Reserve carbohydrates and lipids from the seeds of four tropical tree species with different sensitivity to desiccation BABT
Mello,Juliana Iura de Oliveira; Barbedo,Claudio José; Salatino,Antonio; Figueiredo-Ribeiro,Rita de Cássia Leone.
Considering the importance of water content for the conservation and storage of seeds, and the involvement of soluble carbohydrates and lipids for embryo development, a comparative study was carried out among the seeds of Inga vera (ingá), Eugenia uniflora (pitanga), both classified as recalcitrant, and Caesalpinia echinata (brazilwood) and Erythrina speciosa (mulungu), considered as orthodox seeds. Low concentrations of cyclitols (0.3-0.5%), raffinose family oligosaccharides (ca. 0.05%) and unsaturated fatty acids (0-19%) were found in the seeds of ingá and pitanga, while larger amounts of cyclitols (2-3%) and raffinose (4.6-13%) were found in brazilwood and mulungu, respectively. These results, in addition to higher proportions of unsaturated fatty acids...

Glycemic index: effect of food storage under low temperature BABT
Carreira,Marina Cassab; Lajolo,Franco Maria; Menezes,Elizabete Wenzel de.
This study was carried out to evaluate the influence of food storage under low temperature (-20ºC) and the resistant starch formation, both on the glycemic index (GI). The GI of only cooked and cooked and stored foods under -20ºC for 30 days was evaluated in short-term tests with humans. Significant increase on the RS content was evidenced for all the stored foods. The food storage resulted in a significant decrease on the GI of beans and chick-peas; the GI of pasta remained the same and the GI of corn meal increased. Thus, the RS formation showed reduced influence on the glycemic index. The storage of starchy foods under low temperature can collaborate to the RS intake but its effect on the GI will depend on the characteristics of the carbohydrates of...

Extracellular amylase production of a thermotolerant Fusarium sp: isolated from Eastern Nigerian soil BABT
Nwagu,Tochukwu Nwamaka; Okolo,Bartholomew Ndubuisi.
In this work, an α- amylase producing Fusarium sp. was isolated from the soil at 50 0C. Growth and enzyme production occurred at 30, 45 and 55 ºC. Soybean meal at 1% concentration, supplemented with 0.2% NH4Cl and 2.5% corn starch elicited the highest amylase yield. Optimum pH for the enzyme was pH 6.5 which retained over 60% of its activity after 24 h incubation at the pH range of 4.5-7.0. The enzyme showed high activity from 40-70 0C with optimal activity at 50 ºC and 78% activity was retained after incubation at 70 ºC for 30min. Catalytic function of the crude amylase was stimulated by Mg (136%), Ca (118%) and Zn (118%) at 2mM concentrations. The enzyme hydrolyzed cassava, potato and yam starches effectively.

Characterization of native and oxidized starches of two varieties of Peruvian carrot (Arracacia xanthorrhiza, B.) from two production areas of Paraná state, Brazil BABT
Matsuguma,Luciana Shizue; Lacerda,Luiz Gustavo; Schnitzler,Egon; Carvalho Filho,Marco Aurélio da Silva; Franco,Célia Maria Landi; Demiate,Ivo Mottin.
Two commercial varieties of Peruvian carrot ('Amarela de Carandaí' and 'Senador Amaral') were processed into flour, starch and bagasse and chemically evaluated. The starch was extracted, modified with H2O2 and characterized by the physicochemical methods. By using the methylene blue dyeing, the granules of the modified starches showed intense blue color. The carboxyl content, the reducing power and the amount of the water liberated from the pastes after the freeze-thawing were higher for the oxidized starches and their pastes were clearer than those of the native starches of the two varieties from the two production areas. The RVA viscoamylography showed that the modified starches had lower viscosities with differences between the varieties. In the thermal...

