Influence of estradiol analogue on cell growth, morphology and death in esophageal carcinoma cells Biocell
Mqoco,Thandi; Marais,Sumari; Joubert,Annie.
2-Methoxyestradiol-bis-sulphamate is a bis-sulphamoylated derivative of the naturally occurring 17-beta-estradiol metabolite namely 2-methoxyestradiol. 2-Methoxyestradiol-bis-sulphamate is regarded as a potential anticancer drug with increased antiproliferative activity when compared to 2-methoxyestradiol. The aim of this pilot in vitro study was to determine the influence of 2-methoxyestradiol-bis-sulphamate on cell growth, morphology and possible induction of certain types of cell death in the SNO esophageal carcinoma cell line. A dose-dependent study (0.2-1.0µM) was conducted with an exposure time of 24 hours. Data revealed that 2-methoxyestradiol-bis-sulphamate reduced cell numbers statistically significantly to 74% after exposure to 0.4µM of the drug....

Effect of sugars on the association between cowpea vicilin (7S storage proteins) and fungal cells Biocell
Rose,T.L.; Gomes,V.M.; Da Cunha,M.; Fernandes,K.V.S.; Xavier-Filho,J..
Vicilins (7S storage proteins) found in various legume seeds have been previously shown to interfere with the germination of spores or conidia of phytopathogenic fungi and inhibit yeast growth and glucose stimulated acidification of the medium by yeast cells. In the present work vicilins from cowpea (Vigna unguiculata) seeds were added to the growth medium of Saccharomyces cerevisiae cells and Fusarium oxysporum conidia. Helix pomatia lectin, wheat germ agglutinin and Ulex europaeus lectin were used to identify differences in the binding of the vicilins to the surface of cells of S. cerevisiae and F. oxysporum treated with this protein. After the growth period, the material in suspension (yeast cells) was centrifuged and the final pellet was also treated...

Effect of age on the myosin-V immunoreactive myenteric neurons of rats ileum Biocell
Ferreira Schoffen,João Paulo; Marçal Natali,Maria Raquel.
Alterations in the gastrointestinal neuromuscular function related to age have been demonstrated in human and animal models. This study analyzes the effects of the aging process on the area of the neuronal cell bodies of the myenteric plexus in the antimesenteric and intermediate regions of the ileal circumference of Wistar, 12 month-old in comparison 3 month-old animals. The ileum was removed and whole-mount preparations immunostained by the antibody anti-myosin-V were processed. The morphometric analyses were performed using a computerized image analysis system, with a subsequent distribution of neurons by size in intervals of 100 μm². The cellular body morphometry revealed a significant increase in the size of the myosin-V- immunoreactive...

Tissue alterations in the Guinea pig lateral prostate following antiandrogen flutamide therapy Biocell
Cordeiro,Renato Simões; Scarano,Wellerson Rodrigo; Góes,Rejane Maira; Taboga,Sebastião Roberto.
The flutamide antiandrogenic effects on the Guinea pig male prostate morphology in puberal, post-puberal and adult ages were evaluated in the present study. Daily-treated group animals received flutamide subcutaneous injection at a dose of 10 mg/Kg body weight for 10 days. The control group animals received a pharmacological vehicle under the same conditions. The lateral prostate was removed, fixed and processed for light and transmission electron microscopy. The results revealed an increase of the acinus diameter in the treated puberal animals and straitness in the stromal compartment around the acini. The epithelial cells exhibited cubic phenotype. In the post-puberal and adult animals, a decrease of the acinus diameter was observed, as well as an...

Morphological study of the spermatogenesis in the teleost Piaractus mesopotamicus Biocell
da Cruz-Landim,Carminda; Camargo Abdalla,Fábio; da Cruz-Höfling,Maria Alice.
The spermatogenesis of Piaractus mesopotamicus was investigated under light and transmission electron microscopy. The specimens were captured from their natural environment (Rio Miranda and Rio Aquidauana, Pantanal Matogrossense, Brazil) during April and September. The results were compared with the spermatogenic data of specimens under captivity condition. In both conditions, P. mesopotamicus presented the typical spermatogenesis pattern of the teleost fishes, showing no significative differences. The spermatozoon was classified as type I, which has a globular head without acrosome, a short middle piece and a long tail constituted only by the flagellum. This type of spermatozoon is considered the basic type in fishes.

