Winke; Villela,Francisco Amaral; Aumonde,Tiago Zanatta.
ABSTRACT Seed vigour is determined by a series of laboratory tests, many of these laborious and time-consuming. In view of this, the present study aimed to compare the determination of seedling length in wheat using image processing, with the traditional method, as well as the ability of the method to determine seed vigour. The experimental design was completely randomised, with five replications comprising a single-factor experimental scheme. The data were analysed for normality by the Shapiro-Wilk test and variance analysis using ANOVA at 5% probability. The response variables were correlated using Pearson's test, and the methodologies for determining the length of the shoots and roots, by measuring with a graduated rule and by image processing, were...

Romualdo,Liliane Maria; Luz,Pedro Henrique de Cerqueira; Baesso,Murilo Mesquita; Devechio,Fernanda de Fatima da Silva; Bet,Jessica Angela.
ABSTRACT Image analysis can provide information extracted from the leaves of crops, and contribute to early identification of nutrient deficiency. The objective of this study was to recognize nutritional nitrogen (N) patterns in maize plants, at the V4 and V7 stages, using digital image analysis based on spectral indexes. The experiment was carried out in a greenhouse under hydroponic cultivation. Treatments consisted of a completely randomized design, in a 4 × 2 factorial arrangement, with four replications. The factors were constituted by the doses of N (0; 3.0; 6.0 e 15 mmol L-1) combined at V4 and V7. In each stage, digital images were taken of leaf blades, with subsequent chemical composition and image analysis. For image recognition and...

Digital-image analysis to predict weight and yields of boneless subprimal beef cuts Scientia Agricola
Teira,Gustavo Adolfo; Tinois,Eduardo; Lotufo,Roberto de Alencar; Felício,Pedro Eduardo de.
For several decades, beef carcass evaluation for grading or research purposes has relied upon subjective visual scores, and manually taken measurements, but in recent times there has been a growing interest in new technologies capable of improving accuracy of estimates. Equations to predict weight and yield of beef pistol subprimal cuts were developed in this work using digital image analysis (VIA) of the 12th rib steak. Equations to predict total pistol subprimal cuts weight (CUTS) had coefficients of determination (CD) of 0.84, or 0.87 to 0.88, when the independent variables were the VIA parameters and the half carcass weight (HC) or the total pistol weight (TP), respectively. The predicted values for the total seven subprimal cuts, as a percentage of...

A short review: Comparisons of high-throughput phenotyping methods for detecting drought tolerance Scientia Agricola
Kim,Jaeyoung; Kim,Ki-Seung; Kim,Yoonha; Chung,Yong Suk.
ABSTRACT Drought is a major threat worldwide for crop production, especially due to the rapid climate changes. Current drought solutions involve improving irrigation system, rainwater harvesting, damming, cloud seeding, and changes of cultivation methods. Despite effective, each solution has economic, environmental, and temporal drawbacks. Among all solutions, the most effective, inexpensive and manageable method is the use of drought-tolerant cultivars via plant breeding. However, conventional plant breeding is a time-consuming and laborious task, especially for phenotypic data acquisition of target traits of numerous progenies. High-throughput phenotyping (HTP) is a recently developed method and has potential to overcome the mentioned issues. HTP offers...

Swingle citrumelo seed vigor and storability associated with fruit maturity classes based on RGB parameters Scientia Agricola
Gomes-Junior,Francisco Guilhien; Arruda,Natália; Marcos-Filho,Julio.
ABSTRACT: Citrus seeds normally have low storability. Identifying an optimal fruit harvest time for production of high vigor seeds is important for nurseries; however, identifying this stage for Swingle citrumelo fruits has been based only on visual color examination, and research related to this parameter has been inconsistent. The main objective of this study was to evaluate a red-green-blue (RGB) color measurement system for successful identification of Swingle citrumelo fruits possessing seeds with maximum physiological potential and storability. Fruits were harvested at three ripening stages identified as green (G), greenish-yellow (GY), and yellow (Y) pericarp, photographed, and the images processed using ImageJ software. Data were expressed as the...

PhenoRoots: an inexpensive non-invasive phenotyping system to assess the variability of the root system architecture Scientia Agricola
Martins,Saulo Muniz; Brito,Giovani Greigh de; Gonçalves,Washington da Conceição; Tripode,Bruna Mendes Diniz; Lartaud,Marc; Duarte,João Batista; Morello,Camilo de Lelis; Giband,Marc.
ABSTRACT: The root system architecture (RSA) of plants and its functioning play a fundamental role in a number of plant growth mechanisms including water and nutrient uptake. Optimization of the RSA is important for stable and increased plant productivity under adverse conditions. Despite its great importance, studying the RSA is notoriously laborious because of the difficulty of accessing the rooting system of plants. We developed a root phenotyping platform, PhenoRoots, which allows for the non-invasive study of plant RSA. The system was built using inexpensive material and was designed to provide medium throughput. Substrate or soil-filled rhizotrons are used to grow plantlets, whose roots are directly visible through a glass plate. An experiment...

