Molecular characterisation of Aspergillus flavus isolates from peanut fields in India using AFLP BJM

Aflatoxin contamination of peanut, due to infection by Aspergillus flavus, is a major problem of rain-fed agriculture in India. In the present study, molecular characterisation of 187 Aspergillus flavus isolates, which were sampled from the peanut fields of Gujarat state in India, was performed using AFLP markers. On a pooled cluster analysis, the markers could successfully discriminate among the ‘A’, ‘B’ and ‘G’ group A. flavus isolates. PCoA analysis also showed equivalent results to the cluster analysis. Most of the isolates from one district could be clustered together, which indicated genetic similarity among the isolates. Mycoplasmas are a heterogeneous group of the smallest organisms capable of self replication and are known to cause many detrimental diseases in both animals and humans. These wall-less prokaryotes are enveloped by a lipoprotein membrane and their small genomes are sufficient to synthesize molecules required for growth and self-replication. Among sixteen species isolated from humans, Mycoplasma pneumoniae, an agent of primary atypical pneumonia, and the urogenital tract species Mycoplasma hominis,Ureaplasma urealyticum and Ureaplasma parvum have been confirmed to be pathogenic. Comparison of serological methods for the diagnostic of Caprine arthritis-encephalitis (CAE) in Rio de Janeiro, Brazil BJM

Serum samples of 562 goats were tested for Caprine arthritis-encephalitis using agar gel immunodiffusion test (AGID) and enzyme linked immunosorbent assay (ELISA). Seventy-nine samples (14.1%) were reactive by both testes but 141 (25.1%) were reactive only by ELISA. ELISA, when compared to AGID, presented 100% sensitivity and 70.8% specificity.

Expression of enterovirus 71 capsid protein VP1 in Escherichia coli and its clinical application BJM

The VPl gene of enterovirus 71 (EV71) was synthesized, construct a recombinant plasmid pET15b/VP1 and expressed in E. coli BL21. The recombinant VP1 protein could specifically react with EV71-infected patient sera without the cross-reaction with serum antibodies of coxsackievirus A16 (CA16), A4, A5, B3 and B5 as well as echovirus 6. In acute and convalescent phases, IgM and IgG antibodies of 182 serum samples were detected by ELISA with recombinant VP1 protein as a coated antigen. Analysis of isotype-specific antibody responses to bovine herpesviruses 1.1 and 1.2a allows to estimate the stage of infection BJM

Specific IgM, IgA, IgG1, IgG2, as well as neutralizing antibody responses were evaluated in sera of calves experimentally infected with two isolates of bovine herpesvirus type 1 (BoHV1) of distinct subtypes (subtype 1, BoHV1.1; subtype 2a, BoHV-1.2a). No significant differences were observed in the antibody responses induced by each BoHV-1 subtype. The antibody responses following primary acute infection were characterized by an increase in specific IgM and IgA levels between days 2 and 14 post inoculation (pi). ELISAs for paratuberculosis employ a preadsorption step with Mycobacterium phlei to diminish unspecific reactions As M. fortuitum is one of the most frequent environmental mycobacteria, the purpose of this pilot study was to evaluate its use as an alternative for the preadsorption in ELISAs for paratuberculosis. Results suggest that M. fortuitum can be an alternative instead of or associated to M. phlei with comparable results (κ > 0.8) to conventional ELISAs using M. phlei as a preadsorption antigen.

Immunoglobulin G proteolytic activity of Actinobacillus actinomycetemcomitans BJM

Actinobacillus actinomycetemcomitans produces a protease to human immunoglobulin G that is an important evasion mechanism. In this study, the proteolytic activity of A. actinomycetemcomitans strain ATCC 43718 on human immunoglobulin G associated with culture supernatant concentrations, the growth period and the period of incubation with immunoglobulin G were evaluated by an enzyme linked immunosorbent assay. Evaluation of an indirect ELISA for the detection of Salmonella in chicken meat BJM

In this work, an indirect ELISA based on a monoclonal antibody (MAb) specific for an outer membrane protein of Salmonellaenterica serovar Enteritidis was used for detection of Salmonella in 154 samples of chicken meat. Its efficiency was determined through comparison with the results obtained from the conventional method. The prevalence of samples contaminated with Salmonella was 23% with the conventional culture method, and 26% with the ELISA. Development of an indirect ELISA based on recombinant capsid protein to detect antibodies to bovine leukemia virus BJM

Abstract Serological testing and culling infected animals are key management practices aiming eradication of bovine leukemia virus infection. Here, we report the development of an indirect ELISA based on BLV recombinant capsid protein (BLVp24r) to detect anti-BLV antibodies in cattle serum. The BLVp24r was expressed in Escherichia coli and purified by affinity chromatography, and then used to set up the ELISA parameters. The Polysorp ® plate coated with 50 ng of antigen/well and bovine serum diluted 1:100 gave the best results during standardization. Up-regulation of human immune system function by Donkey’s Milk BJPS

