S-100 dendritic cells in normal and Dermatobia hominis infested cattle skin Braz. J. Vet. Res. Anim. Sci.
OLIVEIRA-SEQUEIRA,Teresa Cristina Goulart de; BACHI,Carlos Eduardo; LELLO,Edy de.
S-100 dendritic cells immunolabeling were investigated in uninfested skin biopsies from five Nelores, five Holstein-Friesian and five crossbreed (Holstein-Friesian x Nelores) calves. The group of crossbreed animal was experimentally infested with 100 first-instar larvae of Dermatobia hominis and biopsies were obtained 24, 48, 72 and 168 hours after infestation. Samples obtained prior to infestation from these animals were used as control. Rabbit anti-protein S-100 antibody and the avidin-biotin-peroxidase method were used for immunolabeling. Melanocytes, nerves and endothelial cells and dermal dendritic cells (DC) were positive to protein S-100. DCs were exclusively detected in the superficial dermis close to the basal layer of both normal and parasitized...

Ghost protein damage by peroxynitrite and its protection by melatonin BJMBR
Di Mascio,P.; Dewez,B.; Garcia,C.R.S..
We have studied the effect of peroxynitrite (ONOO-) on the membrane cytoskeleton of red blood cells and its protection by melatonin. Analysis of the protein fraction of the preparation by SDS-PAGE revealed a dose-dependent (0-600 µM ONOO-) disappearance at pH 7.4 of the main proteins: spectrin, band 3, and actin, with the concomitant formation of high-molecular weight aggregates resistant to reduction by ß-mercaptoethanol (2%) at room temperature for 20 min. These aggregates were not solubilized by 8 M urea. Incubation of the membrane cytoskeleton with ONOO- was characterized by a marked depletion of free sulfhydryl groups (50% at 250 µM ONOO-). However, a lack of effect of ß-mercaptoethanol suggests that, under our conditions, aggregate formation is not...

Obese women on a low energy rice and bean diet: effects of leucine, arginine or glycine supplementation on protein turnover BJMBR
Marchini,J.S.; Lambertini,C.R.; Ferriolli,E.; Dutra de Oliveira,J.E..
This study examined if leucine, arginine or glycine supplementation in adult obese patients (body mass index of 33 ± 4 kg/m²) consuming a Brazilian low energy and protein diet (4.2 MJ/day and 0.6 g protein/kg) affects protein and amino acid metabolism. After four weeks adaptation to this diet, each subject received supplements of these amino acids (equivalent to 0.2 g protein kg-1 day-1) in random order. On the seventh day of each amino acid supplementation, a single-dose 15N-glycine study was carried out. There were no significant differences in protein flux, synthesis or breakdown. The protein flux (grams of nitrogen, gN/9 h) was 55 ± 24 during the nonsupplemented diet intake and 39 ± 10, 44 ± 22 and 58 ± 35 during the leucine-, glycine- and...

The induction of differentially expressed proteins of Xylella fastidiosa with citrus extract BJM
Bellato,Cláudia de M.; Garcia,Andréia K.M.; Mestrinelli,Fabiana; Tsai,Siu M.; Machado,Marcos A.; Meinhardt,Lyndel W..
An in vitro system was developed to induce and identify Xylella fastidiosa proteins that were differentially expressed in the presence of callus-derived extracts from its host, the citrus cultivar Pêra. To optimize the induction, we first developed a single culture medium for the growth of both, host and bacteria. This medium, CPXPm7, which mimics the citrus xylem sap, showed that X. fastidiosa at 72 h post-incubation had 10(8) colony forming units mL-1, while Pêra cells had the highest fresh weight content (0.79 g). After testing various methods of co-cultivation of the bacteria and host callus grown in this single medium, the best induction procedure was to grow X. fastidiosa in a solid medium amended with an extract of Pêra callus grown in CPXPm7....

Seed phytate content and phosphorus uptake and distribution in dry bean genotypes Braz. J. Plant Physiol.
Coelho,Cileide Maria Medeiros; Santos,Júlio Cesar Pires; Tsai,Siu Mui; Vitorello,Victor Alexandre.
Phytic acid is the major storage form of phosphorus in seeds of legumes and cereals. Since phytate can form complexes with proteins and minerals, reducing the digestive availability of these nutrients, it is usually regarded as an antinutrient, although recent work indicates that it has important beneficial roles as an antioxidant and anticarcinogen. Therefore, there is an interest in the assessment and manipulation of phytate contents in important food grains such as beans. The objective of this work was to compare eleven dry bean genotypes with regard to grain contents of P, phytate, inorganic P and protein and to examine if differences between genotypes could be explained by differences in grain protein content or in uptake and partitioning of P. Plants...

