Use of molecular tools for mollusc disease diagnosis ArchiMer

Recently, an increasing number of research teams are engaged in developing DNA-based diagnostic techniques for mollusc pathogens. These techniques are slowly moving from development in specialised laboratories for research purposes, to routine application and are expected to find an increasing use in routine disease monitoring programs in mollusc aquaculture and in efforts to prevent the spread of pathogens within and between nations. Considering the probable development and interest in these diagnostic techniques, the potential for wide applications in aquaculture and the inherent problems currently associated with their use, it appears necessary to address these issues.

Screening of Vibrio isolates to develop an experimental infection model in the Pacific oyster Crassostrea gigas ArchiMer

In an attempt to develop a reproducible experimental model of bacterial infection in Crassostrea gigas, oysters taken from very localised sub-populations suffering natural mortality outbreaks were used in cohabitation trials under laboratory conditions. From these trials, a collection of Vibrio strains was isolated from moribund and healthy oysters. In a second step, strains were experimentally tested for virulence by means of injection into healthy oysters. This screening revealed a span of virulence among isolated strains from none to medium. Caractérisation de bactéries pathogènes, Vibrio splendidus, isolées de bivalves marins ArchiMer

Nowadays, cupped oyster, Crassostrea gigas, production is the main shellfish production in France. Spats cultivated on most farms are usually collected in the wild, but hatchery spats are becoming more numerous, and represent around 10% of today's cultivated spats. During the summer period, when the temperatures are higher, high mortality rates have been regularly observed in cupped in oyster C. gigas (60 to 100%) spats since 1991, both in hatcheries-nurseries and in their natural habitat. Over the last five years, these episodes have become a big concern for all the professionals.

First record of Marteilia sp in mussels Mytilus galloprovincialis in Croatia ArchiMer

Marteiliosis is a disease of molluscs caused by Marteilia refringens in Europe and M. sydneyi in Australia. During routine examination of cultured mussels Mytilus galloprovinciallis in the northern Adriatic, the occurrence of Marteilia sp. was recorded with a prevalence of 5 %. This parasite was not detected in flat oysters reared in the same area. The affiliation of the detected parasite in M. galloprovinciallis was confirmed by in situ hybridization using a M. refringens probe, specific at the genus level. DNA of these infected mussels originating from the same area will be used to clarify the taxonomic position of this species within the genus Marteilia using a molecular approach.

Mikrocytos roughleyi taxonomic affiliation leads to the genus Bonamia (Haplosporidia) ArchiMer

Microcell-type parasites of oysters are associated with a complex of diseases in different oyster species around the world. The etiological agents are protists of very small size that are very difficult to characterize taxonomically. Associated lesions may vary according to the host species, and their occurrence may be related to variations in tissue structure. Lesion morphology cannot be used to distinguish the different agents involved. Comparative analysis of Vibrio splendidus-related strains isolated during Crassostrea gigas mortality events ArchiMer

French mollusc production is based mainly on the Pacific cupped oyster, Crassostrea gigas. Since 1991, annual mass mortality of juveniles has been reported during summer months. These recurring episodes concern professionals who fear that like Portugese oyster, C. angulata, C gigas could in turn disappear following one of these epizooties. Previously, bacteriological analysis of moribund oyster juveniles yielded an isolate of a Vibrio splendidus biovar H strain, named TNEMF6. Vibriosis in bivalves : review of recent molecular, biochemical, and physiological studies ArchiMer

The emergence of new vibriosis in the European Atlantic coasts has been associated with mass mortalities of molluscs causing important economic losses. A molecular typing study on Vibrio strains implicated in shrimp disease outbreaks in New Caledonia and Japan was conducted by using AP-PCR (arbitrarily primed PCR). It allowed rapid identification of isolates at the genospecies level and studies of infraspecific population structures of epidemiological interest. Clusters identified within the species Vibrio penaeicida were related to their area of origin, allowing discrimination between Japanese and New Caledonian isolates, as well as between those from two different bays in New Caledonia separated by only 50 km. Pacific oysters were introduced into west coast of the USA from Matsushima Bay in Japan, infected with low levels of Haplosporidiumsp. identical to H.nelsoni(MSX) which causes high mortalities of Eastern oyster (C. virginica). A highly specific and sensitive DNA probe for H. nelsonican also detect Haplosporidium sp. in C.gigas of western US and Japan (Burreson and Stokes 2000).In 2002, H. nelsoni was detected in Japan using the same probe (Kamaishi and Yoshinaga 2002).Lesson: it appears that H. nelsoni does not cause serious disease in Pacific oysters; H. nelsoniis now speculated to have been introduced into the Pacific US by apparently healthy but infected C. gigas. A parasite of the dredge oyster Ostrea chilensis Philippi, previously known as Bonamia sp., was recently described and named Bonamia exitiosus, based on ultrastructural and molecular evidence. The rules governing the names of organisms require correspondence of gender, and the name must be changed from B. exitiosus to B. exitiosa.

Responsible for massive epizootics, the Marteilia type parasites are very dangerous infectious agents for oysters. As a matter of fact, in France the spreading out of the parasitosis caused by Marteilia refringens disturbed dangerously the flat oyster, Ostrea edulis, production; destroying a large percentage of the farms on the Atlantic coast. The fight against marteiliose implies a good management of the contamination risk, that is to say the identification of the parasite(s), the characterization of the host(s), vector(s) or reservoir(s) and the understanding of the parasite's transmission mode.

Prevalence and infection intensity of the ovarian parasite Marteilioides chungmuensis during an annual reproductive cycle of the oyster Crassostrea gigas ArchiMer

The occurrence of Marteilioides chungmuensis, a protozoan paramyxean parasite in the reproductive system of the Pacific oyster Crassostrea gigas, was observed at Gosung Bay, Korea. Seasonal variation in gonad development was investigated in a suspended cultured oyster population. Gametogenesis began in February and first-spawning was observed between mid and late June when surface water temperature reached 22 to 25degreesC. Spawning activity extended from mid June to late September, with 2 marked spawning peaks in June and August. Histological examination indicated that gonad development paralleled seasonal fluctuations in water temperature. We isolated complementary DNA (Cg-TIMP) from Crassostrea gigas that codes for a protein highly homologous to vertebrate tissue inhibitors of metalloproteinases (TIMPs). In vertebrates, TIMPs are multifunctional proteins that play a key role in extracellular matrix metabolism. They are involved in physiological processes (e.g. embryonic development, damage repair, angiogenesis) and pathological processes (e.g. inflammatory reactions, cancer).

Les parasites du groupe "Mikrocell" : Qui est qui? ArchiMer

Bonamiose, a disease caused by the protozoa Bonamia ostreae, has destroyed the flat oyster Ostrea edulis production, in France but also in Europe and in the United States since 1979. Another species of Bonamia has been monitored in New-Zealand, in Australia and lately in Chile. In Canada and Australia, two other parasites have been described in cupped oyster: Mikrocytos mackini and Mikrocytos roughleyi. All these small size protozoa are grouped under the name "Mikrocell". For most of them, light and electronic microscope descriptions have been realised. Based on those criteria, different taxonomic positions have been proposed. However, these analysis are still questioned today.