CAS role in the brain apoptosis of Bufo arenarum induced by Cypermethrin Biocell
Izaguirre,M. F.; Vergara,M. N.; Casco,V. H..
CAS might have a key role in the apoptosis induced by toxins, acting as anti-apoptotic factor, stimulating the cellular proliferation and the cell contact stabilization. To start to elucidate their role in the brain apoptosis of Bufo arenarum induced by cypermethrin (CY), the expression patterns of CAS and several cell adhesion molecules (CAMs) were established. Bufo arenarum tadpoles of the control and acute bioassay survival at different doses (39, 156, 625 and 2,500 µg CY/L) and times (24, 48, 72 and 96 h) of CY treatment were fixed in Carnoy, embedded in paraffin and sectioned. CAS and CAMs expression was determined by immunofluorescence and immunohistochemistry, respectively. When the bioassay starts, CAS increases suggesting a proliferative or...

Micropropagation of Ilex dumosa (Aquifoliaceae) from nodal segments in a tissue culture system Biocell
Luna,C.; Sansberro,P.; Mroginski,L.; Tarragó,J..
Micropropagation of Ilex dumosa var. dumosa R. ("yerba señorita") from nodal segments containing one axillary bud was investigated. Shoot regeneration from explants of six-year-old plants was readily achieved in ¹/4 strength Murashige and Skoog medium (¹/4 MS) plus 30 gr·L-1 sucrose and supplemented with 4.4 mM BA. Further multiplication and elongation of the regenerated shoots were obtained by subculture in a fresh medium of similar composition with 1.5 gr·L-1 sucrose. Rooting induction from shoots were achieved in two steps: 1) 7 days in ¹/4 MS (30 gr·L-1 sucrose, 0.25 % Phytagel®) with 7.3 mM IBA and 2) 21 days in the same medium without IBA and 20 mM of cadaverine added. Regenerated plants were successfully transferred to soil. This micropropagation...

Orbicules diversity in Oxalis species from the province of Buenos Aires (Argentina) Biocell
Rosenfeldt,Sonia; Galati,Beatriz Gloria.
Eleven Oxalis L. species from the province of Buenos Aires (Argentina) were investigated with scanning and transmission electron microscopes. We identified four different types and two subtypes of orbicules. We conclude that the close morphological similarity between these species is also reflected in their orbicules, and we suggest that the orbicules morphology may be a useful character in systematic studies.

Distribution of pectins in the pollen apertures of Oenothera hookeri.velans ster/+ster: Biocell
Noher de Halac,I.; Cismondi,I.A.; Rodriguez-Garcia,M.I.; Famá,G..
Cell wall pectins are some of the most complex biopolymers known, and yet their functions remain largely mysterious. The aim of this paper was to deepen the study of the spatial pattern of pectin distribution in the aperture of Oenothera hookeri.velans ster/+ster fertile pollen. We used "in situ" immunocytochemical techniques at electron microscopy, involving monoclonal antibodies JIM5 and JIM7 directed against pectin epitopes in fertile pollen grains of Oenothera hookeri.velans ster/+ster. The same region was also analyzed by classical cytochemistry for polysaccharide detection. Immunogold labelling at the JIM7 epitope showed only in mature pollen labelling mainly located at the intine endo-aperture region. Cytoplasmic structures near the plasma membrane...

Models for the recent evolution of protocadherin gene clusters Biocell
Morgan,Marcos.
The clustered protocadherins (Pcdhs) are single-pass transmembrane proteins that constitute a subfamily within the cadherin superfamily. In mammals, they are arranged in three consecutive clusters named alpha, beta, and gamma. These proteins are expressed in the nervous system and are targeted to mature synapses. Interestingly, different neurons express different subsets of isoforms; however, little is known about the functions and expression of the clustered Pcdhs. Previous phylogenetic analyses that compared rodent and human clusters postulated the recent occurrence of gene duplication events. Using standard phylogenetic methods, I confirmed the prior observations, and I show that duplications are likely to occur through unequal crossing-over events...

