The French ostreiculture history highlights the fragility of its production caused by stocks overexploitation and diseases. Particularly, the flat oyster production decreased a lot after the appearance of two parasitic diseases: marteilliose and bonamiose. The bonamiose is due to a parasite Bonamia ostreae. The study of interactions between the parasite and oysters hemocytes with flow cytometry allowed to identify modifications of hemocytes activities wh en active parasite is present. Les études de génétique des populations des bivalves marins ont fréquemment mit en évidence une relation hétérozygotie / valeur sélective, et un fort déficit en hétérozygotes. Parmi les hypothèses explicatives de ces deux phénomènes, la création d'individus consanguins dans les populations est particulièrement intéressante à tester. The fact that molluscs from the Mediterranean have benefited from a privileged zoosanitary condition compared to those of the other French coasts has for a long time limited the control of farms and natural fields in this area. It is only in the early 70's, because of the pathological problems and risks observed in other areas that Mediterranean molluscs started to be stictly under zoosanitory control. Variété "bicolor" de l'huître plate : Ostrea edulis ArchiMer

Bien que la culture de l'huître portugaise ne soit pas autorisée dans le Bassin Morbihannais, jusqu'ici réservé à la production et à l'élevage de la seule huître plate, Ostrea edulis, il n'est pas rare de trouver chaque année, parmi les jeunes plates qui garnissent les collecteurs, quelques exemplaires de Gruyphea angulata. Celles- sont toujours peu nombreuses : 1 pour 50000 ou même 10 000 huîtres plates. Si les individus adultes de l'une et l'autre espèces peuvent être facilement distingués par leur seule conformation extérieure, cette différentiation n'est pas toujours aussi aisée sur les très jeunes naissains fixés, en raison de l'existence de jeunes plates présentant, au premier examen, des ressemblances marquées avec de jeunes portugaises.

A first generation genetic linkage map of the European flat oyster Ostrea edulis (L.) based on AFLP and microsatellite markers ArchiMer

This study presents the first genetic linkage map for the European flat oyster Ostrea edulis. AFLP and 20 microsatellite markers were genotyped in a three-generation pedigree comprised of 2 grand-parents, 2 parents and 92 progeny. Chi-square goodness of fit tests revealed high segregation distortion, which was significant for 32.8% of markers. Sixteen microsatellites and 235 AFLPs (170 AFLPs type 1:1 and 65 AFLPs type 3:1) were used to build sex-specific linkage maps using CriMap software. La structuration géographique des populations naturelles d'huître plate (Ostrea edulis) a été étudiée à partir de l'analyse du polymorphisme de conformation simple brin (SSCP), d'un fragment d'environ 300 pb du gène de l'ARNr mitochondrial 12S. les échantillons (13 localités, 507 individus) utilisés avaient fait l'objet d'une étude antérieure de structuration des populations avec 5 locus microsatellites. Il ont été analysés ici dans le but d'une comparaison des structurations observées sur marqueurs mitochondrial et nucléaire. Validation in commercial conditions of the response to selection of the European flat oyster Ostrea edulis for resistance to Bonamia ostreae ArchiMer

Since the outbreaks of the parasitosis with Marteilia refringens and Bonamia ostreae in the last 60's, the French production of the European flat oyster dropped from 20000 mT to less than 1500 mT currently, leaving the French shellfish farming in situation of virtually monoculture of the Pacific cupped oyster Crassostrea gigas. To sustain this traditional culture, Ifremer has developed, at the experimental level, a genetic selection for a better tolerance to Bonamia ostreae which led to an improvement of survivals. Characterization of 27 microsatellite loci in the European flat oyster Ostrea edulis ArchiMer

The flat oyster Ostrea edulis is native to Europe and populations have been severely depleted by the parasite Bonamia ostreae since the 1980s. Additional genetic markers are required to improve population genetics study and linkage map development for selection for B. ostrea-resistance in this species. Here, we characterized 27 novel microsatellite loci for O. edulis. Number of alleles per locus ranged from 6 to 25 and observed heterozygosity between 0.375 and 1. Null alleles were suggested at a few loci but most loci were in Hardy-Weinberg agreement enabling their reliable use in further population and mapping genetics approaches.

The history of French oyster culture consists of a succession of developmental phases using different species, followed by collapses caused by diseases. The indigenous species Ostrea edulis was replaced first with Crassostrea angulata, then C. gigas. France is now the top producer and consumer of oysters in Europe, producing around 120,000 t of the cupped oyster C. gigas annually, and an additional 1500 t of the flat oyster O. edulis. Infectious diseases are currently recognised as a major limiting factor to development and sustainability of aquaculture of molluscs. The study is focused on the European flat oyster, Ostrea edulis and its paramyxean parasite Marteilia refringens. Results are contributing to the prevention of risk associated to infected stocks imports from endemic zones, by developing molecular diagnostic tools that are specific, sensible and reliable.