Influence of contrasting environments on seed composition of two quinoa genotypes: nutritional and functional properties Chilean J. Agric. Res.
Miranda,Margarita; Vega-Gálvez,Antonio; Martínez,Enrique A; López,Jéssica; Marín,Rosa; Aranda,Mario; Fuentes,Francisco.
Quinoa (Chenopodium quinoa Willd.) in Chile represents a source of germplasm with high nutritional value. However, there is little information available related to quinoa seed quality grown under contrasting environments. In this study we evaluated the changes on seed composition of seeds of two lowland/coastal quinoa genotypes grown under arid (Vicuna) and cold-temperate (Temuco) conditions in Chile. Results showed that in the case of 'Regalona Baer' and 'Villarrica' genotypes the arid location (with irrigation) led to a significant increase (P < 0.05) in grain yield (4.2 and 5.1 t ha-1, respectively), soluble dietary fiber (16.8 ± 0.4 and 28.9 ± 2.1 g kg-1 DM, respectively), vitamin B3 (2.44 ± 0.005 and 2.26 ± 0.04 mg 100 g-1 DM, respectively),...

Genetic divergence of soybean genotypes in relation to grain components Ciência Rural
Rodrigues,Josiane Isabela da Silva; Arruda,Klever Márcio Antunes; Cruz,Cosme Damião; Barros,Everaldo Gonçalves de; Piovesan,Newton Deniz; Moreira,Maurilio Alves.
ABSTRACT: The objective of this paper was to estimate the genetic divergence among 49 soybean ( Glycine max L. Merrill .) genotypes to assist grain quality-focused breeding programs in the choice of progenitors. The genetic divergence was estimated using the Mahalanobis generalized distance from the percentages of protein, oil, and fatty acids oleic, linoleic and linolenic after cultivation of genotypes in different environments. Genotypes were grouped by agglomerative methods and the two and three-dimensional projections of the distance matrix were obtained. The average protein and oil contents in the four environments ranged from 34.25 to 45.18% and from 16.48 to 23.01%, respectively. The average contents of the fatty acids oleic, linoleic and linolenic...

Vargas,Diego Prado de; Nörnberg,José Laerte; Mello,Renius de Oliveira; Scheibler,Rudolf Brand; Mello,Fernanda Cristina Breda; Rizzo,Fábio Antunes; Bellato,Tatiana Kátia Bertussi.
ABSTRACT: Milk supply chain in Brazil exhibits significant production system heterogeneity in all federal units. Thus, the objective of this study was to form homogeneous groups of bovine milk production units based on the chemical and microbiological quality of the milk via multivariate statistical techniques. A total of 1,541 milk producing units (MPUs), corresponding to 44,089 samples, were analyzed. The first three principal components accounted for 81.38% of the total variation in the data. Principal component 1 (PC1) was associated with the chemical quality of milk (fat, protein [PROT] and total dry extract [TDE] content), while PC2 and PC3 were associated with microbiological quality (somatic cell count [SCC] and total bacterial count [TBC]). The...

Feeding dairy cows with soybean by-products: effects on metabolic profile Ciência Rural
Alves,Marilia; González,Félix; Carvalho,Nelcy; Mühlbach,Paulo; Lima,Verônica; Conceição,Tiago Reis; Wald,Vera.
Soybean by-products are currently used in Rio Grande do Sul, Brazil, as a source of protein in dairy cows. However, the high protein breakdown of this feed source in the rumen causes loss of the intrinsic quality of the protein and increases plasma urea with deleterious consequences to animal metabolism. Thermal treatment of soybean can overcome this limitation. The aim of this study was to evaluate the metabolism of dairy cows fed with thermally treated soybean and raw soybean, through the analyses of the metabolic profile of plasma and milk. Twelve Holstein cows in mid-lactation period were studied using four treatments: commercial concentrated of protein, soybean meal, raw soybean and roasted soybean. Protein and urea levels in milk were measured in...
