Role of mast cells in gastrointestinal mucosal defense Biocell
Penissi,Alicia; Rudolph,María; Piezzi,Ramón.
The purpose of this review, based on studies from our laboratory as well as from others, is to summarize salient features of mast cell immunobiology and to describe their associations with gastrointestinal mucosal defense. Gastrointestinal mast cells are involved in many pathologic effects, such as food hypersensitivity. On the other hand, they also play a protective role in defense against parasitic and microbial infections. Thus, they have both positive and negative effects, but presently the mechanisms that control the balance of these various effects are poorly known. It has been suggested that stabilization of mast cells may be a key mechanism to protect the gastrointestinal tract from injury. Few molecules are known to possess both mast cell...

Secondary zoospores in the algal endoparasite Maullinia ectocarpii (Plasmodiophoromycota) Biocell
Parodi,Elisa R.; Cáceres,Eduardo J.; Westermeier,Renato; Müller,Dieter G..
The present paper deals with the ultrastructure of zoospores produced by the plasmodiophorid Maullinia ectocarpii, living in the marine algal host Ectocarpus siliculosus. The zoospores described here are very similar to secondary zoospores of Polymyxa graminis and Phagomyxa sp.(the latter an algal endoparasite, also). Our results indicate that M. ectocarpii produces two types of plasmodia, and suggest that is a species with a complete life cycle, as it is known for all the Plasmodiophormycota that have been studied. Sporogenic and sporangial plasmodia produce, respectively, primary zoospores with parallel flagella within thick walled resting sporangia, and secondary zoospores with opposite flagella within thin walled sporangia.

Single-channel response of hamster oocytes to ertilization with homologous spermatozoa Biocell
Ituarte,Leonor M.E.; Viera,Teresa B.; Saldeña,Teobaldo A.; De Rosas,Juan C.; Fóscolo,Mabel; Ibáñez,Jorge E.; Saraví,Fernando D..
Electrophysiological events occur early after fertilization, along with changes in intracellular Ca2+ concentration . Passive electrical parameters were determined in golden hamster oocytes by whole cell patch-clamp method. In separate experiments the effect of 4-aminopyridine on resting oocytes was tested. The single-channel patch clamp configuration was employed to assess the electrical response to fertilization with homologous sperm. Structure of oocytes submitted to patch clamp was evaluated with scanning electron microscopy and found to be preserved. Oocyte diameter was 70.2 ±  2.2 µm; their resting parameters were: membrane potential 23.8 ±  0.8 mV; total membrane specific resistance 519.1 ±  94.6 Ω.cm², and specific capacity 0.99 ±  0.03...

Goblet cell number in the ileum of rats denervated during suckling and weaning Biocell
Hernandes,Luzmarina; Marques da Silva Pereira,Lucieni Cristina; Parisi Alvares,Eliana.
The enteric nervous system plays a role on the stimulation of secretory cells of intestinal epithelia. We have demonstrated that ablation of ENS stimulates epithelial cell proliferation. As goblet cells are important constituents of the epithelial sheet, it is mandatory to investigate separately this cell type. The myenteric plexus of the ileum of rats in postnatal development was partially removed by the serosal application of benzalkonium chloride (BAC). Three groups of animals were used: those where BAC application was at 13 days and sacrifice was at 15 (13/28-day-old) or 23 days after treatment (13/36-day-old), and those where BAC was applied at 21 days and rats were killed 15 days after treatment (21/36-day-old) . The number of goblet cells in the...

Fine-structural details of the photoreceptor membranes in the ocellus of the scale-insect parasite Centrodora sp. (Hymenoptera; Aphenelidae):: A case of gene transfer between host and parasite? Biocell
Yang,Mouyu; Meyer-Rochow,V.Benno.
Only one insect (the scale insect Eriococcus sp.) is known, in which photoreceptive lamellae appear to have replaced the usual arthropod rhabdom microvilli. We are now reporting the presence of photoreceptive membranes, which also appear to resemble lamellae rather than microvilli, but they are in the ocellus of the tiny wasp Centrodora sp., which parasitizes scale insect eggs. The apparently optically homogenous lens of the Centrodora ocellus measures approximately 10 µm in diameter and, thus, operates at the limits of diffraction. We calculated that the lens is capable of focusing a parallel bundle of rays on the retina of the ocellus.

