The preservation of the genetic variability of hatchery stocks used to supplement natural populations is a priority. Current study employed RAPD markers to examine the genetic diversity of dourado samples from the middle Paranapanema river and from the broodstock used in the stock enhancement program of the Aquaculture and Hydrobiology Station at the Salto Grande Hydroelectric Power Plant. Nineteen RAPD primers were analyzed, which generated 299 bands and the latter were used for genetic analysis. The percentage of polymorphic fragments was higher in stock captured in the Paranapanema river than from fry stocks. The genetic diversity was lower in the broodstock than in natural population. Recentemente a produção aquícola brasileira tem apresentado grande progresso. Dentre as espécies nativas cultivadas no Brasil, a piapara (Leporinus elongatus) tem sido amplamente preconizada. Com objetivo de avaliar os programas de repovoamento, foram analisadas a variabilidade e a divergência genética de três estoques de piapara com a técnica de RAPD (Random Amplified Polymorphic). Este trabalho teve como objetivo analisar, pela técnica RAPD, a variabilidade e a divergência genética de duas gerações da linhagem GIFT. Foram estimados parâmetros para os reprodutores (G0) e para a progênie (F1). A variabilidade genética foi determinada pela porcentagem de loci polimórficos e pelo índice de Shannon. As gerações apresentaram 69,6% de loci polimórficos (G0) e 60,0% de polimorfismo (F1). Os valores para o índice de Shannon foram de 0,367 para a geração G0 e de 0,317 para a F1. Os valores de divergência genética, calculados pelo teste de Mantel, foram de 0,213 para a G0 e 0,208 para a geração F1. Allozyme electrophoresis analysis were performed in four species of Hypostomus (Loricariidae), H. albopunctatus, H. hermanni, H. regani, e Hypostomus sp. 1/NUP 5612 from the Ivaí river, a tributary of the upper Paraná river. The study of 14 loci revealed diagnostic characters and exclusive alleles in a low frequency. The heterozygosity ranged from 0.000 in H. albopunctatus to 0.199 in H. hermanni, which was higher than the heterozygosity in other samples of Hypostomus in literature, as well as in other fish groups. The tambaqui, Colossoma macropomum, is the most popular fish species used for aquaculture in Brazil but there is no study comparing genetic variation among native and farmed populations of this species. In the present study, we analyzed DNA sequences of the mitochondrial DNA to evaluate the genetic diversity among two wild populations, a fry-producing breeding stock, and a sample of fish farm stocks, all from the region of Santarém, in the west of the Brazilian state of Pará. ABSTRACT Trifolium polymorphum Poir. is an amphicarpic forage legume from southern Brazil, Uruguay, Argentina, Paraguay and Chile. Information on the genetic diversity of natural populations in natural grasslands in southern Brazil is limited. In order to increase the knowledge about this species, an analysis of the genetic diversity was carried out in 10 natural populations of T. polymorphum with the use of 20 microsatellite markers. The expected heterozygosity in T. polymorphum populations ranged from 0.40 to 0.43, with a mean of 0.42. The correct identification of species and strains of these parasitoids is necessary to implement the use of biological control. The genetic variability of 27 individuals from two distinct populations (Northeast and Central Brazil) of the grasshopper Schistocerca pallens (Thunberg) and eight isolates of its natural pathogen, the fungus Metarhizium flavoviride Gams & Rozsypal, was investigated using RAPD analysis. For the grasshoppers, ten different 10-mer oligonucleotide primers of arbitrary sequence were selected for analysis, resulting in 79 scorable binary characters. Foram identificadas a divergência e a variabilidade genética, por meio do polimorfismo de seis marcadores microssatélites, de duas populações de Odontesthes bonariensis, utilizadas em manejos de cultivo e com potencial para fornecimento de reprodutores para programas de melhoramento genético. Do total de seis loci, cinco demonstraram eficiência para análise genética nas duas populações de O. bonariensis. A diferenciação genética significante nas populações analisadas pode fornecer a base para futuros programas de melhoramentos genéticos, através da combinação de material das populações divergentes para o desenvolvimento de linhagens ou execução de um programa de seleção.

