Pressure and cold shock induction of meiotic gynogenesis and triploidy in the European sea bass, Dicentrarchus labrax L.: relative efficiency of methods and parental variability ArchiMer

The optimal conditions for the retention of the second polar body in sea bass eggs were investigated by altering the timing, intensity and duration of application of pressure and cold shocks. Treatment optima for cold shocks were 0-1 degrees C for 15-20 min at 5 min after fertilisation (a.f.) and 8500 psi for 2 min at 6 min a.f. for pressure shocks. Meiogenesis was obtained by fertilising eggs with UV-irradiated homologous sperm (32,000 erg mm(-2)) and pressure or cold shocking eggs as above. 100% triploidy was induced following definition of liable periods for the disruption of the meiotic spindle obtained in gynogenesis. Particulate matter and organic carbon budgets for the Gulf of Lions (NW Mediterranean) ArchiMer

An analysis of sedimentological, sediment trap and hydrological data was performed to investigate the transport and fate of particulate matter in the Gulf of Lions. The sedimentological properties outline the major sedimentary units of the shelf (Rhone prodelta, mid-shelf mud belt, outer shelf) and slope. The geometry of these sedimentary units and the southwestwards increase of particulate fluxes on the slope highlight the influence of the general cyclonic circulation on the dispersion of land-derived particulate matter. The Mediterranean Thau Lagoon is an important oyster farming area in Europe. Oyster growth rates are among the highest in France, although chlorophyll a concentration is low. Previous studies have demonstrated that picophytoplankton, nano-microphytoplankton, dinoflagellates and loricate ciliates such as tintinnids are abundant. However, heterotrophic flagellates and aloricate ciliates have not been investigated. Hepatic glucose-6-phosphatase (G6Pase) plays an important role in glucose metabolism because it catalyzes the release of glucose to the circulatory system in the processes of glycogenolysis and gluconeogenesis. The present study was initiated to analyze the regulation of hepatic G6Pase expression by dietary carbohydrates in rainbow trout. The first step in our study was the identification of a partial G6Pase cDNA in rainbow trout that was highly homologous to that of mammals. An overview of the FADs fishery in the Mediterranean Sea ArchiMer

The Mediterranean small-scale fleet is highly adaptive, showing a dynamic fishing intensity and strategy, and exploiting seasonal abundant resources. In this area, the aggregatory behaviour under floats of juvenile fish has been used since ancient times to exploit oceanic migratory species such as dolphinfish (Coryphaena hippurus), using Fish Aggregating anchored Devices (fads). A total of 2 300 boats are engaged in this fishery from August to December, the main fishing areas being those around Malta, Tunisia, Sicily and Majorca. A white spot disease-like syndrome in the Pacific blue shrimp (Litopenaeus stylirostris) as a form of bacterial shell disease ArchiMer

In May 1997, some white lesions evoking the white spot syndrome disease were observed in Litopenaeus stylirostris broodstock in New Caledonia. The occurrence of these lesions was neither associated with mortality, nor with histological evidence of white spot syndrome baculovirus (WSBV), The evidence suggests that these lesions result from a form of bacterial disease and are associated with an increased bacterial flora on the outer surface of the cuticle, as well as an increased incidence throughout the molt cycle. A microscopic observation of these lesions allows them to be differentiated from WSBV-associated lesions.

Differential distribution and defence involvement of antimicrobial peptides in mussel ArchiMer

In previous papers, we characterised 3 types of 4-kDa, cysteine-rich, cationic antimicrobial peptides: MGDs (for Mytilus galloprovincialis defensins), mytilins and myticins, which are abundant in the mussel hemocytes. In the present work, me revealed a differential distribution of the cells expressing the different genes. In addition, using confocal and electron microscopy, we confirmed that defensins and mytilins were partially located in different sub-types of circulating hemocytes although the peptides can be located in the same cell, and even in the same granule. Marteilia refringens est un protozoaire parasite de l'huître plate Ostrea edulis, à l'origine de mortalités dramatiques en élevage depuis les années 1970. En zone endémique, le seul moyen de contrôle de la marteiliose, est une gestion du risque de contamination. Dans ce cadre, la connaissance du cycle parasitaire de Marteilia refringens est primordiale. Les travaux réalisés à ce sujet ont permis de poser l'hypothèse d'un cycle hétéroxène. Le parasite a été détecté par PCR puis localisé au sein des tissus par hybridation in situ au sein d'une espèce de copépode Calanoide prélevée dans une claire ostréicole. Au niveau du littoral charentais, et en particulier pour la conchyliculture, le fleuve Charente représente le principal apport d'eaux douces, et donc d 'éléments nutritifs, essentiels au développement de la production primaire et des larves d'huîtres. Influence of water temperature and oxygenation on the aerobic metabolic scope of Atlantic cod (Gadus morhua) ArchiMer