Starch levels in refrigerated and frozen chicken based meat products BABT
Labanca,Renata A.; Silva,Cristiane M. G.; Glória,M. Beatriz A..
The levels of starch were determined in chicken based meat products commercialized in Belo Horizonte, MG, Brazil, from December 1996 to March 1997. Samples were analyzed for moisture and starch contents. Starch was acid hydrolyzed and the resulting glucose was determined at 620 nm after reaction with anthrone. The method was observed to be accurate (92.7% recovery), precise (CV = 3.0%), sensitive (quantification limit = 1.25 g/100 g) and simple in the determination of starch in meat products. Among products analyzed, starch was detected in 100% of meat balls and nuggets samples, in 60% of the sausage, 50% of the bologna and 30% of the frankfurter. Starch was not detected in hamburger samples analyzed. Higher mean starch levels were found in nuggets (14.85...

Starch degradation and nutrition value improvement in corn grits by solid state fermentation technique with Coriolus versicolor BJM
Huang,Mian; Zhang,Song.
The study was conducted to evaluate effect of Coriolus versicolor mycelia on degrading starch and improving nutrition value in corn grits through solid state fermentation technique. The results showed that using soybean meal as a nitrogen source, α-amylase secreted from C. versicolor expressed 407.25U/g of activity, leading to 45.15% of starch degraded. The activity grew with fermentation time until the 15th day, after that the amylase was deactivated rapidly. An orthogonal experiment designed for the study illustrated that degradation rate of starch in corn grits attained to maximum, 50.51%, when 100g of corn grits, added 16g of soybean meal, were fermented by C. versicolor for 12 days, in an initial pH 5.5. After fermenting, compared to the nonfermented...

Application of microbial α-amylase in industry - A review BJM
Souza,Paula Monteiro de; Magalhães,Pérola de Oliveira e.
Amylases are one of the main enzymes used in industry. Such enzymes hydrolyze the starch molecules into polymers composed of glucose units. Amylases have potential application in a wide number of industrial processes such as food, fermentation and pharmaceutical industries. α-Amylases can be obtained from plants, animals and microorganisms. However, enzymes from fungal and bacterial sources have dominated applications in industrial sectors. The production of α-amylase is essential for conversion of starches into oligosaccharides. Starch is an important constituent of the human diet and is a major storage product of many economically important crops such as wheat, rice, maize, tapioca, and potato. Starch-converting enzymes are used in the production of...

Synergistic action of starch and honey against Candida albicans in correlation with diastase number BJM
Boukraa,Laïd; Benbarek,Hama; Moussa,Ahmed.
To evaluate the synergistic action of starch on the antifungal activity of honey, a comparative method of adding honey with and without starch to culture media was used. Candida albicans has been used to determine the minimum inhibitory concentration (MIC) of five varieties of honey. In a second step, lower concentrations of honey than the MIC were incubated with a set of concentrations of starch added to media to determine the minimum synergistic inhibitory concentration (MSIC). The MIC for the five varieties of honey without starch against C. albicans ranged between 40% and 45% (v/v). When starch was incubated with honey and then added to media, a MIC drop has been noticed with each variety. It ranged between 7% and 25%. A negative correlation has been...

Koga,Luciana Numata; Fenelon,Vanderson Carvalho; Miyoshi,Juliana Harumi; Moriwaki,Cristiane; Wessel,Kamila Byanca Baldin; Mangolim,Camila Sampaio; Matioli,Graciette.
A repetitive batch process was employed followed by membrane ultrafiltration system to produce low-cost cyclodextrins (CDs) using commercial enzymes Toruzyme® cyclomaltodextrin glucanotransferase (CGTase) and its kinetic parameters were determined. The ultrafiltration system enabled the removalof inhibitory products from the reaction medium, allowing the enzyme to be recovered for reuse. A 10 kDa membrane was used to separate the different CDs produced by the CGTase. The substrates evaluated were maltodextrin, corn starch and cassava starch at 5, 10 and 15% (w/V), in the presence and absence of 10% (V/V) ethanol. After reaction for 132 h, 10% (w/V) cassava starch in the presence of ethanol provided the best results with 32.1 mg/mL of β-CD. Maximum...