The genetic variation among nine soybean-originating isolates of Colletotrichum truncatum from different Brazilian states was studied. Nitrate non-utilizing (nit) mutants were obtained with potassium chlorate and used to characterize vegetative compatibility reactions, heterokaryosis and RAPD profile. Based on pairings of nit mutants from the different isolates, five vegetative complementation groups (VCG) were identified, and barriers to the formation of heterokaryons were observed among isolates derived from the same geographic area. The use of different genetic dissimilarity indicators can help in both the selection of crop improvement strategies and as artificial crosses. The objectives of this work were to estimate the genetic dissimilarity among seven oat genotypes (Avena sativa L.) through molecular (dg) and morphological (dm) markers. Data were obtained from two experiments carried out in Capão do Leão County, RS, Brazil, in 2000. The estimated correlation coefficient (r = 0.33) indicated a low association between dg and dm estimates. Fusarium verticillioides strains isolated from corn feed: characterization by fumonisin production and RAPD fingerprinting BABT

In this study a total of 16 Fusarium verticillioides strains isolated from corn feed samples were characterized by fumonisin (FB) production and random amplified polymorphic DNA (RAPD). All the strains produced FB1 and FB2 with levels ranging from 2.41 to 3996.36 µg/g, and from 1.18 to 1209.91 µg/g, respectively. From the 16 F. verticillioides strains, four were identified as low (3.59 to 1289.84 µg/g), eight as intermediate (>1289.84 to 3772.44 µg/g) and four strains as high (>3772.44 µg/g) fumonisin producers. Random amplified polymorphisms between two South American subspecies of rattlesnakes (Crotalus durissus collilineatus and Crotalus durissus terrificus) BABT

The present work was made to determine the suitability of RAPD analysis for the identification of specimens of two subspecies of South American rattlesnakes (Crotalus durissus terrificus and Crotalus durissus collilineatus). The 11 arbitrary primer sequence tested amplified a total of 161 bands, of which 31% were polymorphics when compared with Crotalus specimens. The combining results from different primers allowed the identification of all the specimens under analysis. Several characteristic bands of each subspecies were identified. Dendrogram, based on RAPD markers, show three groups that corresponded to the subspecies of Crotalus, and to Bothrops jararaca specimen.

Genetic variability in regenerated Metarhizium flavoviride protoplasts BABT

Protoplast isolation and regeneration were evaluated in two wild-type and two colour mutant strains of Metarhizium flavoviride. Cultivation in liquid medium, followed by mycelium treatment with Novozym 234 in the presence of KCl 0.7M as osmotic stabilizer, produced 5.05 x 10(6) to 1.15 x 10(7)x mL-1 protoplasts. Genetic variability of pre and post-fragmentation cohorts of Aspidosperma polyneuron Muell. Arg. (Apocynaceae) BABT

RAPD was used to access the genetic variability in Aspisdosperma polyneuron, a long-lived, late-reproducing tropical tree, and highly important for the Atlantic Forest. RAPD profiles from adults (pre-fragmentation, >300 years old) and seedlings (post-fragmentation, <<50 years old) were analyzed. Results showed a decrease of genetic polymorphism of post-fragmentation cohorts in small fragments and higher genetic diversity within population. The genetic diversity distribution suggested the establishment of fragments as protected reserves, and the transference of seedlings among fragments for conservation of A. polyneuron.

Microsatellite analysis of the parental contribution of Piaractus mesopotamicus to the production of offspring in the semi-natural system of reproduction BABT

The objective of this study was to evaluate the genetic diversity and the parental contribution of Piaractus mesopotamicus in the production of offspring in the semi-natural system of reproduction. Twenty parental fishes (eleven males and nine females) and the total of 100 larvae were evaluated by microsatellite marker. The parents and offspring had thirty-one alleles and heterozygosity of 0.550 and 0.563, respectively. The females were fertilised by two up to six males while the males fertilised three up to five females. The contribution of the females and males to the offspring were 66.6 and 58%, respectively.