Cloning and analysis of IFRG (interferon responsive gene) in rabbit oocytes and preimplantation embryos Biocell
Qi,Bing; Zheng,Xinming; Li,Wenyong; Wei,Qingxin; Chen,Qingxuan.
Although there is more evidence that shows that IFNs (interferons) plays a very important role in the early development of the embryo, the mechanism of IFNs is still unclear. Our study showed that IFRG is expressed from oocytesthrough to the preimplantation embryo in rabbits. This finding provides some clues for better understanding the role of IFNs in the development of the embryo. The full length of rabbit IFRG cDNA (Accession No. AJ584672), with a 2794bp encoding 131 amino acid sequence, was cloned IFRG expression can be detected in 8 different tissues: ovary, heart, lung, liver, kidney, spleen, cerebra, and the 18-day whole-body embryo. Whole-mount in situ hybridization showed that IFRG was highly expressed in the inner-cell mass of rabbit blastula....

Storage lipids and proteins of Euterpe edulis seeds Biocell
Panza,Vïctor; Pighin,Dario; Láinez,Verónica; Pollero,Ricardo J.; Maldonado,Sara.
Comparative studies on fatty acid and protein composition of the endosperm and embryo of palmito (Euterpe edulis Martius) were conducted using gas-liquid chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis. On a dry weight basis, the embryo contained extremely lower amounts of lipids and proteins than did the endosperm, which was associated with the scarce lipid and protein bodies previously reported in axis and cotyledon. The fatty acid composition also exhibited differences between both tissues: (I) the fatty acid diversity was greater in embryo than in endosperm; (II) embryo and endosperm contained predominantly linoleic, palmitic, oleic and stearic acids even though the relative values were different for each tissue. As...

Salicylic acid permeation: A comparative study with different vehicles and membranes Biocell
Olivella,Mónica S.; Debattista,Nora B.; Pappano,Nora B..
Considering the skin´s function, different dermal pharmaceutical forms can be developed according to the type of therapeutic activity, active principle and excipients involved in the formulation, such as “transdermal preparations”. In the present study, the permeation parameters of the non-steroidal antiinflammatory drug, salicylic acid (SA) through synthetic membrane, polyvinyliden difluoride, and a biological membrane, egg shell membrane, with different vehicles, propylene glycol, isopropyl alcohol and carbopol gel, were determined. The reported physicochemical parameters of SA from CG were significantly higher than those obtained using PG and IP. This is attributed to the lipophilic nature of the vehicle that facilitates the release and penetration of...

Influence of genotype and explant source on indirect organogenesis by in vitro culture of leaves of Melia azedarach L. Biocell
VILA,S.K.; REY,H.Y.; MROGINSKI,L.A..
In vitro regeneration of shoots from leaf explants of the Paradise tree (Melia azedarach L.) was studied. Three different portions (proximal portion, distal portion and rachis of the leaflets) of three developmental stages (folded, young still expanding and completely expanded) of leaves of 10 - 15 years old plants of seven genotypes were cultured on Murashige and Skoog (1962) medium (MS) supplemented with 1mg.l-1 benzylaminopurine (BAP) + 0.1mg.l-1 kinetin (KIN) + 3 mg.l-1 adenine sulphate (ADS). The rachis of the leaflets of the completely expanded leaves was found to be the most responsive tissue, in most of the genotypes employed. Shoot regeneration occurred in leaf explants of all the genotypes tested. The best genotype for shoot regeneration was...

