Ce rapport clôture une collaboration, entreprise courant 97, entre l’Agence Nationale pour la Valorisation de la Recherche (ANVAR), l’Agence Française de Sécurité Sanitaire des Aliments(AFSSA), le Syndicat National des Aquaculteurs Marins (SFAM) et l’Institut Français de Recherche pour l’Exploitation de la Mer (IFREMER). The sea bass, Dicentrarchus labrax, has been subjected to intensive aquaculture on the Mediterranean cost for ten years. Marine fish hatcheries are key factors in the aquaculture field, and face different pathologies, especially one of viral origin (nodavirus) that could challenge their feasibility. Consequently, we have tried to provide some data on larvae and spawning stocks. Firstly, we have studied the immunoglobulins, components of the specific humoral immunity, and their role as antibodies. Sea bass (Dicentrarchus tabrax) (135 +/- 4 g) were reared under tank-based recirculating aquaculture system for a 63-day period at four densities: 10, 40, 70, 100 kg m(-3). Fish performance, stress indicators (plasma cortisol, proteonemia plus other blood parameters-Na+, K+, glucose, pH, total CO2- and water quality were monitored. At the end of the 63-day period, resistance to infection was also studied by a nodavirus challenge. A 25-day test was performed on fish from two extreme densities (10 and 100 kg m(3)) and one intermediate density (40 kg m(3)). Reproduction of nodavirus disease was performed by experimental infection of sea bass eggs during fertilization or at larval stage 4 with 2 genetically distinguishable nodavirus strains (Sb1 and Sb2) isolated from sea bass collected along the Atlantic and Mediterranean French coast. The pathogenicity of the virus strains was assigned after detection of the virus by ELISA and immunohistochemistry (IHC). The Atlantic (Sb1) strain was more pathogenic than the Mediterranean (Sb2) strain during the fertilization step whilst both strains were pathogenic following experimental exposure of 4 d old larvae. Feeding motivation is one major indicator of fish welfare and an investigation on the link between feed demand, growth and physiological variables in sea bass juveniles was developed. A computerized on-demand feeding system coupled with a PIT tag monitoring device was used to continuously record for 219 days the triggering activity of 150 individuals (initial average body weight 131.6 +/- 1.80 g and coefficient of variation 16.8%). Each group was held in 400 1 tanks at 22.2 +/- 1.5 degrees C and light regime was 16:8 LD. In aquaculture management it is important to establish objective criteria to assess health and welfare of the fish. Here we show that European sea bass (Dicentrarchus labrax) confronted with husbandry-associated stress (tank cleaning, i.e. scrubbing, and water temperature variation) during early life stages show poorer survival and disease resistance as juveniles. During 84 days, the effects of density on juvenile sea bass (Dicentrarchus labrax) (76 +/- 16 g) were studied in an experimental tank-based flow through system. Performance, stress response and resistance to virus infection were analysed under five stabilized rearing densities: 10, 20, 40, 70 and 100 kg m(-3). Three batches of rabbits were immunized against 2 strains of Vibrio anguillarum (Va V 62 and Va V 408), which cause vibriosis in sea bass Dicentrarchus labrax (L. 1758) Serranidae. Homologous and heterologous reactions were obtained against these Vibrio strains and other bacteria using an improved indirect ELISA technique. Antibody and antigen titrations revealed high immunogenecity (high antibody titers for the antisera) of the 2 Vibrio strains. They correspond to serotype I and show close affinity.
