Plant regeneration of Anthurium andreanum cv Rubrun Electron. J. Biotechnol.

To establish an efficient regeneration system for Anthurium andreanum cv Rubrun, seeds from plant spadixes were germinated on a medium supplemented with 2.2 muM BA. After 2 weeks, 74% of the seeds germinated and four weeks later, micro-cuttings from these plantlets were subcultured on a medium containing 4.4 muM BA and 0.05 muM NAA. On average, 3.6 shoots per explant were obtained. Four weeks old in vitro plants from germinated seeds and the plantlets obtained from micro-cuttings, showed callus proliferation at the stem base. Background The aim of the present study was to assess the genetic diversity of Pantaneiro sheep, using microsatellite markers, in order to assist maintenance and management plans, enhance mating systems and reduce the inbreeding rate. A total of 127 animals were genotyped at eight microsatellite loci. They belonged to populations from the Experimental Farm of the Universidade Federal da Grande Dourados (UFGD) (Dourados/MS/Brazil) and Embrapa Pantanal (Corumbá/MS/Brazil). Background: Owing to the growing interest in biofuels, the concept of a biorefinery where biomass is converted to a variety of useful products is gaining ground. We here present how distillery waste is combined with a by-product from a sugar production, molasses, to form a medium for the growth of Lactobacillus plantarum with yields and biomass densities comparable with conventional industrial media. Genetic diversity analysis of rice cultivars (Oryza sativa L.) differing in salinity tolerance based on RAPD and SSR markers Electron. J. Biotechnol.

Thirty rice cultivars were evaluated for salinity tolerance during the seedling stage and were divided into five tolerance groups including tolerant (T), moderately tolerant (MT), moderately susceptible (MS), susceptible (S) and highly susceptible (HS) which comprised 5, 10, 9, 4 and 2 cultivars respectively. Genetic diversity of all rice cultivars was evaluated using random amplified polymorphic DNA (RAPD) and simple sequence repeats (SSR) markers. In the present study we examined the possibility of propagating different Cyclamen species (C. africanum Boiss. and Reut., C. cilicium Boiss. and Heldr., C. coum Mill., C. hederifolium Ait., C. persicum Mill., C. purpurascens Mill.) including some of their subspecies and cultivars in vitro using explants of adult plants. For this purpose two protocols have been applied to eleven genotypes combined with mostly four explant types (placentas with ovules, leaves, petioles and peduncles). Evaluating Colombian Clostridium spp. Strains hydrogen production using glycerol as substrate Electron. J. Biotechnol.

Background: The ability for hydrogen production of 13 native strains of Clostridium spp. strains isolated from Colombian soil was evaluated using glycerol substrate. Glycerol to hydrogen conversion was investigated using a batch fermentation reactor and industrial glycerol source (50 g.l-1, pH 7.00). Results: The results were quantified regarding acids, hydrogen, biomass and solvent production. The selected strain gave good hydrogen over production output at 14.4 mmol H2.l-1, productivity 0.3167 mg H2.h-1 l-1 culture mediumand yield 0.1962 mol H2.mol-1 glycerol. The removal of contaminants in slow sand filters occurs mainly in the colmation layer or schmutzdecke - a biologically active layer consisting of algae, bacteria, diatoms and zooplankton. A ripening period of 6 - 8 weeks is required for this layer to form, during which time filter performance is sub-optimal. In the current study, an environmental scanning electron microscope was used to visualise the ripening process of a pilot-scale slow sand filter over a period of eight weeks. Designing and validation of genus-specific primers for human gut flora study Electron. J. Biotechnol.

The aim of this study, was to design and validate 16S rRNA targeted oligonucleotide genus specific primers for amplifying the predominant members of gut flora using polymerase chain reaction. Primers were validated against human faecal samples. Gut flora of a normal individual was compared with that of two diseased individuals. Our observations showed that the genera Lactobacillus, Bacteroides, Peptococcus, Bifidobacterium, and E. coli were invariably present in all studied subjects however, the absence of butyrate producing bacteria Ruminococcus and Peptostreptococcus were significant. Presence of the members of the genus, Campylobacter in both the diseased samples were also unusual.

Eucalyptus plantations account for 74% of the forested land, with Eucalyptus globulus being the most widely distributed species. Cloning and expression analysis of peanut (Arachis hypogaea L.) CHI gene Electron. J. Biotechnol.

Chalcone isomerase (CHI) is the key enzyme that catalyzes chalcone into (2S)-flavanol or (2S)-5-desoxidation flavanol. The full length cDNA (1050 bp) of AhCHI (Arachis hypogaea CHI gene) was cloned by large scale EST sequencing using a peanut immature seed cDNA library. Sequence analysis results indicated that it was a type I CHI gene (with the accession number JN660794). The ORF of AhCHI was 768 bp, encoding a peptide of 255 amino acids with a pI of 5.189. Copper removal from aqueous solution by marine green alga Ulva reticulata Electron. J. Biotechnol.

The batch removal of copper (II) ions from aqueous solution under different experimental conditions using Ulva reticulata was investigated in this study. The copper (II) uptake was dependent on initial pH and initial copper concentration, with pH 5.5 being the optimum value. The equilibrium data were fitted using Langmuir and Freundlich isotherm model, with the maximum copper (II) uptake of 74.63 mg/g determined at a pH of 5.5. The Freundlich model regression resulted in high correlation coefficients and the model parameters were largely dependent on initial solution pH. Molecular analysis of genetic stability in long-term micropropagated shoots of banana using RAPD and ISSR markers Electron. J. Biotechnol.

A large number of micropropagated plantlets of banana, Musa acuminata var. Nanjanagudu Rasabale (NR), that were developed from axillary shoot bud explants over 10 years ago were screened for genetic variation, if any, using RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter-Simple Sequence Repeats) markers. Sesame (Sesamum indicum L.) is one of the oldest oil crops and is widely cultivated in Asia and Africa. To determine the level of genetic diversity in relation to geographical origins and morphological characteristics, a total of 96 accessions have been collected from different parts of the world and were analyzed using AFLP techniques. A simple ethanol wash of the tissue homogenates recovers high-quality genomic DNA from Corchorus species characterized by highly acidic and proteinaceous mucilages Electron. J. Biotechnol.

A simple miniprep based on early elimination of highly acidic and proteinaceous mucilages through ethanol washing of the tissue homogenates has been developed for the extraction of genomic DNA from mature leaves and seeds of Corchorus spp. As compared to high cetyltrimethylammonium bromide (CTAB)-NaCl DNA extraction followed by ethanol-based removal of remnant mucilages from the DNA pellet, this simple miniprep consistently and reproducibly recovers high amounts of DNA with good spectral qualities at A260/A280 and A260/A230. The purified DNA is efficiently digested by restriction endonucleases, and is suitable for PCR amplification of nuclear microsatellites with expected allele sizes.