Karyological and electrophoretic differences between Pomacea flagellata and P. patula catemacensis: Caenogastropoda: Ampullariidae Biocell
Diupotex-Chong,María Esther; Cazzaniga,Néstor J; Uribe-Alcocer,Manuel.
The widespread Mexican apple snail Pomacea flagellata (Say 1827) and the strictly endemic "tegogolo" P. patula catemacensis (Baker 1922) (restricted to Lake Catemaco), are the only known American Ampullariidae that have haploid complements n=13. Pomacea patula catemacensis has suffered a critical reduction in abundance due to immoderate fishing for human consumption. Chromosome slides were obtained from colchicine-injected Pomacea snails collected from nine locations along the coastal zone of the Gulf of Mexico, including Lake Catemaco, for use in principal component analysis (PCA). Total proteins in foot homogenates were analyzed through isoelectric focusing (IEF) and native-PAGE electrophoresis on polyacrylamide gels. The chromosome number 2n=26 was...

Effects of the ascorbic acid supplementation on NADH-diaphorase myenteric neurons in the duodenum of diabetic rats Biocell
Pereira,Marli Aparecida Dos Santos; Bagatin,Maria Claúdia; Zanoni,Jacqueline Nelisis.
We assessed the ascorbic acid (AA) supplementation on the myenteric neurons in the duodenum of rats. Fifteen rats with 90 days of age were divided into three groups: control (C), diabetics (D) and ascorbic acid treated diabetics (DA). After 120 days of daily treatment with AA, the duodenum was submitted to the NADH-diaphorase (NADH-d) histochemical technique, which allowed us to evaluate the neuronal density in an area of 8.96 mm² for each duodenum, and also to measure the cellular profile area of 500 neurons per group. The supplementation promoted an increase on AA levels. The neuronal density (p <0.05) was higher in the group DA when compared to group D. There were no significant differences in the neuronal areas, when we compared groups C (204 ±...

A standard weight equation to assess the body condition of pejerrey Odontesthes bonariensis Biocell
Colautti,Darío C.; Remes Lenicov,Mauricio; Berasain,Gustavo E..
We developed a standard weight equation Ws to aid in the analysis of pejerrey Odonthestes bonariensis body condition over time and across populations using the relative weight index Wr. Weight - length data were compiled from 73 populations of pejerrey (N=16.022) from the Argentine pampas region. We used the regression-line-percentile technique, which provides a 75th-percentile standard by length intervals of 10 mm, to develop the Ws equation. The proposed equation is log10 Ws=-5,267+3,163 log10 Lst; Ws is weight in grams and Lst is standard length in millimeters. This equation is proposed for use with pejerrey between 120 and 520 mm of Lst. Values for Wr calculated with the Ws equation did not consistently increase or decrease as function of fish length,...

Protein detection in spermatids and spermatozoa of the butterfly Euptoieta hegesia (Lepidoptera) Biocell
Mancini,Karina; Dolder,Heidi.
This study was undertaken to detect protein components in both sperm types of the butterfly Euptoieta hegesia. These spermatozoa possess complex extracellular structures for which the composition and functional significance are still unclear. In the apyrene sperm head, the proteic cap presented an external ring and an internal dense content; basic proteins were detected only in external portions. In the tail, the paracrystalline core of mitochondrial derivatives and the axoneme are rich in proteins. The extratesticular spermatozoa are covered by a proteic coat, which presented two distinct layers. In eupyrene spermatozoa, acrosome and nucleus were negatively stained, probably because of their high compaction. In the tail, there is no paracrystalline core...

A six-rhabdomere, open rhabdom arrangement in the eye of the chrysanthemum beetle Phytoecia rufiventris: some ecophysiological predictions based on eye anatomy Biocell
Meyer-Rochow,Victor Benno; Mishra,Monalisa.
We are describing a rhabdom organization of the eye of the chrysanthemum beetle Phytoecia rufiventris that to date has not been described from any other insect. In cerambycid beetles free rhabdomeres, forming a circular, open rhabdom, surround a central rhabdom made up of the rhabdomeres of one or two cells. In Phytoecia rufiventris the central rhabdomeres are missing throughout the eye and the microvilli of the outer 6 rhabdomeres are regularly oriented in three directions. Following the classification of rhabdom types suggested by Wachmann (1979), we suggest to name the rhabdom arrangement seen in the retina of Phytoecia rufiventris "Grundmuster 3". This pattern ought to facilitate polarization sensitivity and movement perception, features that agree...