WT-1 mRNA expression is modulated by nitric oxide availability and Hsp70 interaction after neonatal unilateral ureteral obstruction Biocell
Mazzei,Luciana; García,Isabel Mercedes; Cacciamani,Valeria; Benardón,María Eugenia; Manucha,Walter.
Wilms tumor gene 1 (wt-1), a key regulator of mesenchymal-epithelial transformation, is downregulated during congenital obstructive nephropathy, leading to apoptosis. There is a functional interaction between WT-1 and inducible nitric oxide synthase (iNOS). In this regard, we reported that after neonatal unilateral ureteral obstruction, rosuvastatin prevents apoptosis through an increase in nitric oxide bioavailability, which in turn is linked to higher Hsp70 expression. Hence, the goal of this study was to determine whether a nitric oxide/Hsp70 interaction is involved in changes in WT-1 mRNA expression after ureteral obstruction. Neonatal rats submitted to experimental ureteral obstruction were treated with either vehicle or rosuvastatin for 14 days....

Protective effect of aqueous suspension of dried latex of Calotropis procera against oxidative stress and renal damage in diabetic rats Biocell
Kumar,Vijay L.; Padhy,Biswa M..
Calotropis species have been used in the traditional medicinal system for the treatment of diseases of the liver and abdomen. In view of the antioxidant and anti-hyperglycemic properties of an aqueous suspension obtained from the dried latex of Calotropis procera, the present study was carried out to evaluate its efficacy in affording protection against alloxan induced changes in rat kidney. A single intraperitoneal injection of alloxan (150 mg/kg) in rats produced hyperglycemia within 3 days and altered kidney functions over a period of 90 days. Daily oral administration of the aqueous suspension (100 and 400 mg/kg) in diabetic rats produced anti-hyperglycemic effect that was comparable to that of glibenclamide (10 mg/kg). Unlike glibenclamide, the...

Evaluation of a multiplex PCR method to detect enteroaggregative Escherichia coli Biocell
Rüttler,M.E.; Yanzón,C.S.; Cuitiño,M.J.; Renna,N.F.; Pizarro,M.A.; Ortiz,A.M..
Enteroaggregative Escherichia coli (EAEC) has been implicated in sporadic diarrhea in children and adults and has been identified as the cause of several outbreaks worldwide. The HEp-2 test remains the gold standard for identification of this pathotype. A 60-65 MDa plasmid encodes the aggregative adherence fimbriae (AAF/I and AAF/II), a transcriptional activator ( aggR gene), the enteroaggregative heat-stable enterotoxin EAST1 ( astA gene) and a cytotoxin (Pet). The standard assay for EAEC is performed only in research laboratories, because it is expensive, labor intensive and time-consuming. The Polymerase Chain Reaction (PCR) offers the possibility of rapid diagnosis. In the current study, a multiplex PCR assay which checks aggR and astA genes was...

Dalla Nora,Gracieli; Pastori,Tamara; Dail Laughinghouse IV,Haywood; Do Canto-Dorow,Thais Scotti; Bosio Tedesco,Solange.
Mikania glomerata is a plant used in Brazilian traditional medicine, known as ‘guaco'. It possesses anti-inflammatory properties and the aqueous extracts of its leaves are indicated for the treatment of diseases of the respiratory tract. This study aimed at evaluating the antiproliferative and genotoxic effect of Mikania glomerata leaf infusions on the cell cycle of onion. The material used was collected in the native environment from Rio Grande do Sul State, Brazil. Aqueous extracts through infusions were prepared in two concentrations: 4g/L (usual concentration) and 16g/L (4x more concentrated) of each of the populations. Two groups of four onion bulbs for each plant population were used plus a control group. The rootlets were fixed in...