Environmental influences (temperature and oxygenation) on cod metabolism and their impact on the ecology of this species were investigated. Limiting oxygen concentration curves (O-2 level ranging between 15 and 100% air saturation) were established at 2, 5 and 10 degreesC. The standard metabolic rate (SMR), the maximum metabolic rate and the metabolic score were then modelled as functions of temperature and/or oxygen saturation. The mean SMR at 2, 5 and 10 degreesC were 19.8 +/- 4.9, 30.8 +/- 6.1 and 54.3 +/- 4.1 mg O-2 h(-1) kg(-1). respectively. Between 2 and 5 degreesC, the active metabolic rate of cod almost doubled from 65 to 120 mg O-2 h(-1) kg(-1), to reach 177 mg O-2 h(-1) kg(-1) at 10 degreesC. Cette étude s'inscrit dans le cadre du programme expérimental de réhabilitation des fossés à poisson initié au CREAA (1997-2000). Ce programme répond à une demande régionale pour la valorisation des fossés à poissons. Cet écosystème fait partie du patrimoine régional et marque surtout son identité culturelle, une volonté locale a été manifestée pour sa conservation et valorisation. Cependant l'aménagement de ces structures s'avère très coûteux, le grossissement de poissons, essentiellement l'anguille, a été prévu afin de justifier les frais d'aménagement. Evolution technologique des DCP en Polynesie francaise ArchiMer

French Polynesia started it's Fish Aggregating Device programme in June 1981 with the positioning of the first buoy offshore of the main island of Tahiti. Since then, 251 structures have been installed throughout the 5 archipelagoes of this French overseas territory. The shape and size of the devices have evolved 8 times over this period, however, a single model has not yet been adopted. Trials are currently underway using lighter structures similar to those found in the Indian Ocean. The anchoring lines have progressively been modified through better knowledge of material and product availability, as well as better understanding of environmental constraints. Concomitant herpes-like virus infections in hatchery-reared larvae and nursery-cultured spat Crassostrea gigas and Ostrea edulis ArchiMer

Concomitant sporadic high mortalities were reported in France in May 1994 among batches of hatchery-reared larval Pacific oysters Crassostrea gigas and European flat oysters Ostrea edulis in 2 hatcheries, and in June and July 1994 among batches of cultured spat of both species in a shellfish nursery. Histological observation showed the presence of cellular abnormalities in moribund animals. Transmission electron microscopy revealed the presence of herpes-like virus particles in infected larvae and spat of both oyster species. Observations of chondrichthyan fishes (sharks,rays and chimaeras) in the Bay of Biscay (North-eastern Atlantic) from submersibles. ArchiMer

Forty one manned submersible dives were carried out during 2 cruises in the Bay of Biscay covering a range of depth from about 100 to 2100 m. Chondrichthyan species were observed in most of the dives, some of them were easily identified and their behaviour could be studied. Twenty-two chondrichthyan species (9 sharks, 8 rays and 5 chimaeras) were recorded. Shark species were always seen active except Scyliorhinus canicula and Galeus melastomus. Some representatives were found motionless on the bottom, sometimes forming groups for the first species. Chimaeras appeared to strongly react to the presence of the submersible. Mapping of Posidonia oceanica beds around Elba Island (western Mediterranean) with integration of direct and indirect methods ArchiMer

Direct and indirect methods were used for mapping Posidonia oceanica beds around Elba Island. Side scan sonar and a visual infra red scanner were used to detect the extension of beds. Also, direct observations were adopted to assess the bed density and to calibrate the indirect methods. Samples of P. oceanica were collected and phenological variables and macroalgal epiphytes were studied to assess the health of the beds. P. oceanica around Elba Island covered an area of 3 680 ha. The largest beds occupied the four main gulfs of the island, characterized by gentle slopes and sandy bottoms.