Recent approaches for bell pepper seed vigor testing Agronomy
Dias, Marcos Altomani Neves; Mondo, Vitor Henrique Vaz; Cicero, Silvio Moure.
Seed vigor is a major attribute in the determination of whether a seed lot can be commercialized. Different physiological tests are used to assess seed vigor and are generally based on different concepts, which could lead to variable interpretations, particularly when comparing lots presenting different vigor levels. This study was conducted to compare the different approaches used to evaluate bell pepper seed vigor and verify the applicability of the Seed Vigor Imaging System (SVIS) for this purpose. The experiment was performed using four seed lots of bell pepper hybrids in two periods, and the evaluations involved germination, seedling emergence, seedling emergence speed index, saturated salt accelerated aging and image analysis. The use of seedling...

Gamma-ray computed tomography to evaluate changes in the structure of a clayey soil due to agricultural traffic - doi: 10.4025/actasciagron.v33i3.4727 Agronomy
Pires, Luiz Fernando; Universidade Estadual de Ponta Grossa.
A first generation gamma-ray computed tomography (CT) equipped with a 137Cs source was used to investigate changes in soil structure due to agricultural machinery traffic. Core samples with volume of 55 cm3 were collected at the surface (0-10 cm) of a Brazilian clayey soil. Significant alterations in soil structure occurred after two passages of tractor wheels, with important decreases in soil porosity. CT data allowed a continuous analysis of soil bulk density and also soil porosity distribution for 1.25 and 1.20 mm layers, which cannot be provided by traditional methods commonly used in soil physics.

Knockdown of apoptosis-inducing factor disrupts function of respiratory complex I Biocell
Varecha,Miroslav; Páclová,Daniela; Procházková,Jirina; Matula,Pavel; Cmarko,Dusan; Kozubek,Michal.
Recent findings suggest that apoptotic protein apoptosis-inducing factor (AIF) may also play an important non-apoptotic function inside mitochondria. AIF was proposed to be an important component of respiratory chain complex I that is the major producer of superoxide radical. The possible role of AIF is still controversial. Superoxide production could be used as a valuable measure of complex I function, because the majority of superoxide is produced there. Therefore, we employed superoxide-specific mitochondrial fluorescence dye for detection of superoxide production. We studied an impact of AIF knockdown on function of mitochondrial complex I by analyzing superoxide production in selected cell lines. Our results show that tumoral telomerase-positive (TP)...

Quantification of Japanese Quail Eggshell Colour by Image Analysis Biol. Res.
SEZER,METIN; TEKELIOGLU,OGUZ.
The Japanese quail lays eggs with colourful and patterned shells which make the eggshell colour difficult to classify. In this study, the method of measuring colour of patchy eggs using image analyses and its power to discriminate among individual variation were established. Estimated repeatability for egg colour and proportion of patterned areas was high (>0.58), suggesting intermedíate or high heritability of eggshell colour characteristics. Three components have been identified as significant in discriminant function analysis. These three components explained 91.4% of the total variance in egg colour characteristics. In cluster analysis, 78.3% of the eggs that were collected from 15 females were correctly classified. This study indicates that...

Relationship between DAPI-fluorescence fading and nuclear DNA content: An alternative method to DNA quantification? Biol. Res.
GALLARDO-ESCÁRATE,CRISTIAN; ÁLVAREZ-BORREGO,JOSUÉ; VON BRAND,ELISABETH; DUPRÉ,ENRIQUE; DEL RÍO-PORTILLA,MIGUEL ÁNGEL.
In observations by confocal or conventional fluorescence microscopy, important factors should be considered in order to obtain accurate images. One of them, such as the fluorescence bleaching from highest intensity to lowest signal of fluorescence is a common problem with several DNA fluorochromes and especially for DAPI stain. The fluorescence of DAPI fades rapidly when it is exposed to UV light, under optimal conditions of observation. Although the fading process can be retarded using a mounting medium with antifading reagents, the photochemical process underlying the fluorescence decay has not yet been fully explained. In addition, no relationship between fluorescence fading and nuclear DNA content has been tested. In order to test this relationship, we...