Agmatine inhibits hypoxia-induced TNF-alpha release from cultured retinal ganglion cells Biocell
Hong,Samin; Park,Kyoungsoo; Kim,Chan Yun; Seong,Gong Je.
The effect of hypoxia on the release of tumor necrosis factor-alpha (TNF-alpha) in transformed rat retinal ganglion cells (RGCs) and the effect of agmatine on the hypoxia-induced production of TNF-alpha in RGCs were evaluated. RGCs were cultured under hypoxic conditions with 5% oxygen, with or without 100μM agmatine. The expression levels of TNF-alpha and its receptor-1 (TNF-R1) were investigated by Western blot analysis. After 6 hours of hypoxia, we noted an increase in TNF-alpha production in RGCs. Agmatine significantly reduced TNF-alpha level after 12 hours of hypoxic treatment. The expression of TNF-R1 was not affected by the hypoxia or agmatine treatment. Our results show that agmatine inhibits the TNF-alpha production of RGCs in hypoxic...

Pollen morphology of Oxalis species from Buenos Aires province (Argentina) Biocell
Rosenfeldt,Sonia; Galati,Beatriz G.
Pollen morphology and the degree of pollen variability within nine species and two varieties of Oxalis species from Buenos Aires Province (Argentina) were studied using light microscopy (LM), scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Oxalis pollen grain is generally 3-colpate and the shape is prolate spheroidal, oblate spheroidal or spheroidal. The exine is microreticulate. The brochi are circular to polygonal; brochi sizes decrease near the colpi. The colpus membrane observed in unacetolysed material, is crustate or granular. The colpus length varies within a species; the largest grains are produced in stamens with long filaments and the smallest ones in stamens with short filaments. Four pollen types are distinguished...

Intrahost distribution and trasmission of a new species of cyclopoid copepod endosymbiotic to a freshwater snail, Pomacea canaliculata (Caenogastropoda, Ampullariidae) from Argentina Biocell
Gamarra-Luques,C. D.; Vega,I. A.; Koch,E.; Castro-Vazquez,A..
A new species of cyclopoid copepod, Ozmana huarpium, is described as a symbiont to Pomacea canaliculata (Lamarck 1822) (Caenogastropoda, Ampullariidae). Rather large numbers (about one hundred copepods per snail) were found, although there was no evidence of harm to the host. To our knowledge, O. haemophila (symbiont to P. maculata), and the currently described species, O. huarpium, are the only copepod species ever recorded as endosymbionts to freshwater invertebrates. While O. haemophila is restricted to the haemocoel of its host, O. huarpium predominate in the penis sheath, the ctenidium and the mantle cavity, figuring in these pallial organs 63-65 % of total mature forms. The sex ratio of the symbiont is skewed to the female side in these organs,...

Autophagy, apoptosis and organelle features during cell exposure to cadmiumc Biocell
dos Santos Vergilio,Cristiane; Tenório de Melo,Edésio José.
Cadmium (Cd) induces several effects in different tissues, but our knowledge of the toxic effects on organelles is insufficient. To observe the progression of Cd effects on organelle structure and function, HuH-7 cells (human hepatic carcinoma cell line) were exposed to CdCl2 in increasing concentrations (1 μM - 20 μM) and exposure times (2 h - 24 h). During Cd treatment, the cells exhibited a progressive decrease in viability that was both time- and dose-dependent. Cd treated cells displayed progressive morphological changes that included cytoplasm retraction and nuclear condensation preceding a total loss of cell adhesion. Treatment with 10 μM for 12 h led to irreversible damages. Before these drastic and irreparable damages,...

Morphology and structure of the pollen cone and pollen grain of the Araucaria species from Argentina Biocell
Fueyo,Georgina M. del; Caccavari,Marta A.; Dome,Elizabeth A..
The pollen cone and the pollen grain of the two Argentinean species of Araucaria are described with LM, SEM and TEM. Primordia of pollen cones are formed in April and May and reach maturity by mid-October in A. angustifolia (Bert.) O. Kuntze and by mid-November in A. araucana. (Mol.) K. Koch. Characters of the mature pollen cones and microsporophylls between both taxa are clearly differentiated. Pollen grains are spheroidal-subspheroidal, inaperturate, and asaccate with granulate exine and a subequatorial annular area that corresponds to the sexine thickness. Sculpturing consists of irregularly dispersed granules that are sometimes fused to each other (A. angustifolia) or forming microrugulae (A. araucana). Microgranules and microspinules are also present....