Donkey’s milk represents a good alternative to human milk because of its chemical characteristics similar to those of human’s. In present study, the pro- and anti-inflammatory effects of donkey’s milk were evaluated on peripheral blood mononuclear cells (PBMCs). PBMCs were isolated from 12 young and 12 aged normal subjects. PBMCs were cultured with or without the optimal and non-cytotoxic dose of pasteurized donkey’s milk, and polymyxin B was used to inhibit the possible endotoxin contamination. A infecção ocasionada pelo Rhodococcus equi é responsável por elevadas taxas de mortalidade e grandes perdas econômicas na equideocultura. Diante da importância dessa enfermidade, este trabalho objetivou a padronização de um ensaio imunoenzimático indireto (ELISA teste) para a detecção de anticorpos anti-Rhodococcus equi em potros (n=16) antes e após a ingestão de colostro de mães vacinadas. An enzyme-linked immunosorbent assay for the detection of IgG antibodies against Babesia equi in horses Ciência Rural

A crude antigenic preparation of Babesia equi was used to develop and establish the suitability of an enzyme-linked immunosorbent assay (ELISA) for the detection of parasite carriers. Optimal dilutions of the antigen, using positive and negative reference sera, were determined by checkboard titrations. The specificity and sensitivity of the ELISA were 100 %. A total of 90 serum samples were taken from horses from the Northeast region of São Paulo State and examined for diagnosis of equine B. equi infection by ELISA. A herd of cattle (Holstein-Zebu crosses) was screened every two months by ELISA during a period of two years for IgG antibodies against 19 infectious disease agents. Two hundred and ninety five sera were collected from 157 young animals (0-4 months of age), 1037 sera from 292 developing animals (4-36 months of age) and 1468 sera from 259 producing animals (> 36 months of age). The results indicate that the difference in ELISA between positive and negative tests is associated with the overall prevalence of positive tests. Monitoramento da presença de Mycoplasma hyopneumoniae em granjas de suínos durante a implementação de programas de erradicação Ciência Rural

O objetivo deste estudo foi monitorar a presença de M. hyopneumoniae em granjas suínas durante a implementação de programas de erradicação utilizando diferentes técnicas de diagnóstico focalizando no PCR. Trabalhou-se com uma empresa que possuía três granjas, uma parto-terminação (390 matrizes), uma múltiplo-sítio (4100 matrizes) e uma nova granja que povoava suas novas instalações. Nas duas primeiras, foi desenvolvido um programa de despovoamento parcial para erradicar a pneumonia enzoótica suína, a última foi povoada pelos suínos dos anteriores após a erradicação. Elisa indireto na detecção de Salmonella spp. em lingüiça suína Ciência Rural

Um teste ELISA, baseado em um anticorpo monoclonal (MAb) específico para uma proteína de membrana externa de Salmonella enterica serovar Enteritidis foi comparado com o método de cultivo tradicional na detecção de Salmonella spp. em 110 amostras de lingüiça suína frescal. A prevalência do patógeno nas amostras foi de 11,82% de acordo com o cultivo tradicional. O teste ELISA revelou sensibilidade, especificidade, valores preditivos positivos e negativos de 100%, 98%, 87% e 100%, respectivamente. Tais achados indicam que, comparado ao sistema tradicional, o teste imunológico foi bastante eficaz na determinação de Salmonella spp. em amostras de lingüiças naturalmente contaminadas.

Leishmaniose visceral em um canino de Cruz Alta, Rio Grande do Sul, Brasil Ciência Rural

Descreve-se um caso de leishmaniose visceral em um canino, macho, Akita, cinco anos, que foi atendido no Hospital Veterinário da Universidade de Cruz Alta, RS, após residir três anos no Estado de Alagoas. Clinicamente observou-se emagrecimento progressivo, lesões crostosas na ponta das orelhas, focinho e região periorbital, úlcera na região distal do membro torácico direito, opacidade da córnea, vômitos e eventuais episódios de diarréia. MARKOV CHAIN MODELING OF ENDEMIC CATTLE DISEASES IN THE TROPICS OF MEXICO Ciência Rural

Endemic diseases measured by prevalence of IgG antibodies by ELISA all showed remarkably stable overall prevalences over a study period of 3 years. This however does not mean that incidence is low or absent. Observed transition from the state of being positive for a disease to the state of being negative and vice versa suggested fairly high incidence rates (and rates of waning antibodies) for all 19 disease agents tested for. In spite of this dynamic situation Markov Chain simulations indicate that a steady prevalence is reached in a fairly short time. Markov Chain simulations may also be useful in elucidating seasonal changes in prevalence.

O presente estudo teve como objetivo determinar a frequência e os fatores de risco para a infecção pelo vírus da diarreia viral bovina (BVDV) em fêmeas bovinas leiteiras na região amazônica maranhense. Amostras de soro de animais não vacinados contra o BVDV e provenientes de 40 propriedades foram submetidas à técnica de ELISA indireto. Em cada propriedade avaliada, aplicou-se ainda um questionário epidemiológico para investigar os fatores de risco que poderiam estar associados à infecção pelo vírus.
